长白山湿地细菌群落结构分析及优势菌株鉴定

摘    要:对长白山北坡和西坡不同湿地的土壤细菌的群落结构及优势菌株进行了总体研究.采用PCR-DGGE分子生物学技术,对湿地土壤样品中总DNA的16S rDNA的V3区进行PCR扩增,通过变性梯度凝胶电泳(DGGE)对扩增产物进行分离.回收了26个清晰的条带,对其中19个特征性条带进行了测序,除了条带12、13、16为可培养的微生物外,其他16个条带都是不可培养微生物.通过传统培养法,利用MIDI微生物鉴定系统,对8种可培养的最优势菌株进行了分析,各菌种分别为Bacillus sp., Bacillus licheniformis, Virgibacillus pantothenticus, Bacillus cereus, Micrococcus luteus, Paenibacillus alginolyticus, 优势菌株间的相似性水平较低.

Abstract:In this study, the bacteria community structure and dominant strains of the soil samples from different wetlands of Changbai Mountain north slope and western slope were generally investigated. PCR-DGGE(Denaturing Gradient Gel Electrophoresis)molecular biology technique was used to analyze the bacteria community of different wetland in Changbai Mountain. The V3 variable region of the bacterial 16S rDNA gene fragment of total DNA in the wetland was amplified by PCR. The PCR products were checked by DGGE. 26 clear bands were excised from DGGE gel profiles, 19 specific bands were chosen for sequencing, the results show that band 12, 13 and 16 are cultivable bacteria and the other 16 bands belong to uncultivable kind. By the traditional culture method, 8 kinds of dominant strains that isolated and purified from different wetland were identified using MIDI microbial identification system, dominant strains containing Bacillus sp., Bacillus licheniformis, Virgibacillus pantothenticus, Bacillus cereus, Micrococcus luteus, Paenibacillus alginolyticus is identified. Low similarity level appeared between different dominant strains.
