The acquisition of amino acids by Lactobacillus delbrueckii subsp. bulgaricus 2038 (Lb. bulgaricus 2038) when grown in the presence of bovine casein, the major protein in bovine milk, was investigated by examining the expression of genes related to proteolysis and amino acid biosynthesis. To support the growth on bovine casein, Lb. bulgaricus 2038 has to synthesise five kinds of amino acids de novo, as proteolysis from casein does not provide these. The incomplete hydrolysis in combination with amino acids biosynthesis may explain the slow growth of Lb. bulgaricus 2038 in a casein environment. Meanwhile, it was determined that Lb. bulgaricus 2038 uses different intracellular peptidases when grown in casein or whey medium, and initially yields the important amino acid glutamate from the C-terminal or N-terminal end of peptides imported into the cell.  相似文献

A comparative study of the properties of membrane‐bound polyphenol oxidase (mPPO) from three apple cultivars, namely Red Fuji (FJ), Granny Smith (GS) and Golden Delicious (GD), was carried out for the first time. Data indicate that mPPOs from three cultivars exhibit significantly different properties. GS mPPO had the strongest affinity to catechol, but FJ mPPO had the highest maximum velocity. Red Fuji (FJ) mPPO had the significantly higher activity than those of GD and GS mPPOs. Red Fuji (FJ) mPPO had the highest activity at pH 8.00, while GD and GS mPPOs at 4.50 and 7.50–8.00, respectively. Red Fuji (FJ) mPPO was more stable than GD and GS mPPOs over the pH range of 5.0–8.5. The optimal temperature for GS mPPO was within 70–75 °C, which is higher than those for mPPOs from FJ and GD. Thermal inactivation of the three mPPOs followed a first‐order kinetic model with different inactivation kinetic parameters.  相似文献

Abstract: Methicillin‐resistant Staphylococcus aureus (MRSA) is a global health concern, which had been detected in food and food production animals. Conventional testing for detection of MRSA takes 3 to 5 d to yield complete information of the organism and its antibiotic sensitivity pattern. So, a rapid method is needed to diagnose and treat the MRSA infections. The present study focused on the development of a multiplex PCR assay for the rapid and sensitive detection of MRSA. The assay simultaneously detected 4 genes, namely, 16S rRNA of the Staphylococcus genus, femA of S. aureus, mecA that encodes methicillin resistance, and one internal control. It was rapid and yielded results within 4 h. The analytical sensitivity and specificity of the multiplex PCR assay was evaluated by comparing it with the conventional method. The analytical sensitivity of the multiplex PCR assay at the DNA level was 10 ng DNA. The analytical specificity was evaluated with 10 reference staphylococci strains and was 100%. The diagnostic evaluation of MRSA was carried out using 360 foodborne staphylococci isolates, and showed 99.1% of specificity, 96.4% of sensitivity, 97.5% of positive predictive value, and 97.3% of negative predictive value compared to the conventional method. The inclusion of an internal control in the multiplex PCR assay is important to exclude false‐negative cases. This test can be used as an effective diagnostic and surveillance tool to investigate the spread and emergence of MRSA.  相似文献

对应用不同浓度纳他霉素(NT)、乳酸链球菌(Nisin)、双乙酸钠(SDA)和脱氢醋酸钠(DHA)处理的油酥锅盔的保质期效果进行了研究。结果表明,不同浓度的防腐剂可不同程度地抑制油酥锅盔表面的茵落生长。最优组合为NT:Nisin=8:2(0.24g/kg:O.04g/kg,保质期为33d)和NT:SDA=7:3(0.21g/kg:1.2g/kg,保质期为33d),与对照组(保质期为20d)相比,大大延长了保质期。  相似文献

A simple and convenient HPLC-FD detection method for ochratoxin A (OTA) with a high detection limit and a short run time has been developed. OTA has been found in most samples of Chinese market wine, including domestic and imported wines, but the content was not very high. Only a few wines showed an OTA content that exceeded the EC and OIV limits, indicating that most Chinese market wines were safe. The OTA intake for Chinese from wine was 0.017 ng/kg (bw) per day, which was lower than the SCF and JECFA limits and also lower than in many other countries. This was mainly due to the low per capita wine consumption in China, but it is still necessary for the Chinese government and wine makers to monitor OTA levels in wine and to establish relevant regulations. An egg white treatment (0.20 mg/mL, 48 h) was the best removal method for OTA.  相似文献

This paper employs a novel Monte-Carlo based generation portfolio assessment tool to explore the implications of increasing wind penetration and carbon prices within future electricity generation portfolios under considerable uncertainty. This tool combines optimal generation mix techniques with Monte Carlo simulation and portfolio analysis methods to determine expected overall generation costs, associated cost uncertainty and expected CO2 emissions for different possible generation portfolios. A case study of an electricity industry with coal, Combined Cycle Gas Turbines (CCGT), Open Cycle Gas Turbines (OCGT) and wind generation options that faces uncertain future fossil-fuel prices, carbon pricing, electricity demand and plant construction costs is presented to illustrate some of the key issues associated with growing wind penetrations. The case study uses half-hourly demand and wind generation data from South Eastern Australia, and regional estimates of new-build plant costs and characteristics. Results suggest that although wind generation generally increases overall industry costs, it reduces associated cost uncertainties and CO2 emissions. However, there are some cases in which wind generation can reduce the overall costs of generation portfolios. The extent to which wind penetration affects industry expected costs and uncertainties depends on the level of carbon price and the conventional technology mix in the portfolios.  相似文献

In this article, we present a new approach in solving the problem of selecting proper and excellent cider yeast. Eight different yeast strains commonly used in the cider industry were selected for this experiment. Different ciders were fermented by the yeasts. Sensory evaluation was conducted first. Then, aroma components were identified by the headspace‐solid phase microextraction–gas chromatography–mass spectrometry. Their contents were determined using quantitative standards with known amounts of target components and 2‐octanol as internal standards. A model of fuzzy comprehensive evaluation was established and used to assess and rank the cider‐making attributes of all eight yeast strains. Saccharomyces cerevisiae F14 was selected out of the eight yeasts for the best cider. The results suggest that the method described in this article is more accurate and objective than conventional methods.  相似文献
