The effects of plant-based extracts on the solar aging and antimicrobial properties of impregnated ethylene–norbornene (EN) copolymer and poly(lactic acid) (PLA) were investigated. In this study, the impregnation yield of polyolefin, lacking in active centers capable of phytochemical bonding, and polyester, abundant in active sides, was measured. Moreover, two different extracts plentiful in phytochemicals—thyme (TE) and clove (CE)—were employed in the solvent-based impregnation process. The effect of thymol and eugenol, the two main compounds embodied in the extracts, was studied as well. Interestingly, oxidation induction times (OIT) for the impregnation of EN with thyme and clove extracts were established to be, respectively, 27.7 and 39.02 min, which are higher than for thymol (18.4 min) and eugenol (21.1 min). Therefore, an aging experiment, mimicking the full spectrum of sunlight, was carried out to investigate the resistance to common radiation of materials impregnated with antioxidative substances. As expected, the experiment revealed that the natural extracts increased the shelf-life of the polymer matrix by inhibiting the degradation processes. The aging resistance was assessed based on detected changes in the materials’ behavior and structure that were examined with Fourier-transform infrared spectroscopy, contact angle measurements, color quantification, tensile tests, and hardness investigation. Such broad results of solar aging regarding materials impregnated with thyme and clove extracts have not been reported to date. Moreover, CE was found to be the most effective modifying agent for enabling material with antimicrobial activity against Escherichia coli to be obtained.  相似文献

Among the five somatostatin receptors (SST1–SST5), SST4 is the least characterized, which is in part due to the lack of specific monoclonal antibodies. We generated a knockin mouse model that expresses a carboxyl-terminal SST4-eGFP fusion protein. In addition, we extensively characterized the novel rabbit monoclonal anti-human SST4 antibody 7H49L61 using transfected cells and receptor-expressing tissues. 7H49L61 was then subjected to immunohistochemical staining of a series of formalin-fixed, paraffin-embedded normal and neoplastic human tissues. Characterization of SST4-eGFP mice revealed prominent SST4 expression in cortical pyramidal cells and trigeminal ganglion cells. In the human cortex, 7H49L61 disclosed a virtually identical staining pattern. Specificity of 7H49L61 was demonstrated by detection of a broad band migrating at 50–60 kDa in immunoblots. Tissue immunostaining was abolished by preadsorption of 7H49L61 with its immunizing peptide. In the subsequent immunohistochemical study, 7H49L61 yielded a predominant plasma membrane staining in adrenal cortex, exocrine pancreas, and placenta. SST4 was also found in glioblastomas, parathyroid adenomas, gastric and pancreatic adenocarcinomas, pheochromocytomas, and lymphomas. Altogether, we provide the first unequivocal localization of SST4 in normal and neoplastic human tissues. The monoclonal antibody 7H49L61 may also prove of great value for identifying SST4-expressing tumors during routine histopathological examinations.  相似文献

As much as space travel and exploration have been a goal since humankind looked up to the stars, the challenges coming with it are manifold and difficult to overcome. Therefore, researching the changes the human organism undergoes following exposure to weightlessness, on a cellular or a physiological level, is imperative to reach the goal of exploring space and new planets. Building on the results of our CellBox-1 experiment, where thyroid cancer cells were flown to the International Space Station, we are now taking advantage of the newest technological opportunities to gain more insight into the changes in cell–cell communication of these cells. Analyzing the exosomal microRNA composition after several days of microgravity might elucidate some of the proteomic changes we have reported earlier. An array scan of a total of 754 miRNA targets revealed more than 100 differentially expressed miRNAs in our samples, many of which have been implicated in thyroid disease in other studies.  相似文献

Analytical methods for molecular characterization of diagnostic or therapeutic targets have recently gained high interest. This review summarizes the combination of mass spectrometry and surface plasmon resonance (SPR) biosensor analysis for identification and affinity determination of protein interactions with antibodies and DNA-aptamers. The binding constant (KD) of a protein–antibody complex is first determined by immobilizing an antibody or DNA-aptamer on an SPR chip. A proteolytic peptide mixture is then applied to the chip, and following removal of unbound material by washing, the epitope(s) peptide(s) are eluted and identified by MALDI-MS. The SPR-MS combination was applied to a wide range of affinity pairs. Distinct epitope peptides were identified for the cardiac biomarker myoglobin (MG) both from monoclonal and polyclonal antibodies, and binding constants determined for equine and human MG provided molecular assessment of cross immunoreactivities. Mass spectrometric epitope identifications were obtained for linear, as well as for assembled (“conformational”) antibody epitopes, e.g., for the polypeptide chemokine Interleukin-8. Immobilization using protein G substantially improved surface fixation and antibody stabilities for epitope identification and affinity determination. Moreover, epitopes were successfully determined for polyclonal antibodies from biological material, such as from patient antisera upon enzyme replacement therapy of lysosomal diseases. The SPR-MS combination was also successfully applied to identify linear and assembled epitopes for DNA–aptamer interaction complexes of the tumor diagnostic protein C-Met. In summary, the SPR-MS combination has been established as a powerful molecular tool for identification of protein interaction epitopes.  相似文献

The empirical Complex Model developed by the US Environmental Protection Agency (EPA) is used by refiners to predict the toxic emissions of reformulated gasoline with respect to gasoline properties. The difficulty in implementing this model in the blending process stems from the implicit definition of Complex Model through a series of disjunctions assembled by the EPA in the form of spreadsheets. A major breakthrough in the refinery-based Complex Model implementation occurred in 2008 and 2010 through the use of generalized disjunctive and mixed-integer nonlinear programming (MINLP). Nevertheless, the execution time of these MINLP models remains prohibitively long to control emissions with our online gasoline blender. The first objective of this study is to present a new model that decreases the execution time of our online controller. The second objective is to consider toxic thresholds as hard constraints to be verified and search for blends that verify them. Our approach introduces a new way to write the Complex Model without any binary or integer variables. Sigmoid functions are used herein to approximate step functions until the measurement precision for each blend property is reached. By knowing this level of precision, we are able to propose an extremely good and differentiable approximation of the Complex Model. Next, a differentiable objective function is introduced to penalize emission values higher than the threshold emissions. Our optimization module has been implemented and tested with real data. The execution time never exceeded 1 s, which allows the online regulation of emissions the same way as other traditional properties of blended gasoline.  相似文献

Pyrrolizidine alkaloids (PAs) are secondary metabolites of plant families such as Asteraceae or Boraginaceae and are suspected to be genotoxic carcinogens. Recent investigations revealed their frequent occurrence in honey and particularly in tea. To obtain a comprehensive overview of the PA content in animal- and plant-derived food from the European market, and to provide a basis for future risk analysis, a total of 1105 samples were collected in 2014 and 2015. These comprised milk and milk products, eggs, meat and meat products, (herbal) teas, and (herbal) food supplements collected in supermarkets, retail shops, and via the internet. PAs were detected in a large proportion of plant-derived foods: 91% of the (herbal) teas and 60% of the food supplements contained at least one individual PA. All types of (herbal) teas investigated were found to contain PAs, with a mean concentration of 460 µg kg?1 dry tea (corresponding to 6.13 µg L?1 in [herbal] tea infusion). The highest mean concentrations were found in rooibos tea (599 µg kg?1 dry tea, 7.99 µg L?1 tea infusion) and the lowest in camomile tea (274 µg kg?1 dry tea, 3.65 µg L?1 tea infusion). Occurrence of PAs in food supplements was found to be highly variable, but in comparable ranges as for (herbal) tea. The highest concentrations were present in supplements containing plant material from known PA-producing plants. In contrast, only 2% of the animal-derived products, in particular 6% of milk samples and 1% of egg samples, contained PAs. Determined levels in milk were relatively low, ranged between 0.05 and 0.17 µg L?1 and only trace amounts of 0.10–0.12 µg kg?1 were found in eggs. No PAs were detected in the other animal-derived products.  相似文献

In this study, the characteristics of nanocellulose extracted from bleached softwood and hardwood pulps by formic acid hydrolysis followed by TEMPO-mediated oxidation were compared using transmission electron microscopy (TEM), atomic force microscopy (AFM), Fourier transform infrared analysis (FT-IR), X-ray diffraction (XRD), and thermal gravimetric analysis (TGA). The experimental results showed that the nanocellulose products derived from spruce pulp exhibited a relatively larger particle size, higher crystallinity, and higher thermal stability, compared with the corresponding products obtained from aspen pulp under the same conditions. Furthermore, the study helped establish that the properties of the nanocellulose products were highly dependent on the nature of the starting materials under identical processing conditions.  相似文献
