玻璃酸二甲基硅烷醇复合物对鼻黏膜上皮细胞的作用研究

摘    要:目的 探讨玻璃酸二甲基硅烷醇复合物(DSHA)的细胞毒性,及其对苯并芘(BAP)引起的兔鼻黏膜上皮细胞损伤的修复作用.方法 采用噻唑蓝(MTT)法检测不同浓度的DSHA原料药及鼻喷剂对鼻黏膜上皮细胞的细胞毒性,评价其对鼻黏膜上皮细胞增殖的影响;采用BAP刺激离体培养的兔鼻黏膜上皮细胞模拟雾霾对鼻黏膜的损伤,用MTT法检测DSHA对BAP引起的鼻黏膜上皮细胞损伤的保护作用,采用ELISA法检测DSHA对炎性因子肿瘤坏死因子α(TNF-α)、白介素1(IL-1)、IL-8表达水平的影响.结果 0.002%-0.05%的DSHA原料和0.001%-0.005%的鼻喷剂处理细胞24 h,细胞存活率均在99%以上,无细胞毒性;此外,DSHA可显著抑制BAP引起的鼻黏膜细胞存活力的下降,并能降低TNF-α、IL-1、IL-8的表达.结论 DSHA对鼻黏膜上皮细胞无细胞毒性且对大气污染物BAP引起的细胞损伤有修复作用.

Abstract:Objective To investigate the cytotoxicity of dimethyl silanol hyaluronate(DSHA)on nasal epithelial cells,and its repair effect on the injury of rabbit nasal mucosal epithelial cells induced by benzopyrene(BAP).Methods MTT assay was used to detect cytotoxicity of different concentrations of DSHA nasal spray and medicinal raw material on rabbit nasal epithelial cells,and to evaluate its effect on the proliferation of the cells.BAP was used to treat the cultured nasal mucosal epithelial cells to imitate the damage of air pollution.The protective effect of DSHA on BAP-induced injury of nasal epithelial cells was detected by MTT method.The inflammatory factors TNF-α,IL-1 and IL-8 expression levels were detected by ELISA.Results 0.002%-0.05%DSHA raw material and 0.001%-0.005%DSHA saline nasal spray were judged to be non-cytotoxic,and the cell survival rates were all above 99%.DSHA can significantly inhibit the decrease of viability of nasal mucosa cells induced by benzopyrene BAP,and down-regulate the expression of TNF-α,IL-1 and IL-8.Conclusion DSHA is not cytotoxic to nasal epithelial cells and could repair cell damage caused by air pollutant BAP.
