A polymerase chain reaction (PCR) assay was developed to rapidly detect and identify West Nile (WN) virus. The RNA from seven isolates of WN virus from six countries and four other flaviviruses (Kunjin, Japanese encephalitis, St. Louis encephalitis, and yellow fever viruses) was reverse-transcribed (RT) and amplified by PCR. The nucleotide sequences of the amplified products were determined by a rapid, automated DNA sequencing method. The WN virus RT/PCR assay detected the target gene segment of sequencing method. The WN virus RT/PCR assay detected the target gene segment of isolates from both the African-Middle Eastern group and the Indian group with a sensitivity of approximately 0.05 pg of viral RNA. Kunjin virus was the only other flavivirus tested that produced a band of the appropriate size. Five of seven WN virus isolates showed 92-98% homology in the nucleotide sequence of their PCR products. The sequence of one isolate was virtually identical to the published sequence of the Nigerian isolate (99.5% homology). No correlation was established between the degree of nucleotide homology, geographic location, time of isolation, or source of the isolates.  相似文献

A test equipment was designed to study thermal shock and thermal fatigue of ceramic materials subjected to fast heating (ascending). The equipment was designed to generate thermal stress in a test specimen by heating one surface of it by an oxy-hydrogen flame while cooling the opposite surface. The sample cracked when thermal stress exceeded its mechanical strength. The in situ crack formation was detected by an acoustic emission system coupled to the set up. The hot zone temperature was measured by an infra red pyrometer. The equipment was also designed to run thermal fatigue test cycles in automatic mode between two selected temperatures. The temperature and thermal stress distribution in the test specimen were modelled using finite element software. The effect of temperature distribution of the top and bottom surfaces on thermal stresses was studied. It was observed that the thermal stress is very sensitive to the temperature distribution on the top surface and maximum near the periphery of the top surface. This was in agreement with the experimental results in which the cracks were originated from the periphery of top surface. It was also observed that the failure temperature was higher for thicker samples.  相似文献

Powder compaction is a critical step in a classical method for the production of zero-defect ceramic products. We have studied the development of this step to determine whether acoustic emission can be used to detect the presence of flaws in a compact. In uniaxial pressing at room temperature, the creation and extension of flaws occur principally during the ejection of the compacts. The type and extent of flaws depend on the mechanical properties of the powder used and on the pressing conditions. Acoustic emission was used to monitor the consolidation by uniaxial pressing of Al2O3 powders and a UO2---PuO2 powder. It was possible to detect flaws in the compacts, to determine their type for a given powder, and to identify the type of powder used by a direct examination of cumulative counts of acoustic emission events and by a statistical analysis of amplitude distributions. Hence, the introduction of this technique in a fabrication process offers the possibility of detecting the presence of defects in compacts as soon as they are created. This would decrease the difficulties involved in reuse in the process of the powder from defective products, and would limit further flaw development during sintering, which are two major problems in the nuclear industry.  相似文献

Knowledge representation issues related to the modelling of imprecision and uncertainty are discussed in the framework of possibility theory. Differences and relations between possibility theory, fuzzy sets, probability theory and Shafer evidence theory are presented. Then, patterns of reasoning and inference and control procedures are studied in presence of uncertainty and imprecision using a possibilistic approach. the basic ideas and the main trends are emphasized rather than the mathematical and logical foundations or the technical details of implementation which can be found in the references.  相似文献

The mechanofusion process, a dry particle coating route, has been successfully applied to coat iron particles with alumina. The high-temperature oxidation resistance of Fe and coated Fe particles has been comparatively investigated in air in the temperature range 350°–900°C. The activation energy calculations demonstrate that, in the temperature range 350°–420°C, the oxidation mechanisms are the same for both particles. Scanning electron microscopy and energy-dispersive spectroscopy analyses show that oxidation products are mainly formed at the iron/alumina interface at low temperatures or at the alumina/gas interface at high temperatures. It is shown that, depending on the temperature range, two different oxidation mechanisms are predominant.  相似文献

Self-aligned single-dot memory devices and arrays were fabricated based on arsenic-assisted etching and oxidation effects. The resulting device has a floating gate of about 5-10 nm, presenting single-electron memory operation at room temperature. The Canadian Armed Forced have been deployed to the republics of the former Yugoslavia since 1992 as part of the United Nations Protection Force and the NATO-led Implementation Force. Most of the combat arms units have been supported by small, self-contained surgical teams for essential surgery. Considerable benefit is gained by cooperating with civilian surgeons. The experience of treating five patients with complicated hydatid disease endemic to the area is examined. Treatment of these patients requires performing major surgical procedures under austere conditions and must be undertaken with care. Careful selection and communication with the surgical nursing team and civilian surgeons is essential. Well selected cases can also pay tremendous dividends in terms of maintaining the skills of personnel who must be prepared for any emergency in addition to providing vital surgical assistance to these patients.  相似文献

The silylation reaction of dextran with 1,1,1,3,3,3-hexamethyldisilazane (HMDS) in DMSO was studied as the first step of the synthesis of new amphiphilic polyester-grafted dextrans. According to the experimental conditions, i.e. dextran molar weight, medium temperature and reaction time, HMDS/OH ratio, addition of a catalyst and co-solvent, partially or totally silylated dextrans were recovered. The highest silylation yields were obtained with the lowest molecular weight dextrans. The increase in temperature medium and/or reaction time, the presence of catalyst or co-solvent favored the protection yield. Whatever the dextran used, complete silylation of the polysaccharide chain could be achieved by adequate selection of the experimental conditions. The thermal properties of resulting silylated polysaccharides were investigated by temperature modulated DSC. It was observed that Tg values of partially silylated dextran were maintained between 120 and 140 °C, independently of the dextran molecular weight. Interestingly, DMSO proved to behave as an efficient plasticizer of (partially) silylated dextrans. The partially silylated dextrans were efficiently used as multifunctional macroinitiators for the controlled ring-opening polymerization (ROP) of lactone. The ROP was then promoted from the remaining hydroxyl groups in the presence of tin or aluminium activator. After polymerization and ultimate deprotection of the silylated dextran backbone, amphiphilic polyester-grafted dextrans were readily recovered.  相似文献

New vanadium oxide supported on mesoporous silica catalysts for the oxidation of methane to formaldehyde were investigated by infrared and Raman spectroscopies to identify and characterize the molecular structure of the most active and selective catalytic sites. In situ and operando experiments have been conducted in order to understand the redox and hydroxylation/dehydroxylation processes of the vanadium species. (SiO)2VO(OH) species were identified in these catalysts in reaction conditions and shown to undergo a deprotonation at 580 °C under vacuum, leading to a site giving a photoluminescence band at 550 nm attributed to reverse radiative decay from the excited triplet state:

(V4+–O)*  (V5+O2−). An activation mechanism of vanadium monomeric species with electrophilic oxygen species is proposed.  相似文献
