微小隐孢子虫gp40基因的克隆与核苷酸序列分析

摘    要:目的 获取微小隐孢子虫侵入蛋白40基因(gp40),并将其克隆到 pMD18-T载体中进行核苷酸序列分析。方法 应用PCR技术,从牛源微小隐孢子虫基因组中扩增出 975bp的gp40基因,然后将其克隆到 pMD18-T载体中,用Sanger’s双脱氧法对重组质粒中的插入片段进行序列分析。结果 测得的核苷酸序列及其推导的氨基酸序列与Cenbank公布的序列核苷酸同源性为98%,氨基酸同源性为98%。结论 获得了序列正确的gp40基因,为其相关研究奠定了基础。

Abstract:Objective To obtain the invasion protein 40( gp40) gene of Cryptosporidium parvum and clone it into plasmid pMD18-T for nucleotide sequencing. Methods gp40 gene was amplificated from genom-ic DNA of Cryptosporidium parvum by polymerase chain reaction (PCR) ,PCR product was inserted into vector pMD18-T directly. Then they were sequenced by Sanger's method. Results Comparison and analysis of the nucleotide and deduced amino sequences was performed with that reported in Genebank, The results showed the homology of the nucleotide and amino acid sequences were 98% and 98% . Conclusion A confirmed gp40 gene has been obtained.The study laid a good foundation of the future research about this gene.
