摘    要:建立了固相萃取-高效液相色谱法测定食品中脱氢乙酸的方法。方法采用C18(5μm,4.6mm×250mmi.d)反向色谱柱,流动相:V(甲醇)∶V(0.02mol/L乙酸铵溶液)=7∶93,流速1.0mL/min,检测波长290nm,柱温30℃,进样量5μL。该方法的检出限在1.5~4.8mg/kg,线性范围0.01~0.08mg/mL,相对标准偏差(RSD)0.95%~2.10%(n=6),加标回收率94.5%~100.4%。该方法简单、快速、灵敏度高,并具有良好的精密度与准确度,可作为食品中检测脱氢乙酸的有效定量方法。

Abstract:HPLC method for determination of dehydroacetic acid in food was introduced. The analysis column was a revered-phase column of C18(5μm,4.6mm×250mm i.d),the mobile phase was methanol -0.02mol/L acetic ammonium solution (7∶93 by volume),the flow was 1.0mL/min,the detect wavelength was 290nm,the column temperature was 30℃,and the injection volume was 5μL. The limit of detection was 1.5 and 4.8 mg/kg. The linear plots were obtained between 0.01 and 0.08 mg/mL. Overall recoveries were between 94.5 and 100.4%, and RSD (n=6) value were 0.95% and 2.10%.This was a simple, rapid, sensitive and effective method for directly determining dehydroacetic acid in food without pre-separation process.
