纤维素酶糖化水解荔枝渣工艺条件的优化研究

摘    要:采用纤维素酶解的方法对荔枝渣进行糖化水解,以增加料液中还原糖的含量,进而提高生物质发酵的利用率。试验研究了酶添加量、酶解初始pH、酶解温度及时间4个因素对糖化效果的影响,先后采用了单因素试验和正交设计试验对糖化工艺进行优化,并经验证得出最佳工艺条件为:纤维素酶添加量50u/g、初始pH4.8、酶解温度50℃、酶解时间持续2.5h,此条件下荔枝渣料液中的还原糖含量可达12.41mg/mL。

Abstract:To advance the reducing sugar content in feed liquid and increase the utilization rate in biomass fermentation, the effects of cellulose hydrolysis on the saccharification of litchi residue were investigated in this paper. The technological conditions involving enzyme amount, initial pH value, enzymolysis temperature and time were studied by single factor experiment first. Based on the results, the orthogonal experiment was designed to obtain the optimum process conditions of saccharification. The results showed that the reducing sugar production was 12.41 mg/mL when litchi residue was treated with 50 u/g cellulase at pH4.8, 50 ℃ for 2.5 h in verify experiment.
