拮抗丁香假单胞菌海洋微生物的筛选与鉴定

摘    要:该研究采用平板涂布法和平板划线法从印度洋沉积物中分离纯化海洋微生物,首先采用滤纸片法对丁香假单胞菌拮抗菌株进行初筛,然后采用牛津杯法进行复筛,并通过形态观察、生理生化试验和分子生物学技术对其进行菌种鉴定,最后研究其抑菌谱。结果表明,从印度洋沉积物中共分离纯化出115株微生物,经过初筛得到33株丁香假单胞菌拮抗菌株,再经过复筛后,筛选出一株对丁香假单胞菌抑菌效果最好的菌株,编号为NO.4.1.3-1,其抑菌圈直径达(12.63±0.06) mm。该菌被鉴定为贝莱斯芽孢杆菌(Bacillus velezensis);除屎肠球菌外,该菌对8株致病菌均有抑制作用,且对肺炎克雷伯氏菌、金黄葡萄球菌的抑菌作用较好,具有较广的抑菌谱。

Abstract:Marine microorganisms were isolated and purified from the sediments of the Indian Ocean using plate coating method and plate marking method. Firstly, the strains obtained against Pseudomonas syringae were preliminarily screened by the filter paper method, and rescreened by Oxford Cup method. Then the strain was identified through morphological observation, physiological and biochemical characteristics and molecular biology techniques. Finally, the antibacterial spectrum was studied. The results showed that a total of 115 strains of microorganisms were isolated and purified from the Indian Ocean sediments. 33 strains were obtained by the preliminary screening. After re-screening, one strain named NO. 4.1.3-1 with the optimal antibacterial effect against P. syringae was selected, and the diameter of the inhibition zone was (12.63±0.06) mm. The strain NO. 4.1.3-1 was identified as Bacillus velezensis. In addition to Enterococcus faecium, the strain NO. 4.1.3-1 had inhibitory effects on 8 pathogenic bacteria, had strong antibacterial effect against Klebsiella pneumoniae and Staphylococcus aureus, and also had a wider antibacterial spectrum.
