Nowadays, studies about the anti‐obesity potential of probiotics are of growing interest. Lactobacilli are one of the well‐studied probiotics owing to their preventing effect on metabolic disorders. This study was undertaken to access the anti‐obesity effect of probiotic dahi containing Lactobacillus casei NCDC 19 on C57BL/6 mice. Feeding of probiotic dahi showed reduce body weight gain and epididymal fat weights. Moreover, blood glucose, plasma lipids and expression level of leptin were reduced and caecal bifidobacteria counts and adiponectin expression levels were significantly increased. It can be concluded that feeding of probiotic dahi containing L. casei NCDC 19 showed potential anti‐obesity effects.  相似文献

The objective of this study was to increase quality and limited shelf‐life of boza (3–15 days), a traditional Balkan origin fermented beverage using lysozyme (LYS) and/or nisin (NIS). For this purpose, the effectiveness of NIS, LYS and LYS:NIS combinations was first tested in a broth medium at 4 °C for 3 weeks on Lactobacillus plantarum, one of the frequently isolated lactic acid bacteria in boza. Stability of LYS and NIS in boza, their effects on LAB counts, and chemical and sensory properties of boza were then evaluated during cold storage at 4 °C. Results of LAB counts as well as pH, d ‐ and l ‐lactic acid, and titratable acidity measurements showed that LAB in boza containing NIS (250 μg g?1) or LYS:NIS (500:250 μg g?1) could be controlled without reducing LAB counts below 6 log CFU mL?1 during 2 weeks shelf‐life. In contrast, LYS (500 μg g?1) alone could not control LAB in boza to delay its acidic spoilage. Positive effects of NIS and LYS:NIS application on quality of boza were also proved with sensory analysis by panelists and e‐nose measurements. This work showed that use of natural GRAS agents in preservation of fermented beverages containing probiotic LAB is possible without affecting their characteristic aroma and flavour.  相似文献

The effect of inoculum level of Bifidobacterium animalis ssp. lactis BB‐12 probiotic strain and ripening period on the quality of dry‐cured neck was studied. The microbiological parameters (Enterobacteriaceae, LAB and TVC) and physicochemical attributes (pH value, aw, TBARS index, colour) were determined directly after fermentation at 15 °C for 3 weeks, after 6 and 12 months of ripening at 4 °C. The highest LAB count and a lower pH value were found in the meat inoculated with probiotic strain at 6.6 log cfu g?1 (B2) followed by inoculation with probiotic strain at 6.3 log cfu g?1 (B1). Level of inoculation had not had an influence on water activity, TBARS index and total colour parameters. Changes of fat oxidation during half‐year of ripening were limited in probiotic meat samples compared to naturally fermented control meat (C). Based on the results, it can be concluded that the most favourable physicochemical and microbiological parameters of the dry‐cured neck were obtained after 6 months of ripening. At that time, the Bifidobacterium BB‐12 at both levels is a good potential starter for meat fermentation.  相似文献

This study aimed to evaluate the vacuum impregnation (VI) and soaking methods in the addition of Lactobacillus acidophilusLA‐3 to minimally processed melon (MPM). The melons were washed, sanitised in chlorine solution (200 mg L?1), peeled and cutted into cubes. Lactobacillus acidophilusLA‐3 (1.4 × 1010 CFU g?1) were added to the MPM through both techniques. The L. acidophilusLA‐3 count in MPM was similar to those commonly found in dairy products having probiotic claim, but VI was more efficient than soaking in maintaining the viability (8.61 and 7.98 Log CFU g?1, respectively). The pH, acidity and soluble solids were not affected by probiotic culture and the incorporation technique; however, the VI affected the firmness of fruit. The MPM was within Brazilian standards for their microbiological characteristics. MPM may be used as a carrier of probiotic bacteria, being one more alternative for individuals who consume probiotic products.  相似文献

Myricetin is a natural flavonol widely occurring in wines. Many beneficial effects of myricetin in alcoholic beverages have been reported before, but never including anti‐obesity. In the present study, we fed obese male Sprague–Dawley rats with ethanol solutions containing various concentrations of myricetin and found that myricetin may maintain the food intake while reduce the weight‐gain, feed efficiency, level of blood lipids, adipocyte size, and weight and size of the perirenal and epididymal adipose tissues (P < 0.01). Our experiment data also show that the anti‐obesity effect may be associated with the upregulation of adropin and β‐endorphin levels. Based on the above‐described findings, we propose the potential for myricetin‐containing alcoholic beverages to be developed into anti‐obesity health food.  相似文献

Scope

Modifications in intestinal microbiota and its metabolites, the short-chain fatty acids (SCFA) are main factors altering intestinal epithelial barrier integrity and eliciting the onset of a meta-inflammation observed in obesity. The present study is aimed at evaluating the efficacy of Enterococcus faecium (SF68) administration in counteracting the impairment of gut barrier and enteric inflammation in a model of diet-induced obesity, characterizing the molecular mechanisms underlying such beneficial effects.

Methods and Results

Male C57BL/6J mice, fed with standard diet (SD) or high-fat diet (HFD), are treated with SF68 (108 CFU day−1). After 8 weeks, plasma interleukin (IL)-1β and lipopolysaccharide binding protein (LBP) are measured, analysis of fecal microbiota composition and butyrate content as well as intestinal malondialdehyde, myeloperoxidase, mucins, tight junction protein, and butyrate transporter expression are investigated. After 8 weeks, SF68 administration counteracts the body weight gain in HFD mice, reducing plasma IL-1β and LBP. In parallel, SF68 treatment acts against the intestinal inflammation in HFD-fed animals and improves the intestinal barrier integrity and functionality in obese mice via the increase in tight junction protein and intestinal butyrate transporter (sodium-coupled monocarboxylate transporter 1 ) expression.

Conclusions

Supplementation with SF68 reduces intestinal inflammation and reinforces the enteric epithelial barrier in obese mice, improving the transport and utilization of butyrate.  相似文献

This research studied the use of banana purée from three indigenous Thai cultivars (Kluai Hom Thong, Kluai Namwa and Kluai Khai) as probiotic carrying agents in food and application in bio‐set‐type yoghurt. Kluai Khai purée had promising indication of prebiotic activity on Lactobacillus paracasei Lpc‐37 with the highest probiotic activity score. When Lpc‐37 cell was incorporated into Kluai Khai purée, its cell counts increased by 0.8 log CFU g?1 and it adhered to purée, as seen by the proliferating cells. Therefore, Kluai Khai purée mixed with Lpc‐37 was selected for the production of bio‐set‐type yoghurt. The results showed that viable cells increased more than 0.8 log CFU g?1 during storage. This study reflected the achievement of using Kluai Khai purée as food matrix for carrying ‘active probiotic cells’ in bio‐yoghurt production. This methodologically developed prototype could be an interesting alternative for value adding of indigenous fruits.  相似文献

Relationship between intestinal flora and obesity has aroused great interest. The probiotic effects of Lactobacillus plantarum (LP) and Lactobacillus fermentum (LF) have been documented extensively. This study aimed to explore the effects of LP and LF alone or in combination on lipid‐lowering effects of the high‐fat diet rat via intestinal flora modulation and systemic immunity. The rats in high‐fat diet plus LP (LP 108 cfu day?1) showed significantly lower IL‐6 and endotoxin (ET) content, increased the number of Bifidobacterium and Lactobacillus and decreased the liver steatosis and fat vehicle sizes. The Lactobacillus LP and LF in combination can regulate intestinal flora and systemic immune function in rats with high‐fat diet. It provides the foundation for the further development and application of multiple Lactobacillus strain in controlling obesity‐related syndrome.  相似文献

The probiotic Lactobacillus acidophilus was encapsulated in biodegradable and biocompatible capsules prepared by ionic gelation between phytic acid (PA) and chitosan (CS) with an electrostatic extrusion method. Calcium carbonate (CaCO3) and starch were used as co‐encapsulants for improvement of capsule stability. Capsules were characterised and evaluated for survival of encapsulated L. acidophilus cells in simulated gastric fluid (SGF) and during refrigerated storage. Loading capacity values of PA‐CS capsules, PA‐CS‐starch capsules and PA‐CS‐CaCO3 capsules were 8.20, 8.12 and 7.81 log CFU g?1 of wet capsule, respectively. Capsules showed particle sizes of 1.3–1.5 mm and a uniform spherical shape. PA‐CS‐CaCO3 capsules were the most stable vehicles for the protection of probiotic cells against acidic damage, particularly at pH 1.5 and pH 2. L. acidophilus cells from PA‐CS‐CaCO3 capsules showed only a 0.64 log CFU reduction in numbers after 2 h in pH 1.5 SGF conditions. The numbers of L. acidophilus encapsulated in PA‐CS‐CaCO3 capsules were decreased by only 0.69 log CFU g?1, while PA‐CS capsules and PA‐CS‐starch capsule numbers were reduced by more than 1.45 log CFU g?1 after 4 weeks at 4 °C. Addition of calcium carbonate to PA‐CS capsules provided protection against acid injury via antacid and buffering effects for encapsulation of L. acidophilus.  相似文献

Gluten‐free (GF) sourdough was prepared from wheat sourdough and analysed both in fresh (GFS) and dried forms (DGFS). The gluten content in each GF sourdough sample was <20 mg kg?1. The dough leavening capacity and the properties of the bread samples were investigated and compared to those of bread prepared using bakery yeast (Saccharomyces cerevisiae). Two commercial rice‐based mixtures (different for the presence/absence of buckwheat flour) were used to prepare bread samples. In GFS, lactic acid bacteria (LAB) and yeasts were found in amounts corresponding to 108 and 107 CFU g?1, respectively, whereas both LAB and yeasts were detected in lower amounts (about 106 CFU g?1) in DGFS. When used in bread‐making, both GFS types produced significant dough acidification and exhibited good dough development during proofing, resulting in loaves with specific volume values between 3.00 and 4.12 mL g?1, values similar to those obtained for reference bread (3.05÷4.15 mL g?1). The use of GFS was effective in lowering the bread staling rate during storage for up to 7 days.  相似文献

Three medium‐ and long‐chain triacylglycerols (MLCT) with different contents of medium‐chain fatty acids (MCFA) (10% to 30%, w/w) were prepared and evaluated for their anti‐obesity potential in C57BL/6J mice. The group fed with a high fat diet of MLCT containing 30% (w/w) MCFA showed significantly decreased body weight and fat mass (P < 0.05) relative to the control mice fed an obesity‐inducing high fat rapeseed oil diet. In addition, serum parameters including triacylglycerols, total cholesterol, glucose, high‐density lipoprotein cholesterol, low‐density lipoprotein cholesterol, apolipoprotein A1 and apolipoprotein B in the treatment group fed with 30% (w/w) MCFA were close to those of mice fed with a low fat rapeseed oil diet, but significantly different (P < 0.05) from those of the obesity control group. Moreover, the intake of MLCT with high content of MCFA reduced the size of adipocytes. In addition, the visceral fat and liver weights, as well as the liver triacylglycerol for 3 treatment groups were lower than those of the obesity control group. These results demonstrate the great potential of MLCT with high content of MCFA in weight loss.  相似文献

Given the relatively immature state of the neonatal gut and gut-associated immune system, the safety of probiotic strains for use as ingredients in infant milk formulae must be demonstrated in infant populations. As part of a double-blind placebo-controlled clinical trial of two commercially available probiotic strains in the reduction of risk for infant eczema, a number of safety outcomes were measured. Infants received daily doses of Lactobacillus rhamnosus HN001 (6 × 109 cfu day?1) or Bifidobacterium animalis subsp. lactis HN019 (9 × 109 cfu day?1), or placebo from birth to 24 months. Mothers received the same treatment from 35 weeks gestation, for up to 6 months postnatally while breastfeeding. No statistically significant differences were observed between the treatment groups for study withdrawal, incidence of adverse events, morphometric data, wheeze, and antibiotic use over the treatment period. We conclude that probiotics strains HN001 and HN019 were safe and well tolerated in infants, and did not affect normal growth.  相似文献

BACKGROUND: Stevia (Stevia rebaudiana Bertoni) is a non‐caloric natural‐source alternative to artificially produced sugar substitutes. This study investigated the effect of stevia extract on lipid profiles in C57BL/6J mice. Forty mice were divided into four groups: N‐C (normal diet and distilled water), H‐C (high‐fat diet and distilled water), H‐SC (high fat diet and sucrose, 1 mL kg?1 per day), and H‐SV (high‐fat diet and stevia extract, 1 mL kg?1 per day). RESULTS: Body weight gain was significantly higher in the H‐SC group than in the H‐SV group. Triglyceride concentrations in serum and liver were lower in the H‐SV group than in the H‐SC group. Serum total cholesterol concentrations were lower in the H‐SV and H‐C groups compared to the H‐SC group. The concentrations of acid‐insoluble acylcarnitine (AIAC) in serum were higher in the H‐SV group than in the H‐C and H‐SC groups and the acyl/free carnitine level in liver was significantly higher in the H‐SV group than in the N‐C group. These results were supported by mRNA expression of enzymes related to lipid metabolism (ACO, PPARα, ACS, CPT‐I, ACC) assessed by real‐time polymerase chain reaction. CONCLUSION: These results suggest that the supplementation of stevia extract might have an anti‐obesity effect on high‐fat diet induced obese mice. BACKGROUND: The effects of a high protein diet on insulin secretion and glucose metabolism have been quite controversial. The aim of this study was to evaluate the effects of long‐term isocaloric high animal protein intake on insulin secretion in diet‐induced obese rats. RESULTS: After the experimental period (24 weeks), the high‐fat diet‐induced obese rats that were fed isocaloric high‐protein diets (HP) had lower body weight gain (P < 0.01) and lower visceral fat (P < 0.05) than normal protein (NP) rats. Fasting plasma glucagon‐like peptide‐1 (GLP‐1) was also reduced significantly (P < 0.05), as well as serum insulin levels at 5 min and 10 min by intravenous insulin releasing test. In addition, insulin mRNA and pancreatic duodenal homeodomain‐1 (PDX‐1), GLP‐1 protein expression were both markedly lower in HP rats (P < 0.05), while PDX‐1 mRNA in HP rats had no difference from NP rats. CONCLUSION: These results suggest that long‐term isocaloric high animal protein intake reduces the acute insulin response in obese rats and the decrease of insulin is associated with both reduced weight gain and inhibition of PDX‐1 expression. Effects of different levels of fat and inulin on bacterial cell counts, degree of proteolysis and concentrations of organic acids in the yogurt containing inactivated cells of probiotic strains Bifidobacterium animalis and Lactobacillus acidophilus were investigated. Results showed that both L. acidophilus and B. animalis grew well in the yogurt samples reaching cell counts higher than 106 CFU mL?1 at the final pH of 4.5. Inulin at the concentration of 1% had no significant effects on the production of organic acids and cell counts of L. acidophilus, but promoted the growth of B. animalis with a reduction in the degree of proteolysis. Generally, different fat levels showed significant effects on the production of organic acids and nonsignificant effects on the cell counts of probiotic bacteria and degree of proteolysis. In case of lactic acid, the ratio of L‐ (+)to D‐ (?) isomer ranged from 50/50 to 80/20 in yogurt samples.  相似文献

The human health implications of exposure to environmental pollutants and food components (e.g. phytoestrogens) that mimic the female hormone 17β‐estradiol (xenoestrogens) are a subject of significant debate. In this study, we use published data on human dietary phytoestrogen intake and sperm concentrations in the USA and China to assess effects on sperm concentration as a marker of male reproductive health. Comparison of the phytoestrogen intake and sperm concentration trends shows that in the USA phytoestrogen intake has increased from 0.8 mg day?1 in 1993 to 13.7 mg day?1 in 2005 and sperm concentration is trending down (not statistically significant), whereas in China phytoestrogen intake has decreased from 64.7 mg day?1 in 1991 to 15.6 mg day?1 in 2008, and the sperm concentration has increased from 55 × 106 mL?1 in 1999 to 74 × 106 mL?1 in 2008. We present this as evidence that phytoestrogen intake via food might, at least in part, be responsible for sperm concentration trends.  相似文献

The objective of this study was to develop a casein-based edible film for the entrapment of probiotic Enterococcus faecium Rp1. Casein, pectin, sodium alginate and glycerol were used to prepare the film. In this study, the physicochemical and morphological properties of casein-based edible film and its impact on the stability of probiotic were evaluated. Surface morphology and properties of the film were tested using a scanning electron microscope, fluorescence microscopy, Raman spectroscopy, Fourier transform infrared spectroscopy and X-ray diffraction. Probiotic-incorporated casein-based edible film showed significant improvement in the antimicrobial and antioxidant properties and enhanced the structural, optical and thermal properties. Furthermore, the film was found to be desirable to carry probiotics, with the viability of 107 CFU mL−1 rate up to 30 days of storage at 4 °C. Hence, the current study suggests a probiotic-incorporated casein-based edible film for active packaging of food products.  相似文献

Antimicrobial activities of high molecular weight water‐soluble chitosans (HMWWS) against selected Gram‐negative and Gram‐positive foodborne pathogens (initial inoculation of ca. 6.5 Log CFU mL?1) were evaluated. Chitosans with 789 kDa and/or 1017 kDa were dissolved in aspartic acid (AS) to obtain 1–4% w/v solutions. Among HMWWS, only 4% 789 kDa AS chitosan reduced E. coli counts by 2 Log CFU mL?1 from 7.33 at 0 h to 5.16 Log CFU mL?1 at 96 h, and they were not effective against S. Typhimurium. Depending on the concentrations, HMWWS completely inhibited V. cholerae, V. vulnificus and Vparahaemolyticus as well as B. cereus and L. monocytogenes after 48 h or 96 h of incubation. Compared with the control (no HMWWS), 2% or 3% 1017 kDa AS chitosans showed about 3 Log CFU mL?1 lower (4.72–4.86 vs. 7.71) for S. aureus at 96 h of incubation.  相似文献

Three GRAS antimicrobials including, lauric arginate (LAE), bacteriophage P100 (phage P100) and bacteriocin nisin, were evaluated either singly or in combinations for the reduction of initial load of Listeria monocytogenes in cold‐smoked salmon (CSS). The stability of phage P100 in the presence of LAE (200 ppm) and nisin (500 ppm) or at 10× and 100× of these concentrations was determined at 4 °C or 30 °C for 24 h in a broth model. Phage P100 was found to be highly stable in the presence of these antimicrobial agents as plaque‐forming units (PFU) did not vary between control and antimicrobial‐treated phage. The survival of L. monocytogenes in the presence of phage P100, nisin and LAE showed remarkable reduction within 24 h both at 4 °C or 30 °C in broth. Treatment of CSS containing 3.5 log CFU cm?2 L. monocytogenes with phage P100 (10PFU mL?1), nisin (500 ppm) and LAE (200 ppm) showed strong listericidal action and reduced the L. monocytogenes by 2–3 log CFU cm?2 after 24 h. Among the combined treatments, phage P100 + LAE or nisin + LAE exhibited the most listericidal action in which L. monocytogenes cells were reduced to undetectable level within 24 h in CSS.  相似文献

Lactic acid bacteria (LAB) include species very closely related both physiologically and genotypically. Therefore, the identification of this bacteria group using conventional phenotypic methods is ambiguous and cumbersome. In this study, we have analyzed a recA gene fragment from 30 bacteria, including LAB and species common in the human gastrointestinal tract, aiming to evaluate the gene conservation among them and the development of primers and PCR conditions able to discriminate Lactobacillus plantarum strains from LAB closely related. The fragment with 995 bp of recA gene has grouped LAB, enterobacteria and bifidobacteria, in different clusters. A novel primer pair, LPrecAF and LPrecAR with 23 and 18 bp, respectively, has allowed the single amplification of a 108 bp fragment of L. plantarum strains contained in culture broth and fermented dairy samples. The observed detection limit for food samples and for cultures broth were 1 × 103 and 7 × 102 CFU mL−1, respectively. This approach proved to be a simple and efficient method for the identification and monitoring of L. plantarum in food, feeds, and culture broth. Moreover, the assay could be used in the studies from human or environmental microbiota.  相似文献
