Bcl-2 and Bax are homologous proteins which can heterodimerize with each other. These proteins have opposing effects on cell survival when overexpressed in cells, with Bcl-2 blocking and Bax promoting apoptosis. Here we demonstrate that gene transfer-mediated elevations in Bcl-2 protein levels result in a marked increase in the steady-state levels of endogenous p21Bax protein as determined by immunoblotting in the Jurkat T-cell and 697 pre-B-cell leukemia cell lines, but not in several other cell lines including CEM T-cell leukemia, 32D.3 myeloid progenitor, PC12 pheochromocytoma, and NIH-3T3 fibroblasts. Steady-state levels of p21Bax protein were also elevated in the lymph nodes of Bcl-2 transgenic mice in which a BCL-2 transgene is expressed at high levels in B-cells. Northern blot analysis of BCL-2-transfected and control-transfected Jurkat and 697 leukemia cells revealed no Bcl-2-induced alterations in the steady-state levels of BAX mRNAs. In contrast, L-[35S]methionine pulse-chase analysis indicated a marked increase in the half-life (t1/2) of the p21Bax protein in BCL-2-transfected 697 cells compared to control-transfected cells (t1/2 > 24 h versus approximately 4 h), whereas the rate of Bax degradation was unaltered in Bcl-2-transfected CEM cells. The results demonstrate that levels of the proapoptotic p21Bax protein can be post-translationally regulated by Bcl-2, probably in a tissue-specific fashion, and suggest the existence of a feedback mechanism that may help to maintain the ratio of Bcl-2 to Bax protein in physiologically appropriate ranges. 相似文献

The influence of four industrial wastewater types (organic chemical, dye, leather, and winery industries) on the exopolymeric substances (EPSs) constituents [protein, polysaccharide, and deoxyribonucleic acid (DNA)] and physicochemical properties of flocs (contact angle, surface charge, and bound water) was determined. Four laboratory scale activated sludge systems were used; they were fed with real wastewaters and were operated continuously under steady-state conditions. The EPS analysis showed that between 70 and 80% of the extracellular organic material could be attributed to protein. The proteins in the bound form were quantitatively greater than the soluble protein form in winery industry floc EPSs. The soluble protein ratios were higher than bound protein ratios in organic chemical, dye, and leather industries flocs. Lower amounts of protein and high DNA content were found in the EPS flocs grown on wastewaters containing more complex substrates (organic chemical, leather, and dye). Changing the wastewater from chemical, leather, and dye to that of winery industry resulted in an increase in protein and a decrease in DNA level of floc EPSs. The sludge surface of winery industry flocs were less hydrophobic (small contact angle) and more highly negatively charged compared to chemical, dye, and leather industry flocs. A higher sludge volume index (SVI), an indication of poorer settleability, was associated with both small amounts of EPS and protein and a high amount of DNA. Both high hydrophilic and negatively charged surfaces corresponded to lower levels of SVI and good settling properties. The results of this study demonstrated that wastewater type influenced the EPS composition and physicochemical properties of sludge. The surface properties (surface charge, contact angle, and bound water) and composition of EPS, apart from the carbohydrate content, governed the settleability of sludge and were important in controlling the SVI. 相似文献

The trapping technique of Singleton and Pattee, in which the volatiles are swept to a Porapak Q trap by suction rather than by a sweeping gas, was modified and used to isolate soursop juice volatiles. Using fixed, low vacuum and operating conditions determined specifically for soursop volatiles, very high reproducibility was obtained. The majority of the peaks had a coefficient of variation less than 0.10. The relative amounts of the volatile components obtained by direct headspace sampling was fairly well maintained in the Porapak Q concentrate. The chromatograms showed distinct differences between two commercial brands or between lots of the same brand of soursop juice. The trapping technique appear to have wide applicability provided that the operating conditions are adapted to each product studied. 相似文献

We examined the regulation of apoptosis, radiosensitivity, and spindle checkpoint in response to DNA-damaging agents in ataxia telangiectasia (AT)-derived lymphoblastoid cell lines (AT-LCLs), which lack AT mutated (ATM) protein expression. In addition to the previous findings that AT-LCLs are defective in regulation of cell cycle at the G1, S, and G2-M checkpoints in response to X-ray irradiation (X-IR) and are highly sensitive to X-IR (J. Biol. Chem., 271: 20486-20493, 1996), we showed for the first time that AT-LCLs were defective in X-IR-associated spindle checkpoint control. The cells were also resistant to early apoptosis as much as LCLs derived from patients with Li-Fraumeni syndrome (LFS-LCLs). Terminal deoxynucleotidyl transferase-mediated nick end labeling assay of LCLs, however, demonstrated a significant increase in apoptotic cells among AT-LCLs cultured over a longer period after X-IR. These findings were in contrast to those of LFS-LCL, which showed very little increase in terminal deoxynucleotidyl transferase-mediated nick end labeling-positive population, even in cells with hyperploidy. Thus, although early apoptosis and cell cycle controls in response to DNA damage are disrupted in both ATM and p53 mutations, cells from AT patients are much more susceptible to late-onset apoptosis than those of LFS. These differences may depend on the level of accumulation of DNA damage and/or threshold that triggers late-onset cell death in ATM or p53 mutations. Prussian Blue-modified graphite electrodes (G/PB) with electrocatalytic activity toward H2O2 reduction were obtained by PB potentiostatic electrodeposition from a mixture containing 2.5 mm FeCl3 + 2.5 mm K3[Fe(CN)6] + 0.1 m KCl + 0.1 m HCl. From cyclic voltammetric measurements, performed in KCl aqueous solutions of different concentrations (5 × 10−2–1 m), the rate constant for the heterogeneous electron transfer (k
s) was estimated by using the Laviron treatment. The highest ks value (10.7 s−1) was found for 1 m KCl solution. The differences between the electrochemical parameters of the voltammetric response, as well as the shift of the formal potential, observed in the presence of Cl− and NO3− compared to those observed in the presence of SO42− ions, points to the involvement of anions in the redox reactions of PB. The G/PB electrodes showed a good electrochemical stability proved by a low deactivation rate constant (0.8 × 10−12 mol cm2 s−1). The electrocatalytic efficiency, estimated as the ratio

This paper introduces a multi-agent behavioral-based optimization algorithm for system level radio design. Making multi-standard
wireless communication receivers that meet their specs while keeping the requirements of the individual blocks as relaxed
as possible is the goal of this algorithm. In order to achieve this goal a “divide and conquer” approach is proposed. Different
agents focus on different objectives that are pursued in parallel. Agents adopt different behaviors depending on the status
of the environment and their interaction with other agents. Agents are cooperative by default as they try to meet their spec
without making changes that affect other agents. However, more aggressive behaviors that lead to global changes can be adopted
when needed. The interaction between these simple entities yields an emergent behavior able to deal smoothly with the complexity
