摘    要:为建立一种经济简便的利用荧光漂白后恢复技术检测酵母朊病毒流动性的方法,在参照文献荧光漂白后恢复技术研究酵母朊病毒流动性的基础上,借助表达融合蛋白GFP-Sup35p的酵母朊病毒模型(NGMC),对文献法的样片制备过程进行改良.采用YPAD培养基和价格低廉的普通载玻分别替代文献使用的培养酵母的合成培养基和固定酵母的腔室载玻片.结果表明:改良法在保持文献法准确性的基础上,操作更为简单,极大地降低了实验成本.

Abstract:An economic and convenient method for detecting the mobility of yeast prion by fluorescence recovery after photobleaching(FRAP) was established based on the previous reports.The synthesized culture medium and chamber slides which were used to culture and fix yeast prions in the previous reports respectively were replaced with YPAD medium and low-cost ordinary glass slides.With the expression of fusion protein GFP-Sup35p(NGMC),the samples which were prepared by modified process were assayed by FRAP.The results show that the improved method can not only maintain the accuracy of the literature method,but also simplify the operation and reduce experimental costs greatly.
