摘    要:RNA干扰是真核生物中普遍存在的基因转录后水平调控机制,已成为基因治疗的重要手段。RNA干扰文库的高通量筛选实现了对大量预测的药物靶点的快速筛选,在现代药物开发中起到重要作用。尽管这种高新技术的存在,对于高通量筛选产生的大量数据的处理仍然依赖于简单的数据统计和基本的统计学分析,易忽视筛选过程的系统性和随机性误差,从而对最终筛选结果的鉴定产生影响。文章提出一个完整的高通量筛选数据处理流程,并讨论各个阶段可供选择的基于现代统计学数据分析的方法。

Abstract:RNA interference (RNAi) is a regulatory mechanism of most eukaryotic cells that has become a very promising approach for gene therapy. Nowadays RNAi high-throughput screening(HTS) plays a key role in the development of drugs and makes possible the last screening of a huge number of predicted drug targets Despite this high technology, techniques used to analyze the large-scale data produced by HTS relied on simple and basic statistical analysis, and ignored the systematic and random variances, thus affecting the selection of final screening results. The paper reviews a complete sample workflow of HTS data processing, and discussed in detail some alternative modem statistical methods for data analysis in each step.
