荧光素-环胺-Cu(Ⅱ)离子荧光探针的合成及其在一氧化氮检测中的应用

摘    要:合成了一种新型一氧化氮分子荧光探针并对其结构进行了表征,此荧光探针为连接有1,4,8,11-四氮杂环十四烷环胺化合物的荧光素-环胺-Cu(Ⅱ)离子配合物,其本身无荧光,当与NO反应后,在λex=465nm光激发下产生荧光且最大发射波长λem=524nm。体系的pH值对荧光强度有影响,在pH为4.5时荧光强度最大。在H2O2,ONOO-存在下,荧光探针对NO表现出很好的选择性。以NOC-18为NO释放剂,在NOC-18浓度为4×10-5~14×10-5 mol.L-1范围内建立了NOC-18浓度和荧光强度间的关系曲线,荧光强度随NOC-18浓度的增加而增大,可实现对NO的直接检测。

Abstract:A novel fluorescent probe for NO,fluorescein-Cu(Ⅱ) complex connecting with 1,4,8,11-tetraazacyclotetra-decane,was synthesized and characterized.The complex himself showed non-fluorescent,when reacted with NO,it showed a strong fluorescent peak at λem =524 nm and λex=465 nm.The pH could affect fluorescence intensity and there was a maximum value when pH=4.5.The fluorescent probe exhibited an excellent selectivity to NO in the presence of H2O2 or ONOO-.A relationship curve was developed between fluorescent intensity and concentration of NOC-18(a donor of NO).At range of 4×10-5 to 14×10-5 mol·L-1 of NOC-18,the fluorescent intensity was increased with increasing of NOC-18 concentration.Therefore,NO can be determined directly through measuring fluorescent intensity.
