摘    要:4-1-(4-二甲胺基)-1-(4-氟苯基)-丁烯基]-3-(羟甲基)-苯腈是抗抑郁药西酞普兰合成中的一个副产物,对研究西酞普兰成品的质量具有较大价值。本文以4-4-(二甲胺基)-1-(4-氟苯基)-羟基丁基]-3-(羟甲基)-苯腈为起始原料,依次用特戊酰氯酰化,对甲苯磺酸脱水,最后用NaOH溶液催化水解特戊酰基,用甲苯重结晶后得到纯度在99%以上的4-1-(4-二甲胺基)-1-(4-氟苯基)-丁烯基]-3-(羟甲基)-苯腈成品,合收率约49%。

Abstract:4-1-(4-dimethylamino)-1-(4-fluorophenyl) butenyl]-3-(hydroxymethyl) benzonitrile was a by-product when producing the antidepressant citalopram.It had a great value to studying the quality of citalopram.In order to synthesize it,4-4-(dimethylamino)-1-(4-fluorophenyl)-hydroxy-butyl]3-(hydroxymethyl)-benzonitrile used as the starting material,acylated by 2,2-dimethylpropanoyl chloride,then dehydrated by TSCL,hydrolyzed by the catalyst of NaOH solution,and finally recrystallization with toluene in order to get the purity of the product more than 99%.The total yield of this synthetic route can get to 49%.
