运动发酵单胞菌发酵产乙醇的中试研究

摘    要:从6株Zymomonas.mobilis基因工程菌中筛选出一株乙醇高产菌Z4,在500ml摇瓶小试的基础上利用5L全自动发酵罐进行了中试放大。结果表明Z4菌株的最适初糖浓度为18%;pH6.0,35℃,发酵48h乙醇最高含量达67.9g·L-1,得率达77.0%。补料分批培养实验结果表明,补料时机以发酵起始后24h为佳,发酵48h后乙醇含量达最高值93.1g·L-1,得率为77.6%。

Fermentation production of ethanol by zymomonas mobilis genetic engineering strain Z4 on pilot scale

Abstract:A high yield ethanol strain was selected from 6 Zymomonas mobilis genetic engineering strains. This strain was named Z4 and its fermentation conditions in 5L auto-fermentor were studied on the basis of 500ml agitation flask. According to the experiment results, the optimum initial glucose concentration was 18% under 35℃, pH6.0. Peak production of the ethanol was at 48h after fermentation with a maximum yield of 67.9 g·L~-1 (77.0% productivity of alcohol). The time of feeding in feeding-batch culture was explored and the experimental results showed that the optimum time of feeding was 24h after fermentation. Peak production of the ethanol was at 48h and the highest yield and productivity of ethanol were 93.1 g·L~-1and 77.6%.
