香蕉低聚糖对鼠李糖乳杆菌增殖条件的优化

摘    要:探寻香蕉低聚糖对鼠李糖乳杆菌的最佳增殖条件。以香蕉低聚糖为碳源,以鼠李糖乳杆菌的数量为指标,在单因素实验的基础上,采用响应曲面法研究培养时间、低聚糖添加量、菌液浓度以及初始菌液pH对鼠李糖乳杆菌增殖数量的影响,建立各种因素与鼠李糖乳杆菌增殖数量关系的数学回归模型。结果表明:当以香蕉低聚糖为碳源时,对鼠李糖乳杆菌增殖的最佳条件为:培养时间37 h,低聚糖添加量为6 g/L,菌液浓度为2.2×106 CFU/mL,pH为6.80。在此条件下,进行三次重复实验,实际测得的鼠李糖乳杆菌数量为1.5×109 CFU/mL,取对数后得9.1706,与预测值的RSD值为0.149%。综上所述,本实验所得模型合适,回归方程对实验拟合度好,其最佳优化条件适合于鼠李糖乳杆菌的增殖。

Abstract:To study the optimal conditions of banana oligosaccharide on the proliferation of Lactobacillus Rhamnus.Using banana oilgosaccharides as carbon source, the quantity of Lactobacillus Rhamnus as the measure, based on the single-factor experiment, the effect of culture time, additive amount of banana oligosaccharides, initial bacterial concentration and pH of basic medium was studied by the response surface methodology (RSM).Regression model between the quantity of Lactobacillus rhamnus and the four factors was built.Results showed that banana oligosaccharides as carbon source, the optimal conditions of Lactobacillus rhamnus proliferation was achieved as follows:Culture time of 37 h, additive amount of banana oligosaccharides of 6 g/L, bacterial concentration of 2.2×106 CFU/mL, pH of basic medium of 6.80. Under this condition, three times repeated experiments were carried out, the actual measured number of Lactobacillus rhamnosus was 1.5×109 CFU/mL, and the logarithm was 9.1706, the predicted RSD value was 0.149%.In summary, this experimental model was suitable, the regression equation has good fitting fitness, and the optimal optimization condition is suitable for the proliferation of Lactobacillus rhamnosus.
