双内参实时荧光定量PCR法检测肝癌中易洛魁族同源盒2基因信使RNA的水平

目的采用双内参实时荧光定量PCR法检测肝癌中易洛魁族同源盒2(Iroquois homebox 2,IRX2)基因信使RNA(mRNA)的水平,探讨其对肝癌诊断及治疗的潜在意义。方法采用荧光定量PCR法检测21份肝癌组织和21份癌旁组织中IRX2基因的表达水平,并以双内参β-actin和GAPDH为标准对照,计算每个样本IRX2基因的相对表达量,经Pfaffl法进行相对定量,分析基因表达差异。结果 42份肝组织中均存在IRX2基因的表达,85.71%(18/21)的肝癌组织中IRX2基因表达水平较癌旁组织明显降低(P<0.01)。结论肝癌组织中IRX2基因表达水平较低,表明IRX2基因在肝癌中出现异常表达,为从基因水平诊断及治疗肝癌提供了一个潜在靶点。

Determination of Iroquois homeobox 2 mRNA level in hepatocellular carcinoma by real-time fluorescent quantitative PCR using double internal references

Objective To determine the Iroquois homeobox 2(IRX2)mRNA level in hepatocellular carcinoma(HCC)by real-time quantitative PCR using double internal references,and investigate its significance in diagnosis and treatment of HCC.Methods The expression levels of IRX2 gene in tumor and normal adjacent tissues of 21 patients with HCC were determined by fluorescent quantitative PCR using β-actin and GADPH as internal references,and the relative expression level in each sample was calculated,between which the difference was analyzed.Results The expressions of IRX2 gene were observed in all the 42 specimens.However,the expression levels in 85.71%(18 / 21)of tumor spe-cimens were lower than those in normal adjacent tissue(P < 0.05).Conclusion The expression level of IRX2 gene in HCC tissue was low,indicating abnormal expression.The study provided a potential target for diagnosis and treatment of HCC at gene level.
