南方刺参性腺多肽酶解工艺优化及抗氧化活性分析

摘    要:研究南方养殖刺参性腺多肽的酶解提取条件,并分析南北刺参性腺多肽抗氧化活性。选用复合蛋白酶对刺参性腺进行水解,以酶解多肽得率为指标,选取酶解温度、底物浓度、pH、酶解时间、酶用量等为主要影响因素,在单因素实验基础上应用响应面分析法对以上5个因子进行优化,确定刺参性腺多肽酶解工艺。试验结果表明,刺参性腺多肽酶解最佳条件:温度为56 ℃,底物浓度11.5%,酶添加量0.5 mKat/g,pH7.1,酶解时间4.5 h,此时刺参性腺多肽得率可达到67.97%±0.96%,与预测值相差0.26%。抗氧化试验结果表明,刺参性腺多肽浓度在40 mg/mL时,羟自由基抑制率与超氧自由基抑制率均大于0.1 mg/mL的维生素C溶液,60 mg/mL刺参性腺多肽的FRAP值大于0.1 mg/mL的维生素C溶液,南、北方养殖刺参性腺酶解多肽的抗氧化能力无显著差异。

Abstract:This study optimized polypeptides extraction conditions of sea cucumber (Apostichopus japonicus) gonads from south of China. And the antioxidant activity of sea cucumber gonads polypeptides from south and north of China were compared. Protamex was used to hydrolyze the sea cucumber gonads. Taking the yield of enzymatic peptide as the index, the main influencing factor experiments such as hydrolysis temperature, substrate concentration, pH, time, enzyme dosage and response surface methodology, the enzymatic condition was selected. The above five factors were optimized by response surface methodology on the basis of single factor experiment. The results showed that the optimum conditions were as follows:temperature of 56℃, substrate concentration of 11.5%, enzyme dosage of 0.5 mKat/g, pH of 7.1, enzymolysis time of 4.5 h. The peptide yield was 67.97%±0.96% in this condition, which was 0.26% from the prediction model. The antioxidant activity of gonads polypetide had been measured. The results showed that the superoxide radical clearance and hydroxyl radical clearance of 40 mg/mL Apostichopus japonicas gonad polypeptide solution from south and north of China were greater than 0.1 mg/mL Vitamin C solution, the FRAP value of 60 mg/mL Apostichopus japonicas gonad polypeptide solution from south and north China were greater than 0.1 mg/mL Vitamin C solution. There was no significant difference between these two gonads polypeptides.
