电子鼻鉴定米曲霉生长繁殖可行性研究

摘    要:目的研究PEN3电子鼻检测郫县豆瓣制曲过程中米曲霉的生长繁殖程度的可行性。方法分别利用PEN3电子鼻检测郫县豆瓣制曲过程中挥发性气体的特征数值及荧光定量PCR(quantitative real-time PCR,QPCR)检测豆瓣制曲过程中米曲霉的生长繁殖程度,通过雷达图分析、主成分分析(principal component analysis,PCA)、线性判别分析(linear discriminant analysis,LDA)、支持向量机(support vector machine,SVM)等数据分析方法,结合2种方法的测定结果,判断电子鼻鉴定米曲霉生长繁殖程度的可行性。结果电子鼻侦测数值结合LDA与SVM模式识别均能实现对接种发酵过程米曲霉的增长程度判别,SVM模式识别出不同生长繁殖程度的米曲霉的正确率超过90%。结论电子鼻检测郫县豆瓣米曲霉的生长繁殖是可行的,为郫县豆瓣行业提供了新的快速无损检测方法。

Abstract:Objective To investigate the feasibility of the growth and reproduction of Aspergillus oryzae in the Pixian soybean paste process by electronic nose. Methods The characteristic values of volatile gas in Pixian soybean paste were measured by PEN3 electronic nose, and the growth and propagation of Aspergillus oryzae in the process of Pixian soybean paste were detected by quantitative real-time PCR. Using four data analysis methods such as radar chart, principal component analysis (PCA), linear discriminant analysis (LDA) and support vector machine (SVM) combined with the results of two methods, the growth of inoculated Aspergillus oryzae in starter propagation process were explored by non-destructive method. Results The LDA and SVM pattern recognition combined with the electronic nose could identify the growth of Aspergillus oryzae realize in different Pixian soybean paste starter propagation process, and the accuracy of SVM was more than 90%. Conclusion It is feasible to detect the growth and reproduction of Aspergillus oryzae by electronic nose, which provides a new, rapid and non-destructive testing technology for Pixian soybean paste industry.
