复合酶解裂壶藻渣制备抗氧化肽的工艺研究

摘    要:以提取油脂后的裂壶藻渣为原料采用蛋白酶进行酶解,以水解度、蛋白回收率及产物抗氧化活性为指标,确定最佳复合酶解工艺。结果表明,在p H为7.5,水解温度为50℃,料水比为1∶12(w/w),加酶量为0.5%(E/S,w/w)条件下,采用复合蛋白酶与Alcalase 2.4 L复配方式(1∶1,w/w)酶解裂壶藻渣6 h后的产物水解效果与抗氧化活性均优于其他单酶或复合酶解产物。在此条件下,酶解产物的水解度为11.15%,蛋白回收率为70.82%,羟自由基半抑制浓度IC50为1.656 mg/m L,还原力为1.417(浓度3 mg/m L),具有较好抗氧化活性。

Abstract:In this paper,Schizochytrium limacinum after oil extraction was hydrolyzed while the degree of hydrolysis,protein recovery and product antioxidant activity were as index to determine the best complex enzymatic processes. The results showed that the optimum conditions were as follows:p H7.5,temperature 50 ℃,ratio of material to water 1∶12,time 4 h,Protamex and Alcalase 2.4 L complex method,total amount of enzyme dosage0.5%(E/S,w/w,1∶1),which was significantly better than the effect of single enzymatic hydrolysis. In this condition,the hydrolysates had the degree of hydrolysis of 11.15%,protein recovery 70.82%. In addition,IC50 of hydroxyl radicals was 1.656 mg/ m L and reducing power was 1.417.
