A novel gene fusion system to express and purify small recombinantproteins in Escherichia coli has been constructed. The conceptallows for affinity purification of soluble gene products bysequential albumin- and Zn2+-affinity chromatography. The dual-affinitysystem is well suited for expression of unstable proteins asonly full-length protein is obtained after purification andproteins gain proteolytic stability in the fusion protein. Herewe show that the dual-affinity approach can be used for theexpression of various unstable derivatives of a single IgG-bindingdomain based on staphylococcal protein A. Analysis of the proteolyticstabilities and the IgG-binding properties of the differentmutant proteins suggest that the model for the structure ofan IgG-binding domain must be re-evaluated.  相似文献

Enterprise architecture is a model-based IT and business management discipline. Enterprise architecture analysis concerns using enterprise architecture models for analysis of selected properties to provide decision support. This paper presents a framework based on the ArchiMate metamodel for the assessment of four properties, viz., application usage, system availability, service response time and data accuracy. The framework integrates four existing metamodels into one and implements these in a tool for enterprise architecture analysis. The paper presents the overall metamodel and four viewpoints, one for each property. The underlying theory and formalization of the four viewpoints is presented. In addition to the tool implementation, a running example as well as guidelines for usage makes the viewpoints easily applicable.  相似文献

Enterprise Architecture (EA) is an approach used to provide decision support based on organization-wide models. The creation of such models is, however, cumbersome as multiple aspects of an organization need to be considered, making manual efforts time-consuming, and error prone. Thus, the EA approach would be significantly more promising if the data used when creating the models could be collected automatically—a topic not yet properly addressed by either academia or industry. This paper proposes network scanning for automatic data collection and uses an existing software tool for generating EA models (ArchiMate is employed as an example) based on the IT infrastructure of enterprises. While some manual effort is required to make the models fully useful to many practical scenarios (e.g., to detail the actual services provided by IT components), empirical results show that the methodology is accurate and (in its default state) require little effort to carry out.  相似文献

River management based solely on physical science has proven to be unsustainable and unsuccessful, evidenced by the fact that the problems this approach intended to solve (e.g., flood hazards, water scarcity, and channel instability) have not been solved and long‐term deterioration in river environments has reduced the capacity of rivers to continue meeting the needs of society. In response, there has been a paradigm shift in management over the past few decades, towards river restoration. But the ecological, morphological, and societal benefits of river restoration have, on the whole, been disappointing. We believe that this stems from the fact that restoration overrelies on the same physical analyses and approaches, with flowing water still regarded as the universally predominant driver of channel form and structural intervention seen as essential to influencing fluvial processes. We argue that if river restoration is to reverse long‐standing declines in river functions, it is necessary to recognize the influence of biology on river forms and processes and re‐envisage what it means to restore a river. This entails shifting the focus of river restoration from designing and constructing stable channels that mimic natural forms to reconnecting streams within balanced and healthy biomes, and so levering the power of biology to influence river processes. We define this new approach as biomic river restoration.  相似文献

Surface patterning is a recent promising approach to promote performance of pressure-driven membranes in water treatment and desalination. Nevertheless, knowledge about foulant deposition mechanisms, especially at early stage of filtration, is still lacking. The applicability of particle imaging velocimetry to study fluid characteristics atop surface patterned thin-film composite membranes was investigated at different operating conditions. This work is an important first step toward reliable understanding of the impacts of topographical membrane surface modification on hydrodynamic conditions and foulant deposition mechanisms.  相似文献

Reusing wastewater from oil-related industries is becoming increasingly important, especially in water-stressed oil-producing countries. Before oily wastewater can be discharged or reused, it must be properly treated, e.g., by membrane-based processes like ultrafiltration. A major issue of the applied membranes is their high fouling propensity. This paper reports on mitigating fouling inside ready-to-use ultrafiltration hollow-fiber modules used in a polishing step in oil/water separation. For this purpose, in-situ polyzwitterionic hydrogel coating was applied. The membrane performance was tested with oil nano-emulsions using a mini-plant system. The main factors influencing fouling were systematically investigated using statistical design of experiments.  相似文献

The potential benefits of using human resources efficiently in the service sector constitute an incentive for decision makers in this industry to intelligently manage the work shifts of their employees, especially those dealing directly with customers. In the long term, they should attempt to find the right balance between employing as few labor resources as possible and keeping a high level of service. In the short run (e.g., 1 week), however, contracted staff levels cannot be adjusted, and management efforts thus focus on the efficient assignment of shifts and activities to each employee. This article proposes a mixed integer program model that solves the short-term multi-skilled workforce tour scheduling problem, enabling decision makers to simultaneously design workers’ shifts and days off, assign activities to shifts and assign those to employees so as to maximize and balance coverage of a firm’s demand for on-duty staff across multiple activities. Our model is simple enough to be solved with a commercial MIP solver calibrated by default without recurring to complex methodologies, such as extended reformulations and exact and/or heuristic column generation subroutines. A wide computational testing over 1000 randomly generated instances suggests that the model’s solution times are compatible with daily use and that multi-skilling is a significant source of labor flexibility to improve coverage of labor requirements, in particular when such requirements are negatively correlated and part-time workers are a scarce resource.  相似文献

Given the strong increase in regulatory requirements for business processes the management of business process compliance becomes a more and more regarded field in IS research. Several methods have been developed to support compliance checking of conceptual models. However, their focus on distinct modeling languages and mostly linear (i.e., predecessor-successor related) compliance rules may hinder widespread adoption and application in practice. Furthermore, hardly any of them has been evaluated in a real-world setting. We address this issue by applying a generic pattern matching approach for conceptual models to business process compliance checking in the financial sector. It consists of a model query language, a search algorithm and a corresponding modelling tool prototype. It is (1) applicable for all graph-based conceptual modeling languages and (2) for different kinds of compliance rules. Furthermore, based on an applicability check, we (3) evaluate the approach in a financial industry project setting against its relevance for decision support of audit and compliance management tasks.  相似文献

Robots that use vision for localization need to handle environments that are subject to seasonal and structural change, and operate under changing lighting and weather conditions. We present a framework for lifelong localization and mapping designed to provide robust and metrically accurate online localization in these kinds of changing environments. Our system iterates between offline map building, map summary, and online localization. The offline mapping fuses data from multiple visually varied datasets, thus dealing with changing environments by incorporating new information. Before passing these data to the online localization system, the map is summarized, selecting only the landmarks that are deemed useful for localization. This Summary Map enables online localization that is accurate and robust to the variation of visual information in natural environments while still being computationally efficient. We present a number of summary policies for selecting useful features for localization from the multisession map, and we explore the tradeoff between localization performance and computational complexity. The system is evaluated on 77 recordings, with a total length of 30 kilometers, collected outdoors over 16 months. These datasets cover all seasons, various times of day, and changing weather such as sunshine, rain, fog, and snow. We show that it is possible to build consistent maps that span data collected over an entire year, and cover day‐to‐night transitions. Simple statistics computed on landmark observations are enough to produce a Summary Map that enables robust and accurate localization over a wide range of seasonal, lighting, and weather conditions.  相似文献

Free Material Optimization (FMO) is a powerful approach for conceptual optimal design of composite structures. The design variable in FMO is the entire elastic material tensor which is allowed to vary almost freely over the design domain. The imposed requirements on the tensor are that it is symmetric and positive semidefinite. Most of today’s studies on FMO focus on models for two- and three-dimensional structures. The objective of this article is to extend existing FMO models and methods to laminated plate and shell structures, which are used in many engineering applications. In FMO, the resulting optimization problem is generally a non convex semidefinite program with many matrix inequalities which requires special-purpose optimization methods. The FMO problems are efficiently solved by a primal-dual interior point method developed and implemented by the authors. The quality of the proposed FMO models and the method are supported by several large-scale numerical experiments.  相似文献
