聚乳酸/Fe_3O_4载阿奇霉素微球的制备及其体外释药特性的研究

摘    要:利用共沉淀法制备了纳米Fe_3O_4颗粒,用TEM、XRD对其进行了表征。采用乳化一溶剂挥发法,制备了聚乳酸(PLA)载阿奇霉素复合微球(PIA-AZM-MS),通过正交试验优化微球配方。在此基础上,加入适量的纳米Fe_3O_4颗粒制备了聚乳酸/Fe_3O_4载阿奇霉素复合微球(PLA/Fe_3O_4-AZM-MS)。对这两种微球的形态、粒径、平均载药量、体外释药特性等进行了研究。结果表明,两种载药微球呈规则球形,粒径分布较均匀;纳米Fe_3O_4的加入可提高载药微球的平均载药量,磁性微球的粒径更小;聚乳酸作为药物载体具有明显的药缓控释作用,而聚乳酸/Fe_3O_4载阿奇霉素微球的药物缓释性更佳。

Abstract:Magnetic Fe_3O_4nano particles were prepared by chemical coprecipitation method and characterized by TEM and XRD. PLA AZM MS ( microsphere of azithromycin carrying polylactic acid) and PLA/Fe_3O_4 AZM MS were prepared by emulsification solvent evaporation method and their optimum formula were obtained by orthogonal test. The morphologies, particle sizes, average drug loadings and invitro release properties of PLA AZM MS and PLA/Fe_3O_4 AZM MS were studied. And the results show that both of them are regular sphere and uniform particle size distribution, PLA/Fe_3O_4 AZM MS load more drug with smaller particle size compared with PLA AZM MS. Polylactic acid as drug carrier has obvious controlled release property, and PLA./Fe_3O_4AZM MS has longer release time.
