Prostate cancer cells adhere to a tumor basement membrane, while secretory epithelial cells reside in a suprabasal cell compartment. Since tumor cells are derived from suprabasal epithelial cells, they experience de-novo substratum adhesion in the context of oncogenesis. We therefore analyzed whether cell-matrix adhesion could affect the protein expression and activity of the AR. In this study, AR protein expression declined upon suspension of BPH-1-AR cells, but not in PC-3-AR cells shown by Western blot. In a time course study, BPH-1 cell lost AR expression within 6 hours, and the synthetic androgen, R1881 reduced the loss of AR expression. We further explored the mechanism of AR loss in suspended BPH-1 cells. BPH-1-AR cells underwent apoptosis (anoikis) when suspended for 2 – 5 hours. Suspension did not induce significant apoptosis or decreasing of AR expression in PC-3 cells. Inhibition of apoptosis in suspended BPH-1-AR cells, either by expression of Bcl-2 or Bcl-xl or by treatment with Z-VAD, a caspase inhibitor, prevented loss of AR protein. In contrast, the calpain protease inhibitor, ALLN, accelerated the loss of AR protein expression. Additionally, cell-matrix adhesion changed the expression of coregulators of AR in the mRNA level of prostate cancer cells. Our results demonstrate that AR protein expression was reduced through activation of cell death pathways, and thus indirectly through cell suspension in BPH-AR cells. The activity of AR can also be regulated by adhesion in PC-3-AR and LNCaP cells through affecting the coregulators level. 相似文献

This paper reports the results of investigations of shellfish toxin contamination of products obtained from Shanghai seafood markets. From May to October 2003, 66 samples were collected from several major seafood markets. Paralytic shellfish poisoning (PSP) and diarrhetic shellfish poisoning (DSP) toxins in shellfish samples were monitored primarily by a mouse bioassay, then analysed by HPLC for the chemical contents of the toxins. According to the mouse bioassay, eight samples were detected to be contaminated by PSP toxins and seven samples were contaminated by DSP toxins. Subsequent HPLC analysis indicated that the concentrations of the PSP toxins ranged from 0.2 to 1.9 µg/100 g tissues and the main components were gonyautoxins 2/3 (GTX2/3). As for DSP, okadaic acid was detected in three samples, and its concentration ranged from 3.2 to 17.5 µg/100 g tissues. Beside okadaic acid, its analogues, dinophysistoxins (DTX1), were found in one sample. According to the results, gastropod (Neverita didyma) and scallop (Argopecten irradians) were more likely contaminated with PSP and DSP toxins, and most of the contaminated samples were collected from Tongchuan and Fuxi markets. In addition, the contaminated samples were always found in May, June and July. Therefore, consumers should be cautious about eating the potential toxic shellfish during this specific period. 相似文献

高层建筑顶部塔冠形状多样,其对建筑整体气动力的影响目前尚不明确。为此,利用高频测力天平技术,在大气边界层风场下对19种不同顶部塔冠的方形截面超高层建筑进行了风洞试验,分析塔冠形状对超高层建筑顺风向基底弯矩系数值及顺风向基底弯矩功率谱的影响。讨论坡面变化、顶部开洞、阶梯变化和锥形变化等四大类塔冠形状对结构整体顺风向气动力的影响规律。试验结果表明:将坡面置于迎风侧能够降低顺风向基底弯矩系数值;相同开洞形状和面积时,顶部敞口开洞对降低顺风向基底弯矩平均值的效果优于闭口开洞;斜坡面阶梯塔冠对降低顺风向弯矩系数值的效果优于直坡面阶梯塔冠;上棱椎下棱台的棱椎式塔冠能够大幅减小顺风向基底弯矩系数平均值。塔冠能够改变顺风向基底弯矩功率谱峰值大小,棱锥形塔冠对降低顺风向弯矩功率谱峰值最为明显。 相似文献

基于对图像拼接技术的分析,提出了一种基于马尔科夫模型与Hilbert-Huang变换(HHT)的图像拼接盲检测算法。该算法计算图像DCT域上的马尔科夫转移概率矩阵,同时对图像进行Hilbert-Huang分析,得到两类特征值集,并通过计算相关系数矩阵分析了两者之间的相关性,最后使用支持向量机进行训练与分类。实验结果表明,相对于已有文献,该算法具有较高的检测准确率。 相似文献

Novel, thiol‐functionalized, and superparamagnetic, silica composite nanospheres (SH‐SSCNs) with diameters smaller than 100 nm are successfully fabricated through the self‐assembly of Fe3O4 nanoparticles and polystyrene100‐block‐poly(acrylic acid)16 and a subsequent sol‐gel process. The size and magnetic properties of the SH‐SSCNs can be easily tuned by simply varying the initial concentrations of the magnetite nanoparticles in the oil phase. By incorporating fluorescent dye molecules into the silica network, the composite nanospheres can be further fluorescent‐functionalized. The toxicity of the SH‐SSCNs is evaluated by choosing three typical cell lines (HUVEC, RAW264.7, and A549) as model cells, and no toxic effects are observed. It is also demonstrated that SH‐SSCNs can be used as a new class of magnetic resonance imaging (MRI) probes, having a remarkably high spin–spin (T2) relaxivity (r2* = 176.1 mM ?1 S?1). The combination of the sub‐100‐nm particle size, monodispersity in aqueous solution, superparamagnetism, and fluorescent properties of the SH‐SSCNs, as well as the non‐cytotoxicity in vitro, provides a novel and potential candidate for an earlier MRI diagnostic method of cancer. 相似文献

上海市高速公路收费站拥堵问题日益突出,已影响到高速公路的服务质量。拥堵原因主要有:交通量明显超过设计通行能力、部分收费站设计通行能力过低、收费站出口疏解不畅。扩容改造措施主要有:收费车道外侧拓建、建设前后分离式网架收费站、不停车电子收费(ETC)改造、可变车道、一岛双亭。目前广泛使用的是拓建收费车道和进行ETC改造。特别是ETC车道通行能力可几倍于人工收费车道,且可降低收费站成本、节约收费人工,将是解决收费站拥堵的有力措施,目前正处于高速发展中。 相似文献
