大孔吸附树脂纯化冬枣总黄酮的研究

摘    要:以总黄酮含量为考察指标,比较了5种大孔树脂对冬枣黄酮的吸附和解吸效果,从中筛选出适合冬枣黄酮分离纯化的树脂,并对其吸附和解吸条件进行了探讨。结果表明,X-5树脂对冬枣总黄酮有较好的吸附和解吸效果。冬枣黄酮在X-5型树脂上的吸附平衡时间为1.5h、上样液的质量浓度为0.8mg/mL、速率为3mL/min、pH值为3.0,树脂对冬枣总黄酮的饱和吸附量为44.34mg/g。最佳洗脱方式为:依次用2BV50%乙醇、1.5BV70%乙醇进行梯度洗脱,冬枣总黄酮洗脱率达82.42%,产品纯度达83.07%。

Abstract:The characteristic of absorption and separation of five kinds of macroporous resins to extract total flavonoids of Chinese Winter Jujube(CWJ) was studied. Suitable resin for purification of total flavonoids was screened from five resins, and its characteristic of absorption and separation was investigated. The results showed that X-5 macroporous resins had better adsorbing and separating property for purification of total flavonoids from Chinese Winter Jujube. The optimum absorption conditions were that the time of absorption equilibrium was 1.5 h, the concentration and the current velocity of the original solution were 0.8 mg/mL and 3 mL/min, and pH was 3.0 while the maximum adsorbing capacity for total flavonoids in Chinese Winter Jujube was 44.34 mg/g. The total flavonoids was separated and purified with 2BV desorbing volume of ethanol concentration 50%, 70% ethanol gradient desorption. In this condition, the elutive ratio was 82.42%,the purity of total flavonoids was 83.07%.
