南瓜多糖对2型糖尿病大鼠胰岛素抵抗及脂联素基因表达的影响

摘    要:目的:探讨南瓜多糖对2型糖尿病(2-diabetes mellitus,2-DM)大鼠糖脂代谢和脂联素(adiponectin)基因表达的影响。方法:将Wistar大鼠随机分为5组,A:正常对照组;B:模型组;C、D、E:南瓜多糖高、中、低剂量治疗组。8周后,测定空腹血糖(fasting bloodglucose,FBG)、空腹胰岛素(fasting insulin,FINS)、血脂和胰岛素抵抗指数(insulin resistance index,IRI);半定量RT-PCR检测adiponectin基因mRNA的表达水平。结果:南瓜多糖治疗组FBG、FINS、TG、LDL、VLDL、CH、IRI均低于模型组,而HDL的含量则高于模型组(P<0.05);adiponectinmRNA的表达高于模型组(P<0.05)。结论:南瓜多糖可上调adiponectin基因表达,降低胰岛素抵抗;降低2-DM大鼠血糖和改善其脂代谢紊乱。

Abstract:【Objective】To observe the effect of pumpkin polysaccharide(PP) on insulin resistance and mRNA expression of adiponection gene in perirenal adipose tissue of type 2 diabetes mellitus(2-DM) rats.【Method】The 2-DM model of rat was established by high-fat diet and streptozocin.The 2-DM Wistar were randomly divided into the model group and high,middle and low doses of PP groups with 10 animals in each group,another 10 rats were selected as the control group.The mRNA expression of adiponection gene in perirenal adipose tissue was determined by RT-PCR.The levels of fasting blood glucose(FBG),fasting insulin(FINS) and blood lipid total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL) and low density lipoprotein(LDL) were detected and insulin resistance index(IRI) was calculated after rats fed eight weeks.【Result】The mRNA expression of adiponection gene increased in the middle and high doses of PP groups,while the levels of FBG,FINS,IRI,TC,TG and LDL decreased in different doses of PP groups,significantly different from those in 2-DM model group.【Conclusion】PP can improve the metabolism disorder of blood lipid and decrease insulin resistance by increasing mRNA expression of adiponection gene in 2-DM rats.
