不同剂量乌司他丁对大鼠百草枯急性肺损伤的保护作用

摘    要:目的探讨不同剂量乌司他丁(UTI)对大鼠百草枯(PQ)急性肺损伤(ALI)的保护作用。方法将50只SD大鼠按随机数字表法分为5组:阴性对照组(NC组),阳性对照组(PC组),UTI治疗小剂量组(SD组)、中剂量组(MD组)、大剂量组(LD组),每组10只。阳性对照组与UTI治疗组大鼠采用百草枯一次性灌胃染毒法(80mg·kg^-1)制作PQ中毒肺损伤模型,阴性对照组采用等量生理盐水灌胃。灌胃30min后,SD组、MD组、LD组分别腹腔注射UTI 100、300、600U·kg^-1,NC组、PC组注射等量生理盐水。腹腔注射24h后观察各组大鼠一般情况;测定支气管肺泡灌洗液(BALF)白细胞总数及中性粒细胞百分比、蛋白含量;光镜下观察肺组织病理学变化,进行肺损伤病理评分。结果 PC组BALF白细胞总数、中性粒细胞百分比、蛋白含量及肺损伤病理评分均明显高于NC组(P〈0.05);SD、MD、LD组BALF白细胞总数、中性粒细胞百分比、蛋白含量及肺损伤病理评分较PC组均明显减少(P〈0.05),且随着UTI用量增加,逐渐降低(P〈0.05)。结论乌司他丁对大鼠百草枯急性肺损伤具有保护作用,且随着剂量增加保护作用逐渐增强。

Abstract:Objective To investigate the effects of different doses of ulinastatin(UTI) on paraquat(PQ)-induced acute lung injury(ALl) in rats. Methods Fifty SD rats were randomly divided into five groups: normal control group(NC group), positive control group(PQ group), low dose UTI treatment group(SD group),middle dose UTI treatment group(MD group) and higi: dose UTI treatment group(LD group) ,with 10 rats in each group. AI.I was induced by paraquat gavage (80 mg ~ kg ~) in NC group and UTI treatment groups. The NC group was given the same vol ume of saline. In addition,rats in SD,MD and LD groups were intraperitoneally injected with UTI at doses of 100,300 and 600U· kg^-1 30 minutes after gavage administration , respectively. Ratsin NC and PC groups were injected with the same volume of .'reline. The general conditions of rats were observed 24 hours after intraperitoneal injection. Moreover,the number of white blood cells, percentage of neutrophilic granulocytes and content of protein in bronchoalveolar lavage fluid (BALF) were determined,and pathologic changes in the lung were observed under the light microscope. Results Compared with NC group,the number of white blood cells,percentage of neu- trophilic granulocytes,protein content and pathological score were significantly increased in PQ group(P〈0.05). Compared with PC group, these parameters were significantly decreased by UTI treatment in a doseependent manner(P〈0.05). Conclusion UTI treatment can protect against PQ-induced ALI in a dose-dependent manner in rats.
