Abstract:Based on single factor experiment,response surface methodology was used to optimize the extraction conditions of polyphenols from lotus root,and the antioxidant,hypoglycemic and uric acid reduction activities of polyphenols were determined.The results showed that the optimal extraction process parameters of lotus root polyphenols were liquid-to-material ratio 22:1(mL/g),extraction temperature 53 ℃,and extraction time 41 min.Under these conditions,the extraction amount of polyphenols was (359.55±1.20)mg/100 g.The 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH) radicals and 2,2’-azinyl-bis-(3-ethylbenzothiazole-6-sulfonic acid radical (ABTS+))scavenging activities,ferric reducing antioxidant power (FRAP) value and N,N-dimethyl-p-phenylenediamine dihydrochloride (DMPD) radical scavenging capacity were (1292.84±9.35)μmol Trolox/g,(695.90±19.53)μmol Trolox/g,(353.31±0.99) μmol Trolox/g and (1029.15±2.54)μmol Trolox/g,respectively.The inhibition rates of α-glucosidase and α-amylase were (77.54±0.2)% and (33.86±2.0)%,respectively.The inhibition rate of xanthine oxidase was (46.87±3.1)%.In vitro experiments showed that lotus root polyphenols not only had good antioxidant activity,but also showed good effect in lowering blood glucose and uric acid.
