2-氨基喹啉-3-甲酸乙酯的合成工艺改进

摘    要:以铁粉作还原剂,冰醋酸为溶剂,回流条件下,2-氰基-3-(2-硝基苯基)-丙烯酸乙酯经还原-加成环合得到2-氨基喹啉-3-甲酸乙酯。实验优化原料比为n[2-氰基-3-(2-硝基苯基)-丙烯酸乙酯]∶n(铁粉)=1∶5。在此反应条件下,标题化合物经还原-加成环合后的收率达到94.7%,其结构经1H NMR和GC-MS表征确认。

Abstract:2-Cyano-3-(2-nitrophenyl)-ethyl acrylate was converted to ethyl 2-aminoquinoline-3-carboxylate under reflux in acetic acid with iron powder as the reducing agent.The results showed that the optimum molar ratio of 2-cyano-3-(2-nitrophenyl)-ethyl acrylate to iron powder was 1 to 5,and the target compound was obtained in 94.7% yield with the structure confirmed by 1H NMR and GC-MS.
