摘 要: | 把与玉米对生叶突变体对生性状两个显性位点紧密连锁的RAPD标记S312377与S360602成功地转化成SCAR标记CBJ1与CBJ2。对RAPD片段的纯化、T-A克隆、SCAR PCR引物的设计等SCAR标记转化进行了探讨,并对CBJ1与CBJ2进行了验证,为对生玉米的分子标记辅助选择与定位克隆奠定了基础。

Obtaining of SCAR Markers of Two Dominant Genes for Opposite Leaves and Fruits Trait of Zea Mays |

Abstract: | S312 377 and S360 602, two RAPD markers, were successfully transformed into SCAR makers respectively. The two SCAR markers, nominated CBJ1 and CBJ2, are closely linked to the two dominant genes for opposite trait in Zea mays opposite leaves and fruits mutant. The purification of the RAPD polymorphic band, clone of it by T-A vector, design of the SCAR primers and other protocol related the transformation were all study in this paper. The SCAR markers were validated as a good foundation of MAS (markers-assisted selection) and positional clone of the corresponding genes in Zea mays. |
