The objective of this study was to achieve an optimal formulation of hydrophilic–hydrophobic conjugates for nano-sized solid dispersions (SDs) with enhanced dissolution of multiple drugs in different gastrointestinal (GI) tract environments. A new conjugate powder with an optimized process was used to fabricate SDs that contained three poorly water-soluble drugs that were also poorly soluble in different dissolution media. The self-assembled nanoparticle formation, drug crystallinity and SD molecular interactions were investigated by measuring the particle size during dissolution testing and physicochemical property analysis (powder X-ray diffraction and Fourier transform infrared spectroscopy). Drug release studies indicated that SD containing conjugated powder significantly improved the dissolution rates of these poorly water-soluble drugs in the GI tract. In addition, particle size analysis showed nano-sized particles in the dissolution media in the early stage with a tendency to reduce smaller particles over time. Physicochemical characterizations demonstrated almost amorphous drug states and hydrogen bonding interactions between the drugs and conjugates in the SD. This study optimized a promising material for SD, and the material was shown to have a promising performance under various pH medium conditions with poorly water-soluble drugs.  相似文献

The physicochemical and physiological attributes of three contrasting commercial varieties of Musaceae, Dominico Harton (plantain), Guineo (cooking banana) and Gros Michel (dessert banana), were evaluated and statistically analysed during post-harvest ripening. Quality attributes differed markedly among varieties, both in fresh fruits and during ripening. Variety (V) had a significant effect (P < 0.001) on all attributes except total soluble solids (TSS), carotenes and total chlorophyll. Storage time (ST) had a significant effect on all attributes except colour parameter b* and total carotenes. Starch levels decreased significantly (P < 0.001) during ripening, with nearly complete hydrolysis in Gros Michel, followed by Guineo and Dominico Harton. Discriminant analysis showed that central diameter, TSS of the pulp, colour parameter a* and total starch had the highest weight in the differentiation among varieties. These results point out which parameters may help improve current methods for monitoring ripening of bananas, in particular the commercially important varieties in this study.  相似文献

Effective reversal of tumor immunosuppression is of critical importance in cancer therapy. A multifunctional delivery vector that can effectively deliver CRISPR-Cas9 plasmid for β-catenin knockout to reverse tumor immunosuppression is constructed. The multi-functionalized delivery vector is decorated with aptamer-conjugated hyaluronic acid and peptide-conjugated hyaluronic acid to combine the tumor cell/nuclear targeting function of AS1411 with the cell penetrating/nuclear translocation function of TAT-NLS. Due to the significantly enhanced plasmid enrichment in malignant cell nuclei, the genome editing system can induce effective β-catenin knockout and suppress Wnt/β-catenin pathway, resulting in notably downregulated proteins involved in tumor progression and immunosuppression. Programmed death-ligand 1 (PD-L1) downregulation in edited tumor cells not only releases the PD-1/PD-L1 brake to improve the cancer killing capability of CD8+ T cells, but also enhances antitumor immune responses of immune cells. This provides a facile strategy to reverse tumor immunosuppression and to restore immunosurveillance and activate anti-tumor immunity.  相似文献

Chatter causes machining instability and reduces productivity in the metal cutting process. It has negative effects on the surface finish, dimensional accuracy, tool life and machine life. Chatter identification is therefore necessary to control, prevent, or eliminate chatter and to determine the stable machining condition. Previous studies of chatter detection used either model-based or signal-based methods, and each of them has its drawback. Model-based methods use cutting dynamics to develop stability lobe diagram to predict the occurrence of chatter, but the off-line stability estimation couldn’t detect chatter in real time. Signal-based methods apply mostly Fourier analysis to the cutting or vibration signals to identify chatter, but they are heuristic methods and do not consider the cutting dynamics. In this study, the model-based and signal-based chatter detection methods were thoroughly investigated. As a result, a hybrid model- and signal-based chatter detection method was proposed. By analyzing the residual between the force measurement and the output of the cutting force model, milling chatter could be detected and identified efficiently during the milling process.

New methods for synthesis of active materials have been developed to improve capacity and cycle life performance of lithium‐ion batteries. Past studies have focused on routes of development of materials and new processes, which might not be economical for large‐scale production. In this regard, this study examines a widely employed carbothermal reduction technology for the synthesis of lithium‐iron phosphate (LiFePO4/C) and investigates effects of process conditions during this synthesis on final battery performance. An experimental combined genetic programming approach is used to model the effects of crucial process conditions (sintering time, the carbon content, and the sintering temperature) on the discharge capacity of the assembled battery. Experiments are conducted to collect the discharge capacity data based on varying LiFePO4/C synthesis conditions, and genetic programming is employed to develop a suitable functional relationship between them. The results show that the battery discharge capacity is controlled significantly by adjusting sintering temperature and carbon content, while the effect of sintering time is found to be insignificant. Further, the interaction effect of the sintering time and carbon content is much more obvious than that of the sintering time and the sintering temperature. The findings from the study pave the way for the optimum design of the synthesis process of LiFePO4/C for a higher battery performance.  相似文献

The grain growth kinetics and mechanical properties of graphene platelets(GPLs) reinforced ZrO2/Al2O3(ZTA) composites prepared by microwave sintering were investigated. The calculated grain growth kinetics exponent n indicated that the GPLs could accelerate the process of the Al2O3 columnar crystal growth. And the grain growth activation energy of the Al2O3 columnar crystal indicated that the grain growth activation energy of the GPLs doped ZTA composites is much higher than those of pure Al2O3 and ZTA in microwave sintering. The optimal mechanical properties were achieved with 0.4?vol% GPLs, whose relative density, Vickers hardness and fracture toughness were 98.76%, 18.10?GPa and 8.86?MPa?m1/2, respectively. The toughening mechanisms were crack deflection, bridging, branching and pull-out of GPLs. The results suggested that GPLs-doped are good for the Al2O3 columnar crystal growth in the ZTA ceramic and have a potentially improvement for the fracture toughness of the ceramics.  相似文献

This work aimed at producing and characterising xylose‐glycosylated peanut protein isolate (PPI‐X) films by dissolving PPI‐X powder in water at ambient temperature and further plasticising with glycerol. The effect of powder dissolution temperature (20–100 °C) and glycerol concentration (15.0–45.0%, w/w) on mechanical properties and integrity of these films was quantified. The results showed that the powder dissolution temperature had no significant effect on the mechanical and water resistance properties of PPI‐X films within the temperature range tested. With increasing concentration of glycerol, the tensile strength and water resistance of PPI‐X films decreased and elongation increased. The films produced by dissolving the PPI‐X powder at 20 °C and plasticising with 25.0% glycerol had comparable mechanical properties and better water resistance compared to some other plant protein films plasticising with glycerol. The results suggested that PPI‐X films could potentially be used as biodegradable packaging materials.  相似文献

用电沉积方法制备了非晶态铬镀层,用X射线衍射仪分析了镀层的相结构,对镀层的组织与性能以及热处理对镀层组织与性能的影响进行了研究。结果表明,非晶态铬镀层的硬度值约为969HV0.025,热处理后由于Cr7C3相的析出使硬度最高可升至1345HV0.025,非晶态铬镀层表现出良好的耐磨性和耐蚀性能,明显优于普通晶态铬镀层。热处理后,镀层与基体的结合力也有明显提高。  相似文献

采用刚塑性有限元法对钛合金精锻叶片的锻造过程进行了二维有限元数值模拟。在数值模拟过程中 ,考虑了摩擦边界的作用和畸变网格的局部重划分。根据数值模拟结果 ,研究了不同锻造工艺参数 (变形温度、变形速率、压下量等 )对叶片锻造过程中的变形力、应力和应变变化的影响规律。  相似文献
