直投式乳酸菌剂对便秘模型小鼠肠道功能的影响

摘    要:探讨直投式乳酸菌剂(direct vat set lactobacillus,DVSL)对便秘模型小鼠肠道功能的影响。小肠运动实验:将50只清洁级昆明雄性小鼠按体重随机分为5组,每组10只,即空白组,模型组和低、中、高剂量组。低、中、高剂量组小鼠每天分别灌胃2.09、4.17、8.34×107CFU/mL DVSL水悬液,空白组和模型组灌胃蒸馏水,每只小鼠的灌胃体积为0.2mL/10g体重,均自由摄食饮水,每周测定空腹体重1次。第15d空白组给予蒸馏水,其余四组小鼠灌胃浓度为5mg/kg复方地芬诺酯悬液,0.5h后各组均灌胃墨汁,观察各组小鼠的小肠推进率;小鼠排便实验:方法同上,不同的是实验19d时复方地芬诺酯灌胃浓度为10mg/kg,测定指标为首次黑便时间、6h内黑便粒数及其重量。各时间点各组小鼠体重均无统计学差异(P〉0.05);模型组小鼠小肠推进率、6h内黑便粒数及其重量均低于空白组,首次黑便时间延长,两组间差异均有统计学意义(P〈0.01),说明小鼠便秘模型建立成功;中剂量小鼠小肠推进率、6h内黑便粒数均高于模型组(P〈0.05),高剂量组小鼠6h内黑便粒数及其重量均高于模型组(P〈0.01),而首次黑便时间短于模型组(P〈0.01)。按照根据《保健食品检验与评价技术规范(2003版)》结果判定原则,直投式乳酸菌对便秘模型小鼠具有调节肠道功能的作用。

Abstract:To investigate the effect of direct vat set lactobacillus (DVSL) on intestinal function in mice with constipation. According to body weight, 50 male clean KM mice were randomly divided into five groups with 10 in each for the small intestinal movement experiment, which including control group, model group and the low, middle and high dosage group. Mice of the low, middle and high dosage group were respectively given the water solution of DVSL with 2.09, 4.17, 8.34×107CFU/mL at 0.2mL/10g body weight once a day, while mice of control group and model group were given water, each mouse was free access to diet and water, and the body weight of mice were recorded weekly. Mice of control group were given water, while that of model group and 3 dosage groups were compound dipheoxylate with 5mg/kg at the 15th day. Half an hour later, mice of control and model group were treated with ink, and that of the three dosage groups were the ink mixed with corresponding dose of DVSL respectively, the intestinal propulsive rate was measured after 25 minutes. According to the above methods, the time of the first black feces, the amount of feces within 6 hours and its weight were measured after mice were administrated compound dipheoxylate with 10mg/kg and ink in the defection experiment with 19 days. There were no statistical differences in body weight of mice in each group at different time (P〉0.05). And the intestinal propulsive rate, the time of the first black feces, the amount of feces within 6 hours and its weight had statistical differences in mice of control group and model group (P〈0.01), which suggested that mice constipation model was successfully established. The intestinal propulsive rate and the amount of feces within 6 hours of mice in middle dosage group were higher than that of model group (P〈0.05), and the amount of feces within 6 hours and its weight of mice in high dosages group were higher than that of model group (P〈0.01), while time of the first black feces was shorter (P〈0.01). According to the Technical Standards for Testing & Assessment of Health Food (2003 edition), the solution of direct vast set lactobacillus has the regulation of intestinal function in mice with constipation.
