茶树菇多糖的提取、气相色谱分析和AFM观测以及活性研究

摘    要:利用水提得到茶树菇水提多糖S-ACP,并对水提残渣进行碱提得多糖J-ACP。分别用考马斯亮蓝G-250法及苯酚硫酸法测定蛋白含量及总糖含量,气相色谱(GC)分析多糖组分,原子力显微镜(AFM)观测结构形貌,并进行了体外抗氧化活性测定。结果表明,S-ACP的单糖组成为木糖、甘露糖、葡萄糖、半乳糖,其摩尔比为2.527:0.140:1.284:1.738;J-ACP的单糖组成为鼠李糖、木糖、甘露糖、葡萄糖、半乳糖,其摩尔比为1.327:2.525:0.140:3.641:1.765;AFM观测表明S-ACP为多侧支的雪花状结构,J-ACP为少侧支的简单环状结构,且二者均非单链存在;抗氧化活性结果表明S-ACP活性高于J-ACP。

Abstract:A polysaccharide named S-ACP was extracted from Agrocybe aegerita by hot water,then a polysaccharide named J-ACP was isolated from the dried leaf mustard with alkali.The content of the protein and the total sugars were determined by Coomassic brilliant blue G-250 method and phenol-sulfuric acid method.The monosaccharide compositions of polysaccharides were analyzed by gas chromatography(GC).The structure of polysaccharides were investigated by atomic force microscopy(AFM).Meanwhile,the antioxidant activity in vitro of polysaccharides were also tested.Conclusion:Xyl,Man,Glu and Gal were determined to be the components of S-ACP in the mol ratios of 2.527:0.140:1.284:1.738.Rha,Xyl,Man,Glu and Gal were determined to be the components of J-ACP in the mol ratios of 1.327:2.525:0.140:3.641:1.765.It was found that S-ACP on mica formed Multi-lateral Snowflake-like structure by AFM,while J-ACP existed in the form of simple ring-like structure.The results showed that S-ACP had stronger antioxidant activity than J-ACP.
