不同提取方法对红松籽油甾醇浸提及功能性质影响的研究

摘    要:以有机溶剂浸提法为对照,研究超声波、微波、光波辅助溶剂法对红松籽油甾醇浸提效果及不同方法提取物的抗氧化、抑制胰脂肪酶活性和与胆酸盐结合能力。通过磷硫铁比色法测定甾醇得率,采用自由基清除体系、对硝基苯酚法、模拟体外消化实验法分别测定四种方法提取甾醇的自由基清除能力、脂肪酶活性抑制效果及胆酸盐结合能力。结果表明,微波辅助溶剂法(500 W,1 min)提取的红松籽油甾醇得率(2.263±0.013)mg/g最大(P<0.05)。四种提取方法中,微波辅助溶剂法提取的红松籽油甾醇对DPPH·清除效果最好,超声波辅助溶剂法提取的红松籽油甾醇对ABTS+·清除效果最好,超声波辅助溶剂法提取的红松籽油甾醇可显著抑制胰脂肪酶活性(P<0.05),IC50值为(1.322±0.020)mg/mL,与牛磺胆酸钠、甘氨胆酸钠结合能力最强,分别为(77.593±1.401)%、(77.601±1.103)%。超声波辅助溶剂法提取的红松籽油甾醇得率略低于微波辅助溶剂法,但抗氧化能力、胰脂肪酶活性的抑制作用及胆酸盐结合能力均为最强,本研究为红松籽油甾醇的制备及功能性产品研发提供理论依据。

Effects of different extraction methods on extraction effect and functions of Korean pine oil sterol
Abstract:Compared with organic solvent extraction, the effects of ultrasonic, microwave and light wave assisted organic solvent on extraction effect of Korean pine oil sterol were studied, and the antioxidant, inhibition of pancreatic lipase activity as well as cholate binding capacity of the extracts by different methods were also studied. The yield of sterols was determined by phosphorus sulfur iron colorimetry, the free radical scavenging ability, lipase activity inhibition effect and cholate binding capacity of sterols extracted by four methods were determined by free radical scavenging system, p-nitrophenol method and stimulated in vitro digestion experiment. The results showed that microwave assisted solvent extraction (500 W, 1 min) had the highest yield of Korean pine oil sterol (2.263±0.013) mg/g (P<0.05). The results showed among the four extraction methods, microwave assisted solvent extraction of Korean pine oil sterol had the best DPPH· scavenging effect, ultrasonic assisted solvent extraction of Korean pine oil sterol had the best ABTS+· scavenging effect. The sterols extracted from Korean pine seed oil by ultrasonic assisted solvent method could significantly inhibit the activity of pancreatic lipase (P<0.05), and the IC50 value was (1.322±0.020) mg/mL. The sterols extracted by ultrasonic assisted method had the strong binding ability with sodium taurocholate and sodium glycocholate, which were (77.593±1.401) % and (77.601±1.103) %, respectively. The yield of sterol from Korean pine seed oil extracted by ultrasonic assisted solvent method was slightly lower than that by microwave assisted solvent method, but the antioxidant ability, lipase activity inhibition effect and cholate binding capacity were the highest, this study provides a theoretical basis for the preparation of Korean pine seed oil sterols and the development of functional products.
