嵌合抗CD20抗体分子在CHO细胞中的表达

摘    要:利用PCR方法从抗CD20抗体Fab′片段的表达载体上扩增抗CD20抗体的轻链和重链可变区,将轻链和重链可变区组装到含有人抗体恒定区的表达载体中,构建抗CD20抗体IgG的轻链和重链表达载体PK100VL和PGL05VH,并用质脂体法共转染CHO细胞,RT-PCR和ELISA检测结果证实了抗CD20抗体IgG全分子在CHO细胞中的分泌表达。免疫结合结果表明CHO细胞分泌表达的抗CD20抗体IgG全分子可与CD20阳性的B淋巴瘤Raji细胞结合。

Abstract:The genes of the variable region of heavy chain (VH) and light chain (VL) of the antibody against CD 20 were amplified from the expression vector pYZF1cd20 by PCR. The VH and VL were cloned into the plasmid PK100 and PGL05 to construct the light chain and heavy chain expression vector, respectively. The whole antibody against CD 20 was expressed in CHO cells. The competitive inhibition of immune fluorescence experiment shows that the antibody against CD 20 IgG inhibits the binding of monoclonal antibody HI47 to CD 20 on the surface of Raji cells.
