The aim of the present study was to evaluate and compare the phenolic content and antioxidant and antimicrobial properties of olive leaf extracts obtained by ultrasound-assisted extraction (UAE) and conventional extraction (CSE). UAE of olive leaf extracts yielded a higher total phenolic content (TPC) and total flavonoid content (TFC) of 14.31% and 19.50%, respectively. Higher antioxidant activities were found from the extracts prepared with UAE (for 18.5%, 12.5%, 10.9% and 17.6% higher determined by DPPH, ABTS, FRAP and CUPRAC methods, respectively). Good antibacterial inhibitory activity (as MIC and MBC) was observed against both Y. enterocolitica and S. aureus (1.40 ± 0.40 mg mL−1 and 4.00 ± 1.60 mg mL−1, respectively) with the extract prepared with UAE. In conclusion, olive leaf extracts prepared with UAE exhibited higher antioxidant and antimicrobial activities against common food-borne pathogens than CSE extracts and thus could be beneficial in ensuring food quality and food safety.  相似文献

This study investigated the antiglycation capacity and antioxidant activities of fifteen Thai rice varieties. Purple variety, Kum Rai showed the highest value of total phenolic content (245.06 ± 7.87 mg GAE/gram extract dry weight), DPPH radical scavenging activity (92.33 ± 0.49 mg Trolox/gram extract dry weight), ABTS radical scavenging activity (221.01 ± 0.25 mg Trolox/gram extract dry weight) and percentage inhibition of AGEs formation (82.03 ± 0.19%). We found that, total phenolic content and antioxidant activities (ABTS and DPPH assay) in rice varieties were highly correlated (P < 0.05) with their antiglycation capacity. These results indicated that the total phenolic content was responsible for antioxidant and antiglycation capacities of rice samples. This study is the first report on a correlation between anti‐AGEs capacity and antioxidant activities of Thai rice varieties. Our data have provided useful information for selection of rice varieties for the bioactive compounds that may improve the health of the aged and diabetic complications.  相似文献

The present study investigates the phenolic contents and antioxidant activities of silk sericin extracted (SSE) from Thai mulberry silkworms (Bombyx mori) and non-mulberry silkworm (Samia ricini). The SSE from all strains revealed the mainly presence of flavonoid compounds including (+)-catechin (20.63–145.64 mg/100 g), quercetin (7.89–35.95 mg/100 g), and (−)-epicatechin (3.36–44.19 mg/100 g). The antioxidant activity of the SSE with water from Chokumnoui 1 was found to have low EC50 values (0.96 mg/mL), the concentrations of the SSE that exhibit 50% reduction in DPPH, and highest scavenging of ABTS· + radicals (1.73 mg TEAC/g) and highest reduce TPRZ-Fe (III) complex to TPTZ-Fe (II) (8.03 mg Fe(II)/g), thus indicating high antioxidant activities. Total phenolic content and total flavonoid content of the SSE showed positive correlated to the scavenging activity of DPPH and ABTS· + radicals and the ferric reducing ability (FRAP assay).  相似文献

The objective of this study was to assess antioxidant and antiproliferative activities of four different Turkish pomegranate varieties (Hatay, Hicaz, Adana and Antalya) using an in vitro HepG2 cancer cell model. All the pomegranate extracts employed in this study significantly diminish the proliferation of HepG2 cells in a dose‐dependent manner. The total phenolic acid, anthocyanin and flavonoid contents for each of the four varieties were determined. The Hatay pomegranate variety had the highest total phenolic acid (337.4 ± 2.34 mg/100 g) and flavonoid (58.42 ± 2.25 mg/100 g) contents of the pomegranates examined. Antioxidant activities of the pomegranates were determined using DPPH and ABTS radical scavenging assays. The lack of correlation between colour index value and antioxidant–antiproliferative activities suggested that phenolic acids and flavonoids are predominant compounds influencing pomegranate's bioactivity rather than anthocyanins. Individual phenolic acids found in Hatay pomegranates were determined, using an HPLC system, as gallic acid being the most predominant phenolic compound.  相似文献

Phenolic compounds, antioxidant and antibacterial activities of defatted Moringa oleifera seed flour (DMF) were investigated. The total phenolic and flavonoid contents were measured using colorimetric methods. Free phenolic acid and flavonoid profiles were analyzed by high performance liquid chromatography, while antioxidant capacities were evaluated using scavenging assays of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), ferric reducing antioxidant power and total antioxidant capacity. The results showed that extractability of phenolic compounds was significantly higher (p < 0.05) in bound phenolic extract (4173.00 ± 32.22 mg gallic acid equivalents (GAE)/100 g) than in free phenolic extract (780.00 ± 14.2 mg GAE/100 g) and it showed higher antioxidant and antimicrobial activities. The IC50 value for DPPH radical scavenging activity was 0.9 ± 0.05 and 14.9 ± 0.07 mg/mL for bound phenolic and free phenolic extracts, respectively. Bound phenolic extract was more effective (minimum inhibitory concentration (MIC), 0.06–0.157%) than free phenolic extract (MIC, 0.117–0.191%) against tested bacteria. Ten phenolic compounds (gallic acid, p-coumaric acid, ferulic acid, caffeic acid, protocatechuic acid, cinnamic acid, catechin, epicatechin, vanillin and quercetin) were identified and quantified in both extracts. These natural plant phenolics from Moringa seeds could be a good source of antioxidants and antibacterials for food and pharmaceutical industries.  相似文献

Pressurised liquid extraction (PLE) of antioxidant compounds from bitter gourd fruits (Momordica charantia) in aqueous ethanolic solvent was investigated using response surface methodology at laboratory scale to understand key impact of extraction variables. Extraction efficiency was optimised by measuring the yield of extraction, total phenolic content (TPC), total flavonoid content (TFC), ferric reducing/antioxidant power assay (FRAP) and radical scavenging activity (RSA). The optimal extraction conditions were reached at 80% ethanol concentration, 10‐min extraction time and at 160 °C. Under these extraction conditions, values of TPC (5.40 ± 0.30 g GAE per 100 g), TFC (1.50 ± 0.10 g QE per 100 g), FRAP (778.55 ± 10 μmol eq Fe (II) g?1), yield (178.50 ± 5.50 mg g?1 dc) and RSA (75.50 ± 4.50%) were achieved. Furthermore, statistical analysis revealed that antioxidative attributes of bitter gourd extract were strongly and positively correlated with extraction temperature and ethanol concentration rather than processing time. This study illustrated that PLE has the potential to extract antioxidant compounds from tropical fruit vegetables in an accelerated manner. Furthermore, influential parameters affecting the process could be optimised for further industrial intake.  相似文献

Abstract: The phenolic acid and flavonoid contents of Malaysian Tualang, Gelam, and Borneo tropical honeys were compared to those of Manuka honey. Ferric reducing/antioxidant power assay (FRAP) and the 1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) radical‐scavenging activities were also quantified. All honey extracts exhibited high phenolic contents (15.21 ± 0.51– 42.23 ± 0.64 mg/kg), flavonoid contents (11.52 ± 0.27– 25.31 ± 0.37 mg/kg), FRAP values (892.15 ± 4.97– 363.38 ± 10.57 μM Fe[II]/kg), and high IC50 of DPPH radical‐scavenging activities (5.24 ± 0.40– 17.51 ± 0.51 mg/mL). Total of 6 phenolic acids (gallic, syringic, benzoic, trans‐cinnamic, p‐coumaric, and caffeic acids) and 5 flavonoids (catechin, kaempferol, naringenin, luteolin, and apigenin) were identified. Among the Malaysian honey samples, Tualang honey had the highest contents of phenolics, and flavonoids, and DPPH radical‐scavenging activities. We conclude that among Malaysian honey samples, Tualang honey is the richest in phenolic acids, and flavonoid compounds, which have strong free radical‐scavenging activities.  相似文献

以3种不同的青稞为原料,采用不同的提取溶剂(水、60%乙醇、60%甲醇、60%丙酮、95%乙醇、100%甲醇、100%丙酮)对青稞进行提取。对各提取物的总酚、总黄酮进行测定,同时采用3种抗氧化方法:二苯代苦味酰自由基(DPPH)、2,2’-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐自由基(ABTS)和总抗氧化能力(FARP)评价青稞提取物的抗氧化活性。结果表明,95%乙醇和60%丙酮更有利于青稞多酚的提取。藏青2000的各溶剂提取物的总酚、总黄酮及抗氧化活性普遍高于相应溶剂的循化蓝青稞和香格里拉绿青稞。藏青2000的60%丙酮提取物含有最高的总酚含量,达到211.92mg GAE/100g DW;在60%乙醇提取时具有最高的总黄酮含量,达到60.11mg RT/100g DW;在60%乙醇提取时具有最强的DPPH清除能力,达到80.08%;而用60%丙酮提取时,具有最强的ABTS清除能力和总抗氧化能力,分别达到了1.85,9.28 mmol TEAC/100g DW。总酚含量与抗氧化活性均具有显著的相关性,FRAP法与DPPH、ABTS法具有极显著的相关性。综上表明,青稞富含总酚成分,是一种潜在的天然抗氧化剂资源。  相似文献

In this study, the antioxidant, anticholinesterase, and anti-tyrosinase properties of (hexane, acetone, methanol, and water) extracts of Ferula elaeochytris and Sideritis stricta were determined with the total phenolic and flavonoid contents. The phenolic profile of the methanol and water extracts was analysed using HPLC-DAD. Protocatechuic acid was found as the major phenolic compound in the methanol (116.3 ± 3.1 µg/g) and water extracts (69.4 ± 1.3 µg/g) of F. elaeochytris. Coumarins (253.9 ± 4.1 µg/g) and catechin hydrate (175.2 ± 2.9 µg/g) were the most abundant phenolic compounds in the methanol and water extracts of S. stricta. β-carotene–linoleic acid, DPPH?, ABTS?+, CUPRAC, and metal-chelating assays were used to evaluate antioxidant properties of the extracts. The methanol and water extracts of F. elaeochytris and the acetone and methanol extracts of S. stricta containing the highest amount of total phenolic and flavonoid contents showed the highest antioxidant activities in β-carotene–linoleic acid, DPPH?, ABTS?+, and CUPRAC assays. The enzyme inhibitory potential of extracts was investigated against key enzymes involved in neurodegenerative (acetylcholinesterase (AChE) and butyrylcholinesterase (BChE)) and skin (tyrosinase) disorders. In the cholinesterase inhibitory assays, the hexane extracts of two species exhibited the best activity against AChE, while the hexane extract of F. elaeochytris and the methanol extract of S. stricta observed to be the most active against BChE. As for anti-tyrosinase activity results of extracts, the only acetone and methanol extracts showed mild inhibitory activity for both species.  相似文献

To investigate the relationship between bioactive compounds and antioxidant property, total phenolic content (TPC), total flavonoid content (TFC), individual and total isoflavone content of soymilk and their correlations with oxygen‐radical‐absorbing capacity (ORAC), ferric reducing antioxidant power (FRAP) and 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging capacity values were evaluated and compared. Results showed that TPC, TFC, isoflavones (including concentration and profiles) and antioxidation activity significantly varied among ten tested soybean cultivars. Significant correlations were established between DPPH and TFC (= 0.553, < 0.01), DPPH and TPC (= 0.753, < 0.01), FRAP and TFC (= 0.599, < 0.01) and FRAP and TPC (= 0.616, < 0.01). Positive linear correlations were found between subtotal, total isoflavones and ORAC. Unlike other isoflavone monomers, aglycone isoflavones correlated positively with DPPH and FRAP significantly. DPPH well correlated with FRAP, whereas neither DPPH nor FRAP correlated with ORAC. TPC, TFC, individual and total isoflavone values are potentially useful for soymilk antioxidant activity assessment.  相似文献

The methanolic extract of raw Abrus precatorius (Jequirity bean, red‐ and black‐coloured seed coat) seeds contains a total phenolic content of 14.2 ± 0.39 g catechin equivalent/100 g extract. An analysis of phenolic profile revealed the presence of syringic acid, caffeic acid and (?)‐epicatechin in the methanolic extract of A. precatorius seeds. Encouraging levels of ferric reducing/antioxidant power (FRAP, 1247 mmol Fe[II]/mg extract), inhibition of β‐carotene bleaching (53.37%), 2,2‐Diphenyl‐1‐picryl‐hydrazyl (DPPH, 62.13%) and superoxide (64.47%) radical scavenging activities were exhibited by the extract of raw samples. Inhibition characteristics of 68.69%α‐amylase and 51.74%α‐glucosidase enzyme were also recorded under in vitro starch digestion bioassay. Sprouting + oil‐frying caused a significant increase in the total phenolic content and also substantially improved the antioxidant and free radical scavenging capacities, while soaking + cooking as well as open‐pan roasting treatments showed diminishing effects.  相似文献

Spices and condiments are rich sources of potent antioxidants. In the present investigation, total equivalent antioxidant capacities (TEAC) of 39 spices were studied using 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) scavenging, and cupric reducing antioxidant capacity (CUPRAC) assays. Their total phenolic contents (TPC) and total flavonoid contents (TFC) were also determined. TEAC, TPC, and TFC varied significantly (P?<?0.05) among these spices. ABTS, DPPH, and CUPRAC assay values of studied spices ranged from 1.42 to 112.94 mg ascorbic acid equivalents (AAE)/g, 1.14 to 91.09 mg Trolox equivalents (TE)/g, and 0.52 to 54.47 mg TE/g, respectively (dry weight basis; DW). Based on Folin–Ciocalteu assay, TPC ranged from 2.93 to 160.55 mg of gallic acid equivalents (GAE)/g DW. Strong correlations between TPC and TEAC values (R?=?0.966, 0.825, and 0.954 for ABTS, DPPH, and CUPRAC, respectively) were found. This indicates that phenolic compounds are potent antioxidants in these spices. Principal component analysis (PCA) indicated that cloves (flower buds of Syzygium aromaticum) had the most distinct and potent antioxidant capacity, followed by allspice (fruits of Pimenta dioica) and cinnamon (bark of Cinnamomum verum). Results of the present study provide adequate evidence that polyphenols are responsible for their compelling antioxidant capacities of studied spices. Thus, consumption of antioxidant-rich spices such as cloves, allspice, and cinnamon can significantly prevent oxidative stress in the human body.  相似文献

The methanolic extract of ox-eye bean [Mucuna gigantea (Willd) DC.] contained total free phenolic content of 14.80±1.28 g catechin equivalent/100 g extract dry matter. Encouraging levels of ferric reducing/antioxidant power (FRAP, 1,023 mmol Fe[II]/mg extract), inhibition of β-carotene degradation (59.35%) and radical scavenging activity against DPPH (72.12%) and superoxide (43.11%) were exhibited by the raw samples. Further, it also recorded 82.17% of α-amylase and 91.26% of α-glucosidase enzyme inhibition characteristics. Sprouting+oil-frying caused a apparent increase on the total free phenolic content and also significant improvement on the antioxidant and free radical scavenging capacity of methanolic extract, while soaking+cooking as well as open-pan roasting treatments showed diminishing effects. Moreover, inhibition of α-amylase and α-glucosidase enzyme activities was declined to 22.82 and 45.47%, respectively during sprouting+oilfrying treatment, which are more desirable for the dietary management of type II diabetic patients.  相似文献

Dry beans are rich sources of dietary fiber and phytochemicals such as flavonoids and phenolics. In the current study, we determined the effects of two processing methods (soaking and toasting) on total phenolics, flavonoids, proanthocyanidin and antioxidant potential by 2,2-diphenyl-1-picrylhydrazyl (DPPH) activity and ferric reducing antioxidant potential (FRAP) in selected dry beans (red kidney beans (K), black-eyed peas (B), pinto beans (P) and soy beans (S)).Total phenolics (mg/g dwb) expressed as gallic acid equivalents (GAE), total flavonoids (mg/g dwb) as catechin equivalents (CE) and proanthocyanidin expressed as leucocyanidin equivalent (mg LE/g) ranged from 3.42 to 7.21, 0.61 to 0.84 and 0.51 to 3.13 in raw beans; 3.58 to 6.94, 0.19 to 0.99 and 0.43 to 3.13 in soaked beans and 4.55 to 9.52, 0.23 to1.00 and 0.20 to 3.25 in toasted beans, respectively.FRAP (μg/g) in raw, soaked and toasted dry beans ranged from 0.00097 to 0.00424 while DPPH(T30) (%) ranged from 43.9 to 62.61.Our results indicate that processing methods (soaking and roasting) influenced total phenolic, flavonoid and antioxidant contents (DPPH, FRAP) in selected dry beans.  相似文献

This study reports the phenolic composition, antioxidant, antibacterial, larvacidal, and cytotoxic activity of methanol and acetone extracts of Hyacinthella lineata leaves and bulbs. The phenolic composition of H. lineata was determined by HPLC. The most abundant component was gallic acid (421.9µg/g). The β-carotene/DPPH/ABTS/FRAP decoloration method was used to estimate the total antioxidant activity. The total antioxidant activity was the highest for bulb-methanol fraction (65.41 ± 0.05%). The total phenolic content for leaves-methanol extract of the plant was determined as 6.56 ± 4.027mg/mL gallic acid equivalents. Analysis of the antibacterial activity showed that the methanolic-bulb extract are efficient against gram positive and gram negative bacteria. The cytotoxic effect on Artemia salina was assessed by Brine shrimp assay. Brine shrimp lethality assay showed that LC50 values of HBM were obtained as 4.105 ± 2.42μg/ml. The bulb extract of H. lineata showed the highest larvicidal activity against Cx pipiens with value LC50 (64.3275μg/ml). This study suggested that H. lineata may be used as a potential source of antioxidant, and for their biological activity, cytotoxic and antimicrobial properties.  相似文献

The phenolic profiles and antioxidant activity of litchi pulp of 13 varieties were investigated. The free, bound and total phenolic contents were 66.17–226.03, 11.18–40.54, and 101.51–259.18 mg of gallic acid equivalents/100 g, respectively. The free, bound and total flavonoid contents were 16.68–110.33, 10.48–22.75, and 39.43–129.86 mg of catechin equivalents/100 g, respectively. Free phenolics and flavonoids contributed averagely 80.1% and 75% to their total contents, respectively. Six individual phenolics (gallic acid, chlorogenic acid, (+)-catechin, caffeic acid, (?)-epicatechin, and rutin) were detected in litchi pulp by HPLC. The contents of each compound in free and bound fractions were determined. Significant varietal discrepancy in antioxidant activity was also found by FRAP and DPPH scavenging capacity methods. Antioxidant activity was significantly correlated with phenolic and flavonoid contents. Thus, phenolics and flavonoids exist mainly in the free form in litchi pulp. There were significant varietal differences in phytochemical contents and antioxidant activity of litchi pulp.  相似文献

Eight major mulberry cultivars [Nakhonratchasima 60 (NS 60), Buriram 60 (BR 60), Chumphon (CP), Wavee (WV), Chaingmai (CM), Pikultong (PT), Kamphaengsaen (KS) and Kamnanchul (KJ)] cultivated in Thailand were assessed for their flavonoid and phenolic acid composition using HPLC and tested for antioxidant potential using 2,2‐diphenyl‐1‐picrylhydrazyl hydrate (DPPH) assay. The total phenolic content (TPC) ranged from 104.78 to 213.53 mg GAE/100 g DW, and total flavonoid content (TFC) ranged from 69.58 to 211.01 mg CE/100 g DW. The major flavonoid compounds in mulberry fruit cultivars were (+)‐catechin (309.26–750.01 mg/100 g DW), procyanidin B1 (62.59–224.41 mg/100 g DW), quercetin (5.36–58.42 mg/100 g DW), rutin (18.73–26.90 mg/100 g DW) and (?)‐epicatechin (8.47–29.21 mg/100 g DW). Gallic acid, cinnamic acid and p‐hydroxybenzoic acid were found to be the major phenolic acids in mulberry fruit cultivars. The gallic acid and cinnamic acid contents ranged from 7.33 to 23.90 mg/100 g DW and from 11.64 to 15.05 mg/100 g DW, respectively. p‐Hydroxybenzoic acid content ranged from 1.77 mg/100 g DW (PT) to 7.13 mg/100 g DW (KJ). DPPH‐scavenging ability was excellent for ethanolic extract of NS 60, and EC50 value of NS 60 (241.83 μg mL?1) was significantly lower than those of the others (< 0.05). TPC and TFC of the mulberry fruit were positively correlated with the DPPH‐scavenging ability.  相似文献

In this study, fifty‐one monofloral Sardinian honeys from ten various floral origins were screened for their phenolic content, antioxidant activity, colour and electrical conductivity. The total phenolic amounts have been evaluated by Folin–Ciocalteu method, whereas quantification of several phenolic compounds (phenolic acids and flavonoids) has been carried out by HPLC‐DAD technique. The richest sample in phenolic compounds resulted strawberry tree honey with about 40 mg GAE/100 g, as well FRAP test and DPPH˙ test confirm that antioxidant activity of strawberry tree honey extract exceed both honey extracts and synthetic antioxidants like BHA and BHT. Among the studied phenolic compounds a total of five phenolic acids (ferulic, syringic, trans‐cinnamic, chlorogenic and p‐hydroxycinnamic) and nine flavonoids (catechin, kaempferol, rutin, quercetin, luteolin, apigenin, galangin, pinocembrin and pinobanksin) were identified. Our results show good correlations between total polyphenol amount and antioxidant activity and between colour and electrical conductivity.  相似文献

In this study, the chemical composition, total phenolic content and antioxidant activity (DPPH, ORAC and FRAP assays) of A. subrufescens and P. ostreatus, cultivated with handmade and commercials supplements, were compared. Additionally, the compounds ergosterol, saccharopine, and hexitol were identified in A. subrufescens by HPLC-MS/MS. The antioxidant compound p-coumaric acid and dihexoses was found in both mushroom species. A. subrufescens presented higher total phenolic content (73.8 ± 0.6 mg GAE 100 g−1) and antioxidant activity than P. ostreatus (16.6 ± 0.5 mg GAE 100 g−1). The handmade supplement based on the waste of noble grains presented statistically similar phenolic content to the mushrooms cultivated with commercial ones Spawn Mate II SE (86.1 ± 1.4 and 92.9 ± 0.3 mg GAE 100 g−1, respectively). Therefore, the results support the use of handmade supplements based on agro-wastes as a viable alternative to the use of high-cost commercial ones.  相似文献

ABSTRACT: Raspberry (Aksu Kırmızısı, Rubin, Newburgh, Hollanda Boduru, Heritage) and blackberry (Bursa 1, Bursa 2, Jumbo, Chester) cultivars were assayed for antioxidant activity (determined as 2,2-azino-di-[3-ethylbenzothialozine-sulphonic acid][ABTS], 2,2-diphenyl-1-picrylhydrazyl radical [DPPH], and cupric ion reducing antioxidant capacity [CUPRAC]), total phenol, total flavonoid, and total anthocyanin contents. In addition, 10 anthocyanins and anthocyanidins were determined in raspberry and blackberry by liquid chromatography-mass spectrometry (LC-MS/MS). Raspberry and blackberry had the highest ABTS, DPPH, CUPRAC, total phenol, and total flavonoid contents in methanol extracts, whereas total anthocyanin contents were the highest in water extracts. The antioxidant activity of the raspberry and blackberry was directly related to the total amount of phenolic compounds detected in the raspberry and blackberry. All antioxidant activity values were highly correlated with anthocyanin content in blackberry (0.93 ≤ r ≤ 0.99, P = 0.05). On the other hand, high correlation between total flavonoid content and antioxidant activity was recorded in water extract of blackberry (0.91 ≤ r ≤ 0.93, P = 0.05). ABTS value was highly correlated with total flavonoid content in methanol extract (r = 0.90), whereas total flavonoid content was relatively less correlated with DPPH (r = 0.85) and CUPRAC (r = 0.89).  相似文献
