The copper complex of 4,5-diazafluorene-9-one (dafone) and bromine ligands ([Cu(dafone)2]Br2) was prepared and its interaction with double-stranded salmon sperm DNA (dsDNA) in pH 8.0 Britton-Robinson (B-R) buffer solution was studied by electrochemical experiments at the glassy carbon electrode (GCE). It was revealed that Cu(dafone)2Br2 could bind with salmon sperm DNA strands mainly by intercalation mode. The binding number of [Cu(dafone)2]Br2 for each salmon sperm dsDNA chain and equilibrium constant of the binding reaction were calculated to be 3 and 2.8 × 1012 L3 mol−3, respectively. The Cu(dafone)2Br2 was further utilized as a new electrochemical DNA indicator for the detection of human hepatitis B virus (HBV) DNA fragment by differential pulse voltammetry (DPV). The difference of its electrochemical responses occurred between hybridized dsDNA duplex and probe DNA was explored to assess the selectivity of the developed electrochemical DNA biosensor. The constructed electrochemical DNA biosensor achieved a detection limit of 3.18 × 10−9 mol L−1 for complementary target DNA and also realized a robust stability and good reusability.  相似文献

An electrochemical method for the detection of DNA hybridization using a novel electroactive, cationic, and water-soluble branched polyethyleneimine (BPEI)-cobalt(III)-phenanthroline(phen) polymeric indicator and single-stranded neutral peptide nucleic acid (PNA) probe on the Au electrode was developed. The indicator possesses some free amine groups, as well as cationic cobalt complexes in the polymer chain. It does not bind to neutral PNA capture probe alone. However, the indicator strongly interacts with the negatively charged backbone of the complementary oligonucleotide bound to the PNA probe through electrostatic interactions. The coordination spherical moieties also interact with the probe by embedding into the double-helix structure of PNA-DNA. These two interactions enable transduction of hybridization, producing a clear electrical signal in differential pulse voltammetry (DPV). The correlation against non-complementary DNA, three-base and one-base mismatch DNA was sharp, and the signal of indicator for the target DNA demonstrated a linear relationship within the concentration range of 5.0 × 10−9 to 2.5 × 10−7 M (R = 0.9940) with a detection limit of 5.6 × 10−10 M. These studies showed that the novel polymeric indicator and single-stranded PNA probe could be used to fabricate an electrochemical biosensor for DNA detection. This technique can provide an alternative route for expanding the range of detection methods available for DNA hybridization.  相似文献

A highly sensitive electrochemical sensor made of a glassy carbon electrode (GCE) coated with a Langmuir-Blodgett film (LB) containing polyaniline (PAn) doped with p-toluenesulfonic acid (PTSA) (LB/PAn-PTSA/GCE) has been used for the detection of trace concentrations of Ag+. UV-vis absorption spectra indicated that the PAn was doped by PTSA. The surface morphology of the PAn LB film was characterized by atomic force microscopy (AFM). The electrochemical properties of this LB/PAn-PTSA/GCE were studied using electrochemical impedance spectroscopy (EIS) and cyclic voltammetry. The LB/PAn-PTSA/GCE was used as a voltammetric sensor for determination of trace Ag+ at pH 5.0 using linear scanning stripping voltammetry. Under the optimal experimental conditions, the stripping current was proportional to the Ag+ concentration over the range from 6.0 × 10−10 mol L−1 to 1.0 × 10−6 mol L−1, with a detection limit of 4.0 × 10−10 mol L−1. The high sensitivity, selectivity, and stability of this LB/PAn-PTSA/GCE also demonstrated its practical utility for simple, rapid and economical determination of Ag+ in water samples.  相似文献

The complex of rutin-Cu (C81H86Cu2O48, abbreviated by Cu2R3, R = rutin) was synthesized and characterized by elemental analysis and IR spectra. Cyclic voltammetry (CV) and fluorescence spectroscopy were used to investigate the interaction of Cu2R3 with salmon sperm DNA. It was revealed that Cu2R3 could interact with double-stranded DNA (dsDNA) by a major intercalation role. Using Cu2R3 as a novel electroactive indicator, an electrochemical DNA biosensor for the detection of specific DNA fragment was developed and its selectivity for the recognition with different target DNA was assessed by differential pulse voltammetry (DPV). The target DNA related to coliform virus gene could be quantified ranged from 1.62 × 10−8 mol L−1 to 8.10 × 10−7 mol L−1 with a good linearity (r = 0.9989) and a detection limit of 2.3 × 10−9 mol L−1 (3σ, n = 7) was achieved by the constructed electrochemical DNA biosensor.  相似文献

We outline here the fabrication of a sensitive electrochemical DNA biosensor for the detection of sequence-specific target DNA. Zinc oxide nanowires (ZnONWs) were first immobilized on the surface of a glassy carbon electrode. Multi-walled carbon nanotubes (MWCNTs) with carboxyl groups were then dropped onto the surface of the ZnONWs. Gold nanoparticles (AuNPs) were subsequently introduced to the surface of the MWNTs/ZnONWs by electrochemical deposition. A single-stranded DNA probe with a thiol group at the end (HS-ssDNA) was covalently immobilized on the surface of the AuNPs by forming an Au-S bond. Scanning electron microscopy (SEM) and cyclic voltammetry (CV) were used to investigate the film assembly process. Differential pulse voltammetry (DPV) was used to monitor DNA hybridization by measuring the electrochemical signals of [Ru(NH3)6]3+ bounding to double-stranded DNA (dsDNA). The incorporation of ZnONWs and MWCNTs in this sensor design significantly enhances the sensitivity and the selectivity. This DNA biosensor can detect the target DNA quantitatively in the range of 1.0 × 10−13 to 1.0 × 10−7 M, with a detection limit of 3.5 × 10−14 M (S/N = 3). In addition, the DNA biosensor exhibits excellent selectivity, even for single-mismatched DNA detection.  相似文献

A novel and sensitive electrochemical DNA biosensor based on electrochemically fabricated polyaniline nanowire and methylene blue for DNA hybridization detection is presented. Nanowires of conducting polymers were directly synthesized through a three-step electrochemical deposition procedure in an aniline-containing electrolyte solution, by using the glassy carbon electrode (GCE) as the working electrode. The morphology of the polyaniline films was examined using a field emission scanning electron microscope (SEM). The diameters of the nanowires range from 80 to 100 nm. The polyaniline nanowires-coated electrode exhibited very good electrochemical conductivity. Oligonucleotides with phosphate groups at the 5′ end were covalently linked onto the amino groups of polyaniline nanowires on the electrode. The hybridization events were monitored with differential pulse voltammetry (DPV) measurement using methylene blue (MB) as an indicator. The approach described here can effectively discriminate complementary from non-complementary DNA sequence, with a detection limit of 1.0 × 10−12 mol l−1 of complementary target, suggesting that the polyaniline nanowires hold great promises for sensitive electrochemical biosensor applications.  相似文献

5-Hydroxytryptophan (5-HTP) was covalently grafted on the surface of glassy carbon electrodes (GCEs) using cyclic voltammetric method in a phosphate buffer solution. The prepared electrode, denoded as 5-HTP/GCE, was characterized by X-ray photoelectron spectroscopy, cyclic voltammetry and differential pulse voltammetry (DPV). Tryptophan grafted GCE (TRP/GCE) and 5-hydroxytryptamine grafted GCE (5-HTP/GCE) were also prepared by the same method for comparison. It was found that the electrocatalytic activities toward the oxidation of uric acid (UA) and ascorbic acid (AA) was in the order of 5-HT/GCE > 5-HTP/GCE > TRP/GCE for UA oxidation and 5-HT/GCE = 5-HTP/GCE > TRP/GCE for AA oxidation. However, the CV current sensitivity was estimated as 4:2:1 for 5-HTP/GCE:5-HT/GCE:TRP/GCE. The DPV peaks for UA and AA oxidation appeared at 0.07 V and 0.34 V versus SCE, respectively, allowing simultaneous determination in mixtures. A linearly response in the range of: 5.0 × 10−7 to 1.1 × 10−5 M with the detection limit (s/n = 3) of 2.8 × 10−7 M for UA determination, and a linear response in the range of: 5.0 × 10−6 to 1.0 × 10−4 M with the detection limit of 4.2 × 10−6 M for AA determination were obtained. This electrode was used for UA and AA determinations in human urine samples satisfactorily.  相似文献

The electrochemical behavior of bisphenol A (BPA) was investigated on Mg-Al layered double hydroxide (LDH) modified glassy carbon electrode (GCE) by cyclic voltammetry (CV), differential pulse voltammetry (DPV), linear sweep voltammetry (LSV) and chronocoulometry (CC). The cyclic voltammogram of BPA on the modified electrode exhibited a well defined anodic peak at 0.454 V in 0.1 M pH 8.0 phosphate buffer solution (PBS). The experimental parameters were optimized and the kinetic parameters were investigated. The probable oxidation mechanism was proposed. Under the optimized conditions, the oxidation peak current was proportional to BPA concentration in the range from 1 × 10−8 to 1.05 × 10−6 M with the correlation coefficient of 0.9959. The detection limit was 5.0 × 10−9 M (S/N = 3). The fabricated electrode showed good reproducibility, stability and anti-interference. The proposed method was successfully applied to determine BPA in plastic products and the results were satisfactory.  相似文献

A sensitive and novel DNA electrochemical biosensor for the detection of the transgenic plants gene fragment by electrochemical impedance spectroscopy (EIS) was presented. The well-dispersed carboxylic group-functionalized single-walled carbon nanotubes (SWNTs) were dripped onto the carbon paste electrode (CPE) surface firstly, and poly-l-lysine films (pLys) were subsequently electropolymerized by cyclic voltammetry (CV) to prepare pLys/SWNTs/CPE. The morphology of pLys/SWNTs films was examined using a field emission scanning electron microscope (SEM). The pLys/SWNTs films modified electrode exhibited very good conductivity. DNA probes were easily immobilized on the poly-l-lysine films via electrostatic adsorption. The hybridization events were monitored with electrochemical impedance spectroscopy using [Fe(CN)6]3−/4− as indicator. The PAT gene fragment from phosphinothricin acetyltransferase gene was detected by this DNA electrochemical sensor. The dynamic detection range of this sensor to the PAT gene fragment was from 1.0 × 10−12 to 1.0 × 10−7 mol/L. A detection limit of 3.1 × 10−13 mol/L could be estimated. The PCR amplification of NOS gene from the sample of a kind of transgenic modified bean was also detected satisfactorily by EIS.  相似文献

A highly sensitive electrochemical biosensor for the detection of trace amounts of methotrexate has been designed. Double stranded (ds)DNA molecules are immobilized onto a pretreated glassy carbon electrode (GCE(ox)) surface with Langmuir-Blodgett (LB) technique. The adsorptive voltammetric behaviors of methotrexate on DNA-modified electrode were explored by means of cyclic voltammetry (CV) and square wave voltammetry (SWV). The oxidation mechanism was proposed and discussed in this work. In addition, the optimum experimental conditions for the detection of methotrexate were explored, and the currents measured by SWV presented a good linear property as a function of the concentrations of methotrexate in the range of 2.0 × 10−8 to 4.0 × 10−6 mol L−1, with an LOD of 5.0 × 10−9 mol L−1. The method proposed was applied for the determination of methotrexate in pharmaceutical dosage and diluted human urine with wonderful satisfactory successfully.  相似文献

A novel electroactive material for ascorbic acid (AA) determination was successfully prepared by plating/potential cycling method. The cobalt film was first deposited on the surface of glassy carbon electrode (GCE) in CoSO4 solution by potential cycling, and then a cobalt film on the surface of GCE was activated by potential cycling in 0.1 mol L−1 NaOH. The electrochemical performance of the resulted film (Co/GCE) and factors affecting its electrochemical activity were investigated by cyclic voltammetry and amperometry. This film electrode exhibited good electrocatalytic activity to the oxidation of AA. This biosensor had a fast response of AA less than 3 s and excellent linear relationships were obtained in the concentration range of 3 × 10−7 to 1 × 10−4 mol L−1 with a detection limit of 2 × 10−7 mol L−1 (S/N = 3) under the optimum conditions. Moreover, the selectivity, stability and reproducibility of this biosensor were evaluated with satisfactory results.  相似文献

Nano-γ-Al2O3 is dispersed onto the glass carbon electrode (GCE) by polishing. This nanostructured modified GCE exhibits a great enhancement to the redox responses of 3-nitrobenzaldehyde thiosemicarbazone (3-NBT). In comparison with bare GCE, 3-NBT gives a more sensitive voltammetric response because of the nanoparticle’s unique properties. The lowest detectable concentration (3σ) of 3-NBT is estimated to be 1.18 × 10−6 M (accumulation for 4 min). The linear relationship between peak current and concentration of 3-NBT holds in the range 1.0 × 10−5 M to 1.0 × 10−4 M (r = 0.9981). The electrochemical properties of 3-NBT on this modified electrode have been investigated with various electrochemical methods. The results indicate that the transference of one electron and one proton involves electrode radical reaction processes I and II, respectively. The coverage value (Γ) of 1.62 × 10−9 mol cm−2 was calculated and the electrochemical parameters, diffusion coefficient D (2.54 × 10−3 cm2 s−1, 2.03 × 10−3 cm2 s−1) and reaction rate constant ks (5.9573 s−1, 7.15 × 10−2 cm s−1) were obtained for quasi-reversible system I and irreversible system II, respectively.  相似文献

A new method for the determination of nimesulide was established based on the multiwalled carbon nanotubes (MWCNTs) modified glassy carbon electrode (MWCNTs/GCE). In 0.2 M PBS (pH 6.6) buffer solution, the MWCNTs/GCE showed a remarkable catalytic and enhancement effect on reduction of the nimesulide. The reduction peak potential of nimesulide shifted positively from −0.665 V at bare GCE to −0.553 V at MWCNTs/GCE, and the sensitivity increased ca. 7 times. A linear dynamic range of 3.2 × 10−7-6.5 × 10−5 M (R = 0.9992) with a detection limit of 1.6 × 10−7 M was obtained. The electrochemical behaviors of nimesulide were studied and electron-transfer coefficient (α = 0.45), proton number (X = 1) and electron-transfer number (n = 2) have been determined. This method has been used to determine the content of nimesulide in medical tablets. The recovery was determined to be 93.2-106.2% by means of standard addition method. Compared with UV-vis spectrometry, the method was not remarkable difference.  相似文献

A multiwall carbon nanotubes (MWNTs)-chitosan modified glassy carbon electrode (GCE) exhibits attractive ability for highly sensitive cathodic stripping voltammetric measurements of bromide (Br). In pH 1.8 H2SO4 solution, a substantial increase in the stripping peak current of Br (compared to bare GCE and chitosan modified GCE) is observed using MWNTs-chitosan modified electrode. Operational parameters were optimized and the electrochemical behaviors of Br were studied by different electrochemical methods. The kinetics parameters were measured, the number of electron transfer (n) was 1 and the transfer coefficient (α) is 0.17. A wide linear calibration range (3.6 × 10−7-1.4 × 10−5 g mL−1) was achieved, with a detection limit of 9.6 × 10−8 g mL−1. The mechanism of electrode reaction was fully discussed.  相似文献

In this paper, a new DNA hybridization detection strategy was developed based on the immobilization of capture probe DNA on a chitosan (CS)–carbon nanotubes (CNTs) composite modified glassy carbon electrode (CS–CNTs/GCE) and the use of a copper complex, [Cu(bpy)(MBZ)2(H2O)] (bpy = 2,2′-bipyridine, MBZ = p-methylbenzoate), as a new redox hybridization indicator. The electrochemical characterization experiments showed that the nanocomposite film of CS–CNTs could effectively immobilize the capture probe DNA and greatly improve the electron-transfer reactions of the electroactive molecules. Electrochemical and fluorescent spectroscopic analysis revealed that the polypyridyl copper complex of [Cu(bpy)(MBZ)2(H2O)] bound to DNA via a typical intercalation mode. Surface studies further showed that the copper complex can discriminate between double-stranded and single-stranded DNA that immobilized on the surface of CS–CNTs/GCE. When being utilized as a redox indicator for the detection of hybridization for short DNA species related to phosphinothricin acetyltransferase (PAT), the indicator showed good specificity for recognizing the complementary, three-base mismatched and non-complementary DNA. Under the optimized conditions, the oxidation peak currents of the copper complex enhanced linearly with increases in the concentration of the complementary sequence in the range from 5.0 × 10−10 to 1.0 × 10−8 M. A detection limit of 5.0 × 10−10 M was also obtained based on the constructed DNA biosensor.  相似文献

The heterogeneous electron transfer reaction for the reduction of some nitroso aromatic derivatives in aqueous-alcoholic medium was studied on both mercury and glassy carbon electrodes (GCE) by using cyclic voltammetry (CV) and scanning electrochemical microscopy techniques (SECM).The nitrosoaromatic derivatives followed a two-electron two-proton mechanism producing a quasi-reversible overall process. This strongly pH dependent mechanism varied from ECCE mechanism at pH < 8.5 to ECEC mechanism at pH > 8.5.The apparent heterogeneous rate constant for the reduction of the nitroso derivatives was calculated using CV or SECM. The rate constant for the electron transfer process depends on the nature of the electrode material. The heterogeneous rate constant on the GCE is almost two orders of magnitude smaller than that on mercury electrode i.e. (3.4 ± 0.3) × 10−3 cm s−1 on Hg and (7.0 ± 1.0) × 10−5 cm s−1 on GCE, for the same nitroso compound and pH.The heterogeneous rate constant values were checked by comparison between experimental and simulated cyclic voltammograms.  相似文献

The development of a simple and efficient method to 2-(2-nitrophenyl)-1H-benzimidazole (NB) electrochemical determination using a polymer film coated chemically modified electrode is described. A glassy carbon (GC) electrode was modified employing an electro-polymerized film of 1-naphtylamine (1-NAP) followed by an over-oxidation treatment in 0.2 M sodium hydroxide solution (poly-1-NAPox electrode).The electrochemical behaviour of NB at the poly-1-NAPox electrode was investigated in a mixture of 10% ethanol + 90% buffer solution (pH 2) by cyclic voltammetry (CV) and square-wave voltammetry (SWV). The experimental results suggested that the poly-1-NAPox electrode had a good effect on NB electrochemical response because it avoided the electrode surface fouling as a consequence of the adsorption of NB reduction products, which was found when a bare GC electrode was employed as the working electrode. The NB cathodic current was dependent on the polymeric film over-oxidation degree (α).NB could be determined in the range from 2 × 10−6 to 5 × 10−5 M. The NB detection and quantification limits were 5 × 10−7 and 1.7 × 10−6 M, respectively. The percent relative standard deviation of the peak current to 10-replicated measurement using 1.2 × 10−5 M NB solution was 1.4%. The method showed to be rapid, simple and with a good sensitivity.  相似文献

A carbon-paste electrode (CPE) chemically modified with the cobalt(II)-4-methylsalophen (CoMSal) as a Schiff base complex was used as a highly sensitive and fairly selective electrochemical sensor for simultaneous determination of minor mounts of ascorbic acid (AA) and cysteine. This modified electrode shows very efficient electrocatalytic activity for anodic oxidation of both AA and cysteine via substantially decreasing of anodic overpotentials for both compounds. The mechanism of electrochemical oxidation of both AA and cysteine using CoMSal-modified electrode was thoroughly investigated by cyclic voltammetry and polarization studies. Results of cyclic voltammetry (CV) and differential pulse voltammetry (DPV) using this modified electrode show two well-resolved anodic waves for the oxidation of AA and cysteine, which makes it possible for simultaneous determination of both compounds. A linear range of 1 × 10−4 to 5 × 10−7 M for cysteine in a constant concentration of 1 × 10−4 M AA in buffered solution (as a background electrolyte) was obtained from DPV measurements using this electrode. The linear range, which is obtained for AA in the presence of 1 × 10−4 M cysteine, was in the range of 1 × 10−4 to 1 × 10−6 M. The modified electrode has good reproducibility (RSD ≤ 2.5%), low detection limit (sub-micromolar) and high sensitivity for the detection of both AA and cysteine with a very high stability in its voltammetric response. Differential pulse voltammetry using the modified electrode exhibited a reasonable recovery for a relatively wide concentration range of cysteine spiked to a synthetic human serum sample.  相似文献

A self-assembled bilayer lipid-like membrane (BLM) supported on glassy carbon electrode (GCE) was fabricated using 5,5-ditetradecyl-2-(2-trimethyl-ammonioethyl)-1,3-dioxane bromide (DTDB) for epinephrine (EP) determination in the presence of ascorbic acid (AA). This modified electrode (DTDB/GCE) has strong membrane adsorption accumulation and electrocatalytic ability toward EP and AA. The oxidation of EP was controlled by double step adsorption accumulation process of the DTDB-BLM. The parameters of fitted Langmuir isotherm Γmax, BADS, and ΔGADS values were determined as 1.0×10−11 mol cm−2, 2.04×106 dm3 mol−1, and −45.17 kJ mol−1 for the fist step for EP concentration less than 1 mM, and 4.92×10−11 mol cm−2, 7.35×104 dm3 mol−1, and −37.1 kJ mol−1 for the second step for EP concentration higher than 1 μM. The DPV peaks for EP and AA oxidations were appeared at 0.220 and 0.085 V versus SCE, respectively, allowing the determination of EP in the presence of high concentration of AA. The advantage of DTDB-BLM was demonstrated experimentally in comparison with other three BLMs, and attributed to the dioxane group as well as the suitable length of the carbon chain of DTDB molecule. The current response of the DTDB/GCE was fast and reproducible, suitable for the electrochemical sensing in flow-injection systems. A linear range of 1×10−8 to 1×10−4 M EP was preliminary obtained using a simple setup.  相似文献

Stable Nafion-Au colloids were immobilized on a glassy carbon electrode (GCE) for detection of β-agonist clenbuterol by electroanalysis. The Au colloids were prepared by a one-step electrodeposition onto GCE, with obvious electrocatalytic activity present. The negatively charged Nafion film was an efficient barrier to negatively charged interfering compounds, resulting in accumulation of positively charged clenbuterol at the Nafion film. The electrochemical characters of the electrode during various modified steps in a redox probe system of K4[Fe(CN)6]/K3[Fe(CN)6] were confirmed by cyclic voltammetry (CV) and AC-impedance. In Britton-Robinson (B-R) buffer solution (pH = 2.0) and the potential range of −0.2 to 1.2 V, the Nafion-Au colloid modified electrode, compared to a bare GCE, exhibits obvious electrocatalytic activity towards the redox of clenbuterol by greatly enhancing the peak current with a linear calibration curve from 8.0 × 10−7 to 1.0 × 10−5 mol/L and a detection limit of (1.0 × 10−7 mol/L) (R = 0.996). The modified electrode shows high sensitivity, selectivity and reproducibility. The recovery for detecting clenbuterol (∼10−6 mol/L) in human serum is up to 98.19%.  相似文献
