Rhodococcus jostii RHA1 accumulates chlorobenzoates (CBA) during the degradation of polychlorinated biphenyls (PCBs). CBA degradation is considered one of the rate-limiting steps in the complete degradation of PCBs. To reduce the accumulation of CBAs, the upper pathway enzyme genes for PCB degradation of RHA1 were introduced into a CBA-degrading bacterium, Burkholderia sp. NK8. The resulting recombinant strain exhibited no biphenyl 2,3-dioxygenase (BphA) activity encoded by bphAaAbAcAd genes, which encode the large and small subunits of the terminal oxygenase component and the ferredoxin and reductase subunits responsible for electron transfer from NADH to the large subunit. The remaining enzyme genes involved in the transformation of biphenyl to benzoate, bphB2C1D1, which encode dehydrogenase, ring-cleavage dioxygenase and hydrolase, conferred activities to NK8. To obtain the BphA activity of RHA1 in NK8, sets of BphA genes were constructed by combining the bphAaAbAcAd genes of RHA1 and bphA3A4 of Pseudomonas pseudoalcaligenes KF707, encoding the ferredoxin and reductase subunits. Hybrid derivatives of BphA containing the KF707 bphA3 conferred BphA activity to NK8, and a derivative containing the RHA1 bphAaAb and KF707 bphA3A4 genes exhibited the highest BphA activity. A plasmid containing the RHA1 bphAaAb and KF707 bphA3A4 genes plus the RHA1 bphB2C1D1 genes was constructed and introduced into NK8. The resulting recombinant strain efficiently degraded 2-, 3- and 4-chlorobiphenyls with an apparent reduction in CBA accumulation in comparison to the recombinant mutant strain, which had an insertion in the cbeA gene to inactivate CBA dioxygenase.  相似文献

Ca3Co4O9 (p-type) and Zn0.98Al0.02O (n-type) pellets were prepared by conventional sintering (CS) and Spark Plasma sintering (SPS) starting from the oxides. The best p-type sample was SPS Ca3Co4O9 obtained from pre-sintered pellets, with electrical conductivity σ = 144 S/cm and Seebeck coefficient S = 172 μV/K at 800 °C, while thermal conductivity κ = 2.00 W/m×K and figure of merit ZT = 0.23. The best n-type sample was CS Zn0.98Al0.02O showing σ = 83 S/cm and S = ?268 μV/K at 800 °C, while = 5.03 W/m×K and ZT = 0.127. The output power of a module based on SPS Ca3Co4O9 and CS Zn0.98Al0.02O legs was 2.26 mW (with T = 500 °C, ΔT = 248 °C).  相似文献

This paper proposes a new force sensorless robust tracking servo system, which detects and suppresses both the periodic disturbance and the sudden disturbance without force sensor. The proposed system estimates both the periodic disturbance and the sudden disturbance and the sudden disturbance by using a new sudden disturbance observer and the memory of tracking error. In the proposed system, the influence of sudden disturbance is suppressed by the proposed sudden disturbance observer. On the other hand, the influence of periodic disturbance is suppressed by means of ZPET feed‐forward servo system. The experimental results point out that the proposed tracking servo system has a precise tracking response against both the periodic disturbance and the sudden disturbance. © 2008 Wiley Periodicals, Inc. BACKGROUND: Neutrophils contribute to the host defense mechanism, but they can cause remote organ injury in peritonitis. The purpose of this study was to examine neutrophil adhesion to the peritoneum and remote organs simultaneously in peritonitis using a fluorescence microscopic method. STUDY DESIGN: Experiment 1: Sprague-Dawley rats (n = 16) were injected intraperitoneally (ip) with saline solution or 10(5), 10(7), or 10(9) Escherichia coli. Five hours after challenge, 1 x 10(6) fluorescein-labeled neutrophils were infused. Two minutes after neutrophil injection, five peritoneal samples (the greater omentum, mesentery, parietal peritoneum, colon, and ileum), both lungs, the liver, and the right kidney were harvested for counting of labeled neutrophils under epifluorescent microscopy. Lung myeloperoxidase (MPO) activity was also determined. Experiment 2: Rats (n = 23) were given 10(9) E. coli ip. Before challenge (0 h) or at 1, 5, or 10 h after challenge, labeled neutrophils were infused. Then, the labeled neutrophil numbers in organs and lung MPO activities were assessed as described for Experiment 1. Hemodynamic and arterial blood gas data were also obtained in another set of rats before and at 1, 5, 8 and 10 h after 10(9) E. coli ip challenge. RESULTS: Experiment 1: The labeled neutrophil numbers in the peritoneum, lungs, and kidney showed significant positive correlations with the injected bacterial numbers. Lung MPO also positively correlated with E. coli number and labeled neutrophil number in the lungs. Experiment 2: Labeled neutrophil numbers in the peritoneum and kidney peaked at 5 h. The pulmonary labeled neutrophil number rose, reaching a plateau at 5 h. No remarkable change was observed in the hepatic labeled neutrophil number. There was a positive correlation between lung MPO activity and pulmonary labeled neutrophil number. Hemodynamic and blood gas data reflected a hyperdynamic state. CONCLUSIONS: Concomitant dose-dependent increases in neutrophil adhesion in the peritoneum, lungs, and kidney were observed in this peritonitis model. Increased neutrophil adhesion was transient in the peritoneum and kidney but persistent in the lungs. Strategies modulating neutrophil adhesion in organs are anticipated to be useful for the treatment of peritonitis.  相似文献

The superconductivity of the tight binding electrons in a magnetic field is studied. We can treat both cases of quasi-one-dimension and quasi-two-dimension in the same manner. We study a various kind of anisotropic superconductivity with line nodes of the energy gap by taking attractive interaction between electrons in nearest sites along each axis. The magnetic field dependence of the transition temperature is calculated for a various pairing symmetry of superconductivity. When a magnetic field is applied in the conducting plane, the transition temperature is shown to increase as the magnetic field increases. In the strong magnetic field the eigenstates approach to those in the absence of hopping between planes, resulting in no orbital frustration.  相似文献

A water-swollen hydrogel with a molecularly-ordered structure was prepared by copolymerizing acrylic acid and acrylic monomer containing hydrophobic long alkyl or mesogenic moiety. The gels with a long alkyl side chain formed the crystalline structure and undergo the reversible order–disorder transition with change in temperature or solvent composition which accompanied a dramatic change in Young's modulus. These gels exhibited such chemomechanical behaviors as the shape memory and the rotational motility on the water surface. On the other hand, the gels with the mesogenic side chain formed the liquid crystalline structure with polymorphism in water, and mesophase diagram was established by changing copolymer composition and water content. Mechanism of these structural and chemomechanical behaviors has been explained in terms of order–disorder transition.  相似文献

A 3-dimensional (3D) simulation for a concentrator photovoltaic (CPV) module using a triple-junction solar cell was developed. By connecting ray-trace simulation for an optics model and 3D equivalent circuit simulation for a triple-junction solar cell, the operating characteristics of a CPV module were calculated. By using the 3D simulation, we considered the influence of tracking error on the CPV module. It was found that maximum output power was not correlated with optical efficiency of the optics system and was strongly dependent on fill factor. We can use this total 3D simulation for the evaluation and optimization of CPV modules.  相似文献
