Leuconostc mesenteroides B-512F and L. mesenteroides B-742 were cultivated in clarified cashew apple juice to produce prebiotic oligosaccharides. Yeast extract (20 g/L); K2HPO4 (g/L) and sucrose (50 g/L) were added to the juice to promote the microbial growth and dextransucrase production. Initial pH was adjusted to 6.5 with H3PO4. Fermentations were carried out at 30 °C and 150 rpm for 24 h. The prebiotic effect of the fermented cashew apple juice, containing oligosaccharides, was evaluated through the Lactobacillus johnsonii B-2178 growth. L. johnsonii was incubated for 48 h using fermented cashew apple juice as substrate. Lactobacillus growth was compared to the microbial growth in non-fermented juice and in MRS broth. L. johnsonii growth in the fermented cashew apple juice was threefolds the observed growth in the non-fermented juice.  相似文献

The stability of nonfermented cashew apple juice (control) and the fermented juice with Lactobacillus casei NRRL B-442 (sweetened and nonsweetened samples) was investigated along the storage at 4 °C for 42 days. The viability of the probiotic bacteria, sugars, lactic acid, and vitamin C content besides color, antioxidant activity, enzyme activity, and sensory characteristics of probiotic cashew apple juice was evaluated. It was observed that viable cell counts increased in the probiotic cashew apple containing sucrose (8 % w/v) along the storage period. Viable cell counts were always higher than 8.00 Log CFU/ml throughout the storage period. Ascorbic acid loss was higher in the nonfermented juice (40 %) compared to the fermented juice (23 %). The same behavior was observed for antioxidant activity and total polyphenolic compounds content, conferring nutritional benefits to this functional food. Browning reactions and nutritional breakdown caused by enzymes were minimized in the fermented samples during storage. In the fermented sample, higher reduction (>70 %) in the polyphenol oxidase enzyme activity was observed. During storage, the increase in the chroma values from 3.2 to 5.0 indicated that the yellow color became more vivid. Sensory analysis of the fermented cashew apple juice revealed that the product was well-accepted, with acceptance percentages above 80 % for the sweetened juice at the end of the storage period.  相似文献

This study was carried out to optimize the fermentation pH and temperature and also the fermentation time for development of a new probiotic fermented drink with cantaloupe juice. The fermented juice was subjected to a storage study during 42?days at 4?°C. The initial pH and temperature influenced the growth of Lactobacillus casei in cantaloupe juice. However, on the microbial viability only the effect of temperature was significant. Optimum conditions for a satisfactory growth and viability of L. casei in cantaloupe juice were as follows: pH?=?6.1 and temperature?=?31?°C. The fermentation time of 8 hours was selected as the optimal fermentation time to prepare the probiotic cantaloupe juice. The cell viability was 8.3 log CFU mL?1 at the end of the fermentation. This level was kept over the 42?days of refrigerated storage. The consistent growth and viability of the probiotic microorganism in cantaloupe juice during fermentation and storage suggested that melon is a suitable vehicle for L. casei delivering. The fermented juice arises as a good alternative to the traditional dairy based products.  相似文献

This study describes the extraction and characterisation of cashew apple polyphenol oxidase (PPO) and the effect of wounding on cashew apple phenolic acid composition, PPO activity and fruit browning. Purification factor was 59 at 95% (NH4)2SO4 saturation. For PPO activity, the optimal substrate was catechol and the optimum pH was 6.5. PPO Km and Vmax values were 18.8 mM and 13.6 U min−1 ml−1, respectively. Ascorbic acid, citric acid, sodium sulphite and sodium metabisulphite decreased PPO activity, while sodium chloride increased PPO activity. Wounding at 2 °C and 27 °C for 24 h increased PPO activity but storage at 40 °C reduced PPO activity. Gallic acid, protocatechuic acid and cinnamic acid (free and conjugate) were identified in cashew apple juice. Cutting and subsequent storage at 40 °C hydrolysed cinnamic acid. 5-Hydroxymethylfurfural content in cashew apple juice increased after injury and storage at higher temperatures, indicating non-enzymatic browning.  相似文献

Lactobacillus casei Zhang is a novel strain that was screened out of koumiss collected in Inner Mongolia, and our previous research showed that L. casei Zhang has health benefits such as cholesterol-reducing and immunomodulating effects. The fermentation characteristics of L. casei Zhang in soymilk and bovine milk and the transit tolerance of L. casei Zhang in fermented milk products during refrigerated storage for 28 d were assessed. A faster decrease in pH and faster growth of L. casei Zhang during fermentation were observed in soymilk compared with bovine milk at various inoculation rates, probably because of the low pH buffering capacity of soymilk. The fermented bovine milk samples had much higher final titratable acidity (TA) values (between 0.80 and 0.93%) than the soymilk samples (between 0.40 and 0.46%). Dramatic increases in TA values in the fermented soymilk samples during storage were observed, and the TA values of the fermented soymilk samples changed from <0.56% to values between 0.86 and 0.98%. On the other hand, only slight increases in TA were observed in the bovine milk samples during the 28 d of storage. The survival rates of freshly prepared cultures of L. casei Zhang in simulated gastric juice at pH 2.0 and 2.5 were 31 and 69%, respectively, and the delivery of L. casei Zhang through fermented soymilk and bovine milk significantly improved the viability of L. casei Zhang in simulated gastric transit. Lactobacillus casei Zhang showed good tolerance to simulated gastric juice and intestinal juice in the fermented soymilk and bovine milk samples, and maintained high viability (>108 cfu/g) during storage at 4°C for 28 d. Our results indicated that both soymilk and bovine milk could serve as vehicles for delivery of probiotic L. casei Zhang, and further research is needed to elucidate the mechanism of the change in pH and TA of L. casei Zhang in fermented milk samples during fermentation and storage and to understand the difference between soy- and milk-based systems.  相似文献

The biocatalyst prepared by immobilization of Saccharomyces cerevisiae strain AXAZ-1 on apple pieces was used for fermentation of grape must both in batch and continuous system. The immobilized yeast showed an important operational stability without any decrease of its activity. In continuous process, the bioreactor was operated for 95 days without any diminution of the ethanol productivity. Concentrations of higher and amyl alcohols were low indicating an improved quality product.Apple pieces were also used as support for immobilizing Kluyveromyces marxianus IMB3 for high-temperature (45 °C) wine-making. The fermented grape must contained 3-4% alcohol and semi-sweet wines were produced by the addition of potable alcohol.Preliminary sensory evaluation of the produced wines ascertained the fruity aroma and the overall improved quality compared to other commercially available products.Lactobaccillus casei cells immobilized on apple pieces were used for probiotic, fermented milk and lactic acid production. Apple-supported L. casei used for successive fermentation batches of whey proved to be very effective and suitable for food-grade lactic acid production. The immobilized biocatalyst was also used for milk fermentation and was able to ferment after storage for 15, 98 and 129 days at 4 °C, while no infection was reported during storage periods.  相似文献

Two types of synbiotic ice cream containing 1% of resistant starch with free and encapsulated Lactobacillus casei (Lc-01) and Bifidobacterium lactis (Bb-12) were manufactured. The survival of L. casei and B. lactis were monitored during the product’s storage for 180 days at −20 °C. The viable cell number of L. casei and B. lactis in the free state in prepared ice cream mixture was 5.1 × 109 and 4.1 × 109 CFU/mL at day one and after 180 days storage at −20 °C, these numbers were decreased to 4.2 × 106 and 1.1 × 107 CFU/mL, respectively. When we encapsulated the mentioned probiotic bacteria in calcium alginate beads, the probiotic survival raised at rate of 30% during the same period of storage at same temperature. In general, the results indicated that encapsulation can significantly increase the survival rate of probiotic bacteria in ice cream over an extended shelf-life. The addition of encapsulated probiotics had no significant effect on the sensory properties of non-fermented ice cream in which we used the resistant starch as prebiotic compound.  相似文献

The aim of the present work was to investigate the use of cashew apple juice as a low cost substrate for Lactobacillus casei B-442 cultivation and lactic acid production. Ammonium sulfate was employed as the only exogenous nitrogen source. The effect of cashew apple juice reducing sugars and ammonium sulfate concentration and the fermentation pH and temperature on biomass formation, lactic acid production, and productivity were evaluated. The highest productivity (2.36 g/L.h) was obtained applying 50 g/L of reducing sugar from the cashew apple juice supplemented with 6 g/L of ammonium sulfate. The process yield was about 95% when fermentation was carried out at 37 °C with pH controlled at pH 6.5 using NaOH (120 g/L).  相似文献

This research investigated the efficacy of gaseous ozone on the inactivation of Escherichia coli ATCC 25922 and NCTC 12900 strains in apple juice of a range of pH levels, using an ozone bubble column. The pH levels investigated were 3.0, 3.5, 4.0, 4.5 and 5.0. Apple juice inoculated with E. coli strains (106 CFU/mL) was treated with ozone gas at a flow rate of 0.12 L/min and ozone concentration of 0.048 mg/min/mL for up to 18 min. Results show that inactivation kinetics of E. coli by ozone were affected by pH of the juice. The ozone treatment duration required for achieving a 5-log reduction was faster (4 min) at the lowest pH than at the highest pH (18 min) studied. The relationship between time required to achieve 5 log reduction (t5d) and pH for both strains was described mathematically by two exponential equations. Ozone treatment appears to be an effective process for reducing bacteria in apple juice and the required applied treatment for producing a safe apple juice is dependant on its acidity level.  相似文献

The influence of organic acid and heating treatments on carotenoid degradation on a simulated cashew apple juice was assessed by high performance liquid chromatography coupled with a photodiode array and mass spectrometry detectors. A total of nineteen carotenoids were separated in unheated simulated cashew apple juice, with all-trans-β-cryptoxanthin and all-trans-β-carotene as the major ones. As a consequence of heating, five xanthophylls disappeared, whereas two new cis isomers and five epoxide or furanoid-derivatives were formed and the levels of all cis isomers increased. In addition, 12′-apo-β-carotenal was formed at 90 °C. Two oxidation compounds (12′-apo-β-carotenal and 5,6-epoxy-β-cryptoxanthin) were formed after β-cryptoxanthin heating at 90 °C in an aqueous-based system. In all systems, the amounts of total carotenoids lost were not compensated by those formed. These facts indicated that isomerisation and oxidation to both coloured and non-coloured compounds were the main reactions occurring during heating of carotenoids in aqueous-based and juice systems.  相似文献

This study was conducted to evaluate probiotic potato juice as a potential substrate for the production of Lactobacillus casei, and the change in the functionality of potato juice was monitored during fermentation. L. casei grew well in potato juice without nutrient supplementation, and lactic acid bacteria of fermented ‘Haryoung’ juice reached 1.7×109 CFU/mL after a 48 h fermentation. DPPH radical scavenging activities of the potato juices decreased after a 72 h fermentation, but fermented colored potato juice still maintained >50% radical scavenging activity. The survival rate of L. casei fermented in’ Haryoung’ juice was 89.0% after exposure to an acidic condition, and L. casei in all fermented potato juice samples showed the ability (50–85%) to survive in the presence of bile. These results suggest that fermented potato juice might serve as a probiotic functional beverage for vegetarians or consumers who are allergic to dairy products.  相似文献

The use of sonicated melon juice as substrate for Lactobacillus casei growth in cantaloupe melon juice and its effect on product quality were investigated. The survival of L. casei in the fermented juice, the product color, the post-acidification and other quality parameters were evaluated in the refrigerated product (4 °C) for a period of 42 days. Storage pH, color, sugar content, lactic acid concentration, biomass and viable cells count were determined. The caloric value of the product was also evaluated. During the refrigerated storage period, acidification and a slight reduction in the number of viable cells were observed. However, a sufficient number of viable cells were observed to guarantee the beneficial properties from probiotic consumption (>8 log CFU?ml?1) throughout the storage period, for both stocked samples (with and without sugar addition). The product color was also preserved during storage. The developed product resulted in a low caloric product. The caloric value was reduced during the storage period due to sugar consumption by the remaining microorganisms.  相似文献

The objective of this study was to evaluate the efficacy of supercritical carbon dioxide (SCCO2) for inactivating Lactobacillus plantarum in apple cider using a continuous system with a gas-liquid metal contactor. Pasteurized apple cider without preservatives was inoculated with L. plantarum and processed using a SCCO2 system at a CO2 concentration range of 0-12% (g CO2/100 g product), outlet temperatures of 34, 38, and 42 °C, a system pressure of 7.6 MPa, and a flow rate of 1 L/min. Processing with SCCO2 significantly (P < 0.05) enhanced inactivation of L. plantarum in apple cider, resulting in a 5 log reduction with 8% CO2 at 42 °C. The response surface model indicated that both CO2 concentration and temperature contributed to the microbial inactivation. The extent of sublethal injury in surviving cells in processed apple cider increased as CO2 concentration and processing temperature increased, however the percent injury dramatically decreased during SCCO2 processing at 42 °C. Structural damage in cell membranes after SCCO2 processing was observed by SEM. Refrigeration (4 °C) after SCCO2 processing effectively inhibited the re-growth of surviving L. plantarum during storage for 28 days. Thus this study suggests that SCCO2 processing is effective in eliminating L. plantarum and could be applicable for nonthermal pasteurization of apple cider.  相似文献

Enterocin AS-48 was tested in apple juice against the cider-spoilage, exopolysaccharide-producing strain Lactobacillus diolivorans 29 in combination with high-intensity pulsed-electric field (HIPEF) treatment (35 kV/cm, 150 Hz, 4 μs and bipolar mode). A response surface methodology was applied to study the bactericidal effects of the combined treatment, with AS-48 concentration and HIPEF treatment time as process variables. At subinhibitory bacteriocin concentrations, microbial inactivation by the combined treatment increased as the bacteriocin concentration and the HIPEF treatment time increased (from 0.5 to 2.0 μg/ml and from 100 to 1000 μs, respectively). Highest inactivation (4.87 logs) was achieved by 1000 μs HIPEF treatment in combination with 2.0 μg/ml AS-48. While application of treatments separately did not protect juice from survivors during storage, survivors to the combined treatment were inactivated within the following 24 h of storage, and the treated samples remained free from detectable lactobacilli for at least 15 days at temperatures of 4 °C as well as 22 °C. The combined treatment could be useful for inactivation of exopolysaccharide-producing L. diolivorans in apple juice.  相似文献

Heat resistance of Neosartorya fischeri was studied in three different juices (apple, pineapple and papaya). The optimum heat activation temperature and time for the ascospores of the N. fischeri (growth for 30 days at 30 °C) was 85 °C for 10 min. Of the three juices tested, apple juice exhibited maximum 1/k values at 80, 85 and 90 °C (208.3, 30.1 and 2.0 min, respectively). The 1/k values for papaya juice (129.9, 19.0 and 1.9 min) and pineapple juice (73.5, 13.2 and 1.5 min) decreased with acidity and °Brix/acidity (ratio) level. The Z* values for apple, papaya and pineapple juices were 5, 5.5 and 5.9 °C, respectively. The sterilization F values (4-log reduction) for apple, pineapple and papaya juices were 56.3, 38.0 and 7.2 s, respectively. Considering the thermal treatments commercially applied to pineapple (96 °C/30 s) and apple juices (95 °C/30 s), it is concluded that such treatments will not guarantee that less than 1 ascospore in each set of 103 packs survive. Only the treatment applied to papaya juice (100 °C/30 s) will be sufficient because the F value is less than 30 s.  相似文献

The effects of selenium enriched green tea (SGT; 85.8–96 mg/kg) in different concentrations of 1 g and 2 g/100 mL, on the in vitro exposure to simulated gastrointestinal juice and refrigerated storage of encapsulated Lactobacillus casei and Lactobacillus plantarum were investigated in chitosan coated alginate beads. The encapsulation yield of viable cells in chitosan coated alginate beads with and without SGT was not significantly different (P < 0.05). These results together with the study about the survival of probiotic bacteria in microspheres with SGT during storage at 4 °C, demonstrated significantly higher number (P < 0.05) of survival bacteria in microcapsules with SGT 2 g/100 mL. Microencapsulated L. casei and L. plantarum with SGT 1 g and 2 g/100 mL were resistant to simulated gastric conditions (pH 2.0, 2 h) and bile solution (3 g/100 mL, 2 h) resulting in significantly (P < 0.05) improved survival when compared with microencapsulation without SGT addition.  相似文献

The study compared the growth capability of probiotic (Lactobacillus acidophilus La05, Lactobacillus casei Lc01 and Bifidobacterium animalis Bb12) and non-probiotic (Lactobacillus delbrueckii subsp bulgaricus and Streptococcus thermophilus) cultures on twenty-one culture media grouped according to selectivity: non-selective agars, selective agars without antibiotics and MRS agars containing different combinations of lithium chloride, cystein, bile salts and antibiotics. Four of these media were selected for quantitative enumeration of L. acidophilus La05, L. casei Lc01, and B. animalis Bb12. The best culture media and incubation conditions for enumeration of the probiotic cultures were: B. animalis: MRS agar with dicloxacillin, 37 °C or 42 °C, anaerobiosis; L. acidophilus: MRS agar with bile salts, 37 °C or 42 °C, aerobiosis; L. casei: MRS agar with lithium chloride and sodium propionate, 37 °C or 42 °C, aerobiosis or anaerobiosis. Plating on MRS with glucose replaced by maltose, 37 °C or 42 °C, anaerobiosis, will distinguish probiotic from non-probiotic cultures. For enumeration of each probiotic in a mixed culture, the following media and incubation conditions were recommended: B. animalis: 4ABC-MRS, 42 °C, anaerobiosis, L. acidophilus: LC medium, 42 °C, aerobiosis or anaerobiosis and L. casei: LP-MRS, 42 °C, aerobiosis or anaerobiosis. In all experiments, differences in counts using pour plating or surface plating were not significant (P ≤ 0.05).  相似文献

ABSTRACT:  High hydrostatic pressure is an alternative to thermal processing to inactivate spoilage and pathogenic microorganisms. Cashew apple juice has a pleasant flavor and is rich in vitamin C. Studies to determine the effect of high pressure on microorganisms in cashew apple juice are still lacking. In this study, the inactivation of natural micropopulation and inoculated Escherichia coli by high pressure was evaluated in fresh cashew apple juice. The microbiological stability of pressure-treated juice was also evaluated. The applied high pressure levels ranged from 250 to 400 MPa for periods of 3 to 7 min. Treatments with 350 MPa for 7 min and 400 MPa for either 3 or 7 min reduced the aerobic mesophilic bacteria count to a level below the detection limit. Pressure treatments were also efficient in inactivating yeast and filamentous fungi. The inoculated E. coli (106 CFU/mL) was reduced to below 10 CFU/mL after a pressure treatment of 400 MPa for 3 min. The inactivation of this microorganism followed a 1st-order reaction kinetics. The decimal reduction time ( D- value) ranged from 1.21 to 16.43 min, while pressure resistance value ( z -value) was 123.46 MPa. Neither natural micropopulation growth nor E. coli repair was observed in postprocessed (400 MPa for 3 min) cashew apple juice kept under refrigerated storage (at 4 °C) during 8 wk. The results of this study demonstrated the efficacy of high-pressure treatment for preserving cashew apple juice.  相似文献

The production of lactic acid by Lactobacillus casei B-442 was studied and modeled. Sugar feedstock was provided using cashew apple juice, an alternative glucose and fructose feedstock that proved to yield high concentrations of lactic acid. The fermentations were carried out in a 1-L fermenter under constant agitation (150 rpm) and controlled pH (6.5). Lactic acid production was evaluated through a dynamic study, varying the initial concentration of sugar in the range of 20 to 60 g/L. Biomass, reducing sugars, and lactic acid concentration were measured throughout the experiments. The highest production of lactic acid (59.3 g/L) was obtained operating the fermentation with 60 g/L of reducing sugar from the cashew apple juice. A rigorous kinetic model was developed for batch fermentation of cashew apple juice for lactic acid production by L. casei B-442. The growth of biomass and lactic acid production were affected by substrate limitation, substrate inhibition and lactic acid inhibition. The model assumed growth- and non-growth-associated lactic acid production and a term for microorganism death was also included in the model. Parameters of the kinetic model were determined based on experimental data by using the least mean squares method and Levenberg–Marquardt algorithm. The model validation was performed and the model was statistically able to fit the profiles for growth of biomass, sugar consumption and lactic acid production. The optimization of the process, using the model, was carried out and the optimum operating conditions aiming highest productivity, lowest production cost and highest gross profit are presented.  相似文献

In this study, ascorbic acid, total polyphenols and proanthocyanidins of fresh-cut cashew apple were quantified. Antioxidant capacity was determined in whole juice and in polyphenols extracts by three methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) and ??-carotene bleaching. Effect of cutting and storage for 24 h at 2 °C, 27 °C and 40 °C on these compounds were also evaluated. Cashew apple presented 163 mg of ascorbic acid per 100 g of fresh weight (FW). Soluble and hydrolysable polyphenols contents were 12.79 mg GAE/100 g FW and 18.53 mg GAE/100 g FW and proanthocyanidins were 9.27 mg/100 g FW. Antioxidant capacity of juice and polyphenols extract was high for DPPH method. Storage temperatures affected bioactive compounds on cut cashew apple. The content of ascorbic acid decreased in all temperatures. Proanthocyanidins were more sensitive to 40 °C than to other temperatures. The content of polyphenols and antioxidant capacity of juice by DPPH assay did not change. However, the reducing power was lower in samples kept at high temperatures. A strong positive correlation between ascorbic acid and FRAP (r = 0.99) and a negative correlation between DPPH and FRAP (r = − 0.79) were observed. No correlations were found between polyphenols and antioxidant capacity indicating the importance of phenolic composition in the extracts. The results confirm the importance of temperature and injury on the quality of cashew apple.  相似文献
