Alcaligenes sp.碱性脂肪酶产生条件及酶学性质的研究

摘    要:对Alcaligenessp .产碱性脂肪酶的工艺条件进行了研究。结果表明 ,适宜产酶培养基为 :3 %黄豆粉 ,1%果糖 ,0 .2 %K3 PO4,1%NaNO3 ,0 .6%柠檬酸钠 ,0 .0 3 %FeSO4,0 .5 %聚乙烯十八烷基醚 ;最佳培养条件为 2 5℃ ,初始pH9.0。脂肪酶活力可高达 12 0IU/mL。该酶在5 5℃以下 ,pH6.0~ 10 .5范围内稳定 ;酶反应适宜温度为 40℃ ,适宜pH9.0。该酶制剂在洗涤和制革工业中有良好的应用前景。

Abstract:The main factors affecting the lipase production by Alcaligenes sp. were studied. The results showed that the suitable medium for lipase producing was: 3% of soybean powder, 1% of fructose, 0.2% of K 3PO 4, 0.6% of sodium citrate, 0.03% FeSO 4·7H 2O, 0.5% of polyethylene stearyl ether. Under optimal culture conditions 25℃ and pH 9.0, the maximal lipase activity could reach 120?IU/mL. The enzyme was stable under 55℃ and pH 6.0~10.5, with optimal temperature and pH for enzymatic reaction were 40℃ and pH 9.0, having a good potential application in detergent and leather industry.
