The need to ensure the microbiological quality and safety of food products has stimulated interest in the use of mathematical models for quantifying and predicting microbial behaviour. For 20 years, predictive microbiology has been developed for predicting the occurrence of food-borne pathogens, although these tools are dedicated to bacteria. Recently, the situation has changed and a growing number of studies are available in the literature dealing with the predictive modelling approach of fungi. To our knowledge the present one is the first review focussed on predictive mycology and food safety, including mycotoxins; existing kinetic and probability models applied to mycotoxigenic fungi germination and growth, and mycotoxin production are reviewed.  相似文献

Growth of a range of 14 common food spoilage fungal species was evaluated along time as a function of both colony diameter and ergosterol content on malt extract agar. Growth was assessed under different environmental conditions following a central composite design. The suitability of using either linear, Gompertz's or Baranyi's models for primary modelling of the results was tested. Regarding colony diameters, using either linear or asymptotic Baranyi's function gave better estimations of growth rate and lag phase when no asymptotic trend was observed. When a decrease in growth rate was observed with time, standard Baranyi's model was chosen, although the search for new mechanistic models specific for moulds would probably improve the estimations. The use of Gompertz equation led, in general, to overestimated parameters. Ergosterol showed good performance as a fungal growth indicator for the whole range of species. Finally, significant correlation coefficients were found between ergosterol and colony diameters, suggesting that both parameters may be useful for primary modelling and thus for subsequent secondary modelling.  相似文献

Recontamination of food products can be the origin of foodborne illnesses and should therefore be included in quantitative microbial risk assessment (MRA) studies. In order to do this, recontamination should be quantified using predictive models. This paper gives an overview of the relevant modelling approaches that are available in the literature to quantify recontamination via factory environment. Different recontamination routes are described: recontamination via air, via processing equipment or via hand contact. Unfortunately, not many available models are directly applicable to the food industry; most models are developed for aquatic or environmental systems. Finally, a general systematic approach is proposed for modelling contamination from surfaces via air, hands or liquid into the product and ranges for the parameters are given.  相似文献

The goals of this study were to evaluate the effectiveness of two hand wash regimens in reducing transient bacteria on the skin following a single hand wash and the subsequent transfer of the bacteria to a ready-to-eat food item, freshly cut cantaloupe melon. The number of bacteria recovered from hands and the quantity transferred to the melon were significantly less following the use of an antibacterial soap compared with plain soap. The antimicrobial soap achieved > 3-log reductions versus Escherichia coli and 3.31- and 2.83-log reductions versus Shigella flexneri. The plain soap failed to achieve a 2-log reduction against either organism. The bacteria recovered from the melon handled by hands treated with antimicrobial hand soap averaged 2 log. Melon handled following hand washing with plain soap had > 3 log bacteria in the experiments. Based on previously published feeding studies, an infection rate in the range of approximately 15 to 25% would be expected after ingesting melon containing 2 log CFU compared with ingesting greater than the 3 log transferred from hands washed with plain soap, which would result in a higher infection attack rate of 50 to 80%. The data thus demonstrate there is a greater potential to reduce the transmission and acquisition of disease through the use of an antimicrobial hand wash than through the use of plain soap.  相似文献

Fungal growth and aflatoxin production by an aflatoxigenic strain during the manufacture of yoghurt are described. This completes previous studies which show that yoghurt may be a good substrate for aflatoxin production. Two factors were of special interest: (a) the temperature of the elaboration process (45 degrees C) and (b) the effect of lactic bacteria. Either during the elaboration of yoghurt or during its cooling from 45 degrees C to 4 degrees C, the necessary conditions required to start fungal growth are given; these are adequate enough for the synthesis of small quantities of aflatoxins. However our results do not clearly show that lactic bacteria have an influence on fungal growth and aflatoxin production.  相似文献

This communication deals with the use of meta-analysis as a valuable tool for the synthesis of food safety research, and in quantitative risk assessment modelling. A common methodology for the conduction of meta-analysis (i.e., systematic review and data extraction, parameterisation of effect size, estimation of overall effect size, assessment of heterogeneity, and presentation of results) is explained by reviewing two meta-analyses derived from separate sets of primary studies of Salmonella in pork. Integrating different primary studies, the first meta-analysis elucidated for the first time a relationship between the proportion of Salmonella-carrier slaughter pigs entering the slaughter lines and the resulting proportion of contaminated carcasses at the point of evisceration; finding that the individual studies on their own could not reveal. On the other hand, the second application showed that meta-analysis can be used to estimate the overall effect of a critical process stage (chilling) on the incidence of the pathogen under study. The derivation of a relationship between variables and a probabilistic distribution is illustrations of the valuable quantitative information synthesised by the meta-analytical tools, which can be incorporated in risk assessment modelling. Strengths and weaknesses of meta-analysis within the context of food safety are also discussed.  相似文献

This paper summarises recent trends in predictive modelling of microbial lag phenomena. The lag phase is approached from both a qualitative and a quantitative point of view. First, a definition of lag and an analysis of the prevailing measuring techniques for the determination of lag time is presented. Furthermore, based on experimental results presented in literature, factors influencing the lag phase are discussed. Major modelling approaches concerning lag phase estimation are critically assessed. In predictive microbiology, a two-step modelling approach is used. Primary models describe the evolution of microbial numbers with time and can be subdivided into deterministic and stochastic models. Primary deterministic models, e.g., Baranyi and Roberts [Int. J. Food Microbiol. 23 (1994) 277], Hills and Wright [J. Theor. Biol. 168 (1994) 31] and McKellar [Int. J. Food Microbiol. 36 (1997) 179], describe the evolution of microorganisms, using one single (deterministic) set of model parameters. In stochastic models, e.g., Buchanan et al. [Food Microbiol. 14 (1997) 313], Baranyi [J. Theor. Biol. 192 (1998) 403] and McKellar [J. Appl. Microbiol. 90 (2001) 407], the model parameters are distributed or random variables. Secondary models describe the relation between primary model parameters and influencing factors (e.g., environmental conditions). This survey mainly focuses on the influence of temperature and culture history on the lag phase during growth of bacteria.  相似文献

Extensive modelling studies on nitrogen (N) dynamics in flooded soil systems have been published. Consequently, many N dynamics models are available for users to select from. With the current research trend, inclined towards multidisciplinary research, and with substantial progress in understanding of N dynamics in flooded soil systems, the objective of this paper is to provide an overview of the modelling concepts and performance of 14 models developed to simulate N dynamics in flooded soil systems. This overview provides breadth of knowledge on the models, and, therefore, is valuable as a first step in the selection of an appropriate model for a specific application. © 2017 The Authors. This review on the use of indigenous solid-state fermentations (SSF) describes the microbiological transformation of plant raw materials into highly nutritious foods and flavor-enhancing ingredients. Traditional fermented foods from most countries of the world may be classified into the following categories: fungal fermentation followed by brining, SSF principally using bacteria, lactic acid fermentation followed by fungal fermentation, production of fermented doughs, alcoholic fermentation, and fermented food ingredients. Scientific studies of the principles behind SSF, identification of the essential microorganisms, development of suitable and versatile equipment, control of the process, and quality control of the substrate and final product can very well lead to a substantial increase at a world level in the availability and consumption of these fermented foods. The application of modern biotechnology in some countries is already changing the ancient methods of making fermented foods.  相似文献

Colony counting by spreading bacterial suspensions on plating media by various techniques is of general concern. Comparative studies between hand plating (Drigalski-spatula technique) for different time intervals and spiral plating resulted in significant differences in colony counts. Lower counts of Gram-negative bacteria were obtained by using hand plating for more than 10 s, compared with short time hand plating (5 s) or spiral plating. Colony counting of Gram-positive bacteria showed no differences between both techniques. Further characterisation of Escherichia coli cells spread with the Drigalski-spatula technique by electron microscopy revealed a large number of damaged cells compared to control samples. The data clearly shows that the mechanical forces during hand plating are sufficient to damage E. coli cells.  相似文献

Ordinary hand washing is well regarded as the primary means of removing bacteria from the hands. Because typical daily hand washings are not always thorough, antibacterial wash products provide an extra measure of protection by depositing a residual antibacterial agent to the skin that persists after washing. However, a more direct means of delivering antibacterial agents to the skin could be through a leave-on hand lotion. This paper describes testing which demonstrated that triclosan, a common antibacterial ingredient when administered in an oil-in-water anionic emulsion, was substantive to skin and effective against bacteria in both laboratory settings and in the normal daily environment. In vitro and in vivo methods were used to demonstrate the antibacterial lotion's spectrum of activity, substantivity on skin, persistence of activity, activity vs. resident flora and overall activity in practical use. The work established that a leave-on lotion was a suitable vehicle for the delivery of an antibacterial active ingredient to skin and that the actives delivered from a leave-on lotion were effective in use. Although it is recognized that antibacterial lotions are not a substitute for thorough hand washing and good hygienic practices, this work shows that antibacterial lotions can complement washing by providing consumers with a demonstrable reduction in bacterial levels on their skin. Furthermore, the work shows that the methods described could effectively quantify the antibacterial effects of a leave-on lotion both in the laboratory and in actual use.  相似文献

In many outbreaks of foodborne illness, the food worker has been implicated as the source of the infection. To decrease the likelihood of cross-contamination, food workers must clean and disinfect their hands frequently. To ensure their effectiveness, hand disinfectants should be tested using rigorous conditions that mimic normal use. Currently, several different methods are used to assess the efficacy of hand disinfectants. However, most of these methods were designed with the health care worker in mind and do not model the specific contamination situations encountered by the food worker. To fill this void, we developed a model that uses soil from fresh meat and a means of quantifying bacteria that is encountered and transferred during food preparation activities. Results of studies using various doses of para-chloro-meta-xylenol and triclosan confirm that the method is reproducible and predictable in measuring the efficacy of sanitizers. Consistent, dose-dependent results were obtained with relatively few subjects. Other studies showed that washing hands with a mild soap and water for 20 s was more effective than applying a 70% alcohol hand sanitizer.  相似文献

The functional food market, including products formulated to maintain a “healthy” gut microbiota, i.e. probiotics and prebiotics, has increased enormously since the end of the last century. In order to favor the competitiveness of this sector, as well as to increase our knowledge of the mechanisms of action upon human health, new probiotic strains and prebiotic substrates are being studied. This review discusses the use of exopolysaccharides (EPS), both homopolysaccharides (HoPS) and heteropolysaccharides (HePS), synthesized by lactic acid bacteria and bifidobacteria as potential prebiotics. These extracellular carbohydrate polymers synthesized by some gut inhabitants seem to be resistant to gastrointestinal digestion; these are susceptible as well to biodegradability by the intestinal microbiota depending on both the physicochemical characteristics of EPS and the pool of glycolytic enzymes harbored by microbiota. Therefore, although the chemical composition of these HoPS and HePS is different, both can be fermentable substrates by intestinal inhabitants and good candidates as prebiotic substrates. However, there are limitations for their use as additives in the food industry due to, on the one hand, their low production yield and, on the other hand, a lack of clinical studies demonstrating the functionality of these biopolymers.  相似文献

Fungal contamination of food and animal feed, especially by mycotoxigenic fungi, is not only a global food quality concern for food manufacturers, but it also poses serious health concerns because of the production of a variety of mycotoxins, some of which present considerable food safety challenges. In today's mega‐scale food and feed productions, which involve a number of processing steps and the use of a variety of ingredients, fungal contamination is regarded as unavoidable, even good manufacturing practices are followed. Chemical preservatives, to some extent, are successful in retarding microbial growth and achieving considerably longer shelf‐life. However, the increasing demand for clean label products requires manufacturers to find natural alternatives to replace chemically derived ingredients to guarantee the clean label. Lactic acid bacteria (LAB), with the status generally recognized as safe (GRAS), are apprehended as an apt choice to be used as natural preservatives in food and animal feed to control fungal growth and subsequent mycotoxin production. LAB species produce a vast spectrum of antifungal metabolites to inhibit fungal growth; and also have the capacity to adsorb, degrade, or detoxify fungal mycotoxins including ochratoxins, aflatoxins, and Fusarium toxins. The potential of many LAB species to circumvent spoilage associated with fungi has been exploited in a variety of human food and animal feed stuff. This review provides the most recent updates on the ability of LAB to serve as antifungal and anti‐mycotoxigenic agents. In addition, some recent trends of the use of LAB as biopreservative agents against fungal growth and mycotoxin production are highlighted.  相似文献

Modelling in food microbiology relies on the development and use of mathematical equations to describe biological processes. These models can be used to perform various tasks from quantitatively describing a phenomenon, testing significant kinetic differences, quantitatively investigate mechanisms and correlations, designing experiments and sampling plans, or predicting phenomena within a food chain to enable optimal control. It is important to clearly identify what one wants to realize, and depending on the purpose of the model, select the most appropriate approach for modelling. In this paper various applications are illustrated and for each application an example is given to highlight the broad use of models and model results in food microbiology.  相似文献

Pre‐clinical development comprises of different procedures that relate drug discovery in the laboratory for commencement of human clinical trials. Pre‐clinical studies can be designed to recognize a lead candidate from a list to develop the procedure for scale‐up, to choose the unsurpassed formulation, to determine the frequency, and duration of exposure; and eventually make the foundation of the anticipated clinical trial design. The foremost aim in the pharmaceutical research and industry is the claim of drug product quality throughout a drug's life cycle. The particulars of the pre‐clinical development process for different candidates may vary; however, all have some common features. Typically in vitro, in vivo or ex vivo studies are elements of pre‐clinical studies. Human pharmacokinetic in vivo studies are often supposed to serve as the ‘gold standard’ to assess product performance. On the other hand, when this general assumption is revisited, it appears that in vitro studies are occasionally better than in vivo studies in assessing dosage forms. The present review is compendious of different such models or approaches that can be used for designing and evaluation of formulations for nail delivery with special reference to anti‐fungal agents.  相似文献

The performance of several steady Reynolds-averaged Navier–Stokes turbulence models and boundary-layer modelling approaches is evaluated for a single sphere, by comparison with empirical data for a Reynolds number range of 10–3.2 × 104. A sphere serves here as a representative model for many spherical food products. The shear stress transport (SST) k–ω turbulence model performs exceptionally well when combined with low-Reynolds number modelling (LRNM) of the boundary layer, which confirms that the turbulence model characteristics are particularly suitable to deal with this specific flow problem. Especially the k–ε turbulence models are less accurate at higher Reynolds numbers (>102). Boundary-layer modelling with wall functions (WFs) leads to inaccurate flow-field and scalar transfer predictions, compared to LRNM. However, LRNM grids and their inherently higher computational cost are often not practically feasible, leaving WFs as the only option. It is shown that using cell sizes on the sphere surface of a few millimetres, typical for CFD studies on food products, can compromise accuracy, and grids with smaller cell sizes are actually required.  相似文献

The purpose of this paper was to estimate microbial growth through predictive modelling as a key element in determining the quantitative microbiological contamination of sea bass stored in ice and cultivated in different seasons of the year. In the present study, two different statistical models were used to analyse changes in microbial growth in whole, ungutted sea bass (Dicentrarchus labrax) stored in ice. The total counts of aerobic mesophilic and psychrotrophic bacteria, Pseudomonas sp., Aeromonas sp., Shewanella putrefaciens, Enterobacteriaceae, sulphide‐reducing Clostridium and Photobacterium phosphoreum were determined in muscle, skin and gills over an 18‐day period using traditional methods and evaluating the seasonal effect. The results showed that specific spoilage bacteria (SSB) were dominant in all tissues analysed, but were mainly found in the gills. Predictive modelling showed a seasonal effect among the fish analysed. The application of these models can contribute to the improvement of food safety control by improving knowledge of the microorganisms responsible for the spoilage and deterioration of sea bass.  相似文献

Field studies were conducted to characterize the concentration vs streamflow relationships (or "flow fingerprints") of fecal pollution and suspended solids in stormwater runoff from the Santa Ana River watershed, the largest watershed in southern California. The concentrations of fecal indicator bacteria and F+ coliphages (viruses infecting E. coli) exhibit little-to-no dependence on streamflow rates, whereas the concentrations of total suspended solids (TSS) exhibit a very strong (power-law) dependence on streamflow rates. The different flow fingerprints observed for fecal pollutants, on one hand, and TSS, on the other hand, reflect different sources and transport pathways for these stormwater constituents. The flow-independent nature of fecal indicator bacteria and F+ coliphages is consistent with the idea that these contaminants are ubiquitously present on the surface of the urban landscape and rapidly partition into the surface water as the landscape is wetted by rainfall. The flow-dependent nature of TSS, on the other hand, is usually ascribed to the shear-induced erosion of channel bed sediments and/or the expansion of drainage area contributing to runoff. The apparent ubiquity of fecal indicator bacteria and F+ coliphages, together with the very high storm-loading rates of fecal indicator bacteria and the low detection frequency of human adenovirus and human enterovirus, suggest that fecal pollution in stormwater runoff from the Santa Ana River watershed is primarily of nonhuman waste origin.  相似文献
