摘    要:运用响应面分析法优化提取野西瓜种子蛋白质的最佳工艺条件,以蛋白质提取率作为响应值,对温度、pH值、提取时间和液料比进行中心组合设计。试验得出的二次模型的相关系数R2 = 96.7%。蛋白质的提取率主要受溶液的pH和液料比的影响。经过响应面分析得出野西瓜种子中蛋白质提取最佳条件为液料比23∶1(mL:g),提取时间50 min,pH值8.40和温度45 ℃,该条件下经模型预测得到的蛋白质提取率为17.63%,实际实验值为(17.19±0.09)%。

Abstract:The optimum conditions for protein extraction from Capparis spinosa seeds were determined by response surface methodology. Using protein extraction ratio as evaluation index, a central composite design was conducted using temperature, pH, extraction time and solid to liquid ratio as indexes. The quadratic correlation coefficient of the model was 96.7%. Results showed that protein yield was primarily affected by pH and solid to liquid ratio, and the optimal protein extraction conditions from C. spinosa seeds were solid to liquid ratio 23:1 (ml:g), time 50 min, pH 8.4 and temperature 45℃. Under these conditions, the predicted protein extraction ratio was 17.63%, and the experimental value was (17.19±0.09)%.
