绿豆酸性磷酸酶的分离纯化和部分性质研究

摘    要:从绿豆芽中提取酸性磷酸酶粗酶液。经分段盐析,DEAE-琼脂糖柱离子交换层析及Superdex-200凝胶过滤后,得到经SDS-聚丙烯酰胺凝胶电泳为单一蛋白区带的酸性磷酸酶。提纯倍数为156.3,酶液比活力为347.0 U/mg。凝胶过滤法测定酶分子质量为34.6 ku,SDS-PAGE测定酶的亚基分子质量为33.1ku。证明绿豆酸性磷酸酶为单亚基酶。该酶最适pH为5.2 ku,最适温度为40℃。以pNPP作底物时Km为4.28×10-3mol/L。Mg2+、Co2+对酶有激活作用,而Hg2+,Cu2+,Pb2+和Zn2+对该酶有明显的抑制作用,依次为:Hg2+>Cu2+>Pb2+>Zn2+。K+,Ca2+对酶影响不明显。

Abstract:Acid Phosphatase(ACP) was extracted crudely from Mung Bean sprout.ACP was a single electrophoretic band in SDS-PAGE by ammonium sulfate fraction precipitation,DEAE-sepharose chromatography and Superdex S-200 gelfiltration.Its purification fold was 156.3,and the specific activity was 347.0 U/mg.Superdex S-200 chromatography showed that the molecular weight of this ACP was 34.6 kD,and SDS-PAGE showed that the molecular weight of ACP subunit was 33.1 kD.It also showed that ACP from Mung Bean was a single subun...
