摘    要:目的优化硒蛋白的分离提取技术。方法以富硒发芽糙米为原料,采用碱提酸沉法提取硒蛋白,研究NaOH溶液浓度、料液比、浸提时间、pH值、(NH_4)_2SO_4饱和度5个因素对硒蛋白产量的影响,并通过正交试验确定硒蛋白分离提取的最优工艺。结果硒蛋白分离提取的最佳条件为:NaOH溶液浓度0.12 mol/L、料液比1:10(g:mL)、浸提时间4 h、pH 4.5、(NH_4)_2SO_4饱和度50%。在此条件下,10 g富硒发芽糙米中硒蛋白产量为327.56 mg,即硒蛋白得率约为3.28%,硒含量为0.73μg,换算成硒得率为48%。结论本方法简便快捷且硒蛋白及硒得率较高,可作为富硒发芽糙米中硒蛋白的分离提取方法。
