永康高山绿茶超高效液相色谱指纹图谱研究

摘    要:采用超高效液相色谱(UPLC)法对永康高山绿茶中主要化学成分的指纹图谱进行研究。方法采用岛津Shim-pack XR-ODSⅡ色谱柱(2.0 mm×150 mm,2.2 μm),以0.05%三氟乙酸水溶液和甲醇为流动相进行梯度洗脱,流速0.2 mL/min,柱温30 ℃。经分析建立了共有模式和18个共有峰,并确定其中9个峰的化学成分。构建的13个永康绿茶样品指纹图谱的相似度>0.98。经聚类分析和判别分析,永康高山绿茶化学性质稳定。构建的永康高山绿茶指纹图谱专一性强、稳定性好,可对永康绿茶进行控制与评价。

Abstract:The fingerprint of main chemical components in Yongkang green tea was studied using UPLC. The method was performed on a Shim-pack XR-ODSⅡ chromatographic column (2.0 mm×150 mm, 2.2 μm) with the mobile phase consisting of 0.05% trifluoroacetic acid and methanol using gradient elution at a flow rate of 0.2 ml/min and column temperature of 30 ℃. 18 common chromatographic peaks and the common pattern of Yongkang green tea were established, and 9 chemical components were identified. The established fingerprint similarity among 13 Yongkang green tea samples was more than 0.98. By cluster analysis and discriminate analysis, the results showed that the chemical properties of Yongkang green tea were stable. The established fingerprint had the advantage of good specificity and stability, which could be used to control and evaluate Yongkang green tea.
