摘    要:研究了薄层扫描法测定万寿菊中叶黄素含量的方法。确定以硅胶G薄层板为固定相,石油醚-氯仿-丙酮(12∶5∶4)为展开剂,扫描波长445 nm,参比波长700 nm,采用反射法锯齿扫描;万寿菊中叶黄素酯的提取与皂化条件为:正己烷-丙酮-无水乙醇-甲苯(10∶7∶6∶7)为提取剂,料液比1∶80,10%KOH-CH3OH溶液为皂化液,皂化温度为45℃,皂化时间为40 min,水浴后暗处静置80 min,再加8 mL正己烷并用10%Na2SO4溶液定容,振荡后静置30 min。实验表明,在0.336~2.016μg范围内,叶黄素浓度与扫描峰面积呈良好的线性关系,该方法具有良好的精密性和稳定性,可用于分析万寿菊中叶黄素的含量。

Abstract:The TLC method was used to determine lutein.The zigzag scanning method was adopted.The fixed phase was silica gel(G) plate;the developing agent was ligroin-chloroform-acetone(volume ratio 12∶5∶4);the scanning wavelength was 445 nm and the reference wavelength was 700 nm.The lutein was exeracted from marigold flower by organic solvent.The extracting agent was a mixture of n-hexane-acetoneabsolute ethyl alcohol-methylbenzene(volume ratio 10∶7∶6∶7),the ratio of extracting agent was 1∶80(g/mL),and the saponification agent was 10% KOH-methanol.The solution was kept in water bath for 40 min of 45 ℃,and then placed in the dark for 80 min.8 mL of n-hexane and 10% Na_2SO_4 were added into the solution,and then it was kept still for 30 min after shaking.The result showed that the linearity of lutein was in the range of 0.056~0.336 μg/mL.The TLC method was reliable to determination the lutein in marigold flowers and the method was accurate and reproducille.
