以转基因水稻中最常用的CaMV35S启动子、NOS终止子、Cry1Ab/Ac基因、HPT基因及SPS水稻内标基因为研究对象,利用5 种不同的荧光信号(FAM、HEX、Taxas Red、Cy5、Cy5.5)进行多重实时聚合酶链式反应(real-time polymerase chain reaction,real-time PCR)检测方法的研究。通过引物组合筛选、反应体系优化、特异性测试、灵敏度测试、适用性测试等一系列实验,建立了5 重real-time PCR方法,灵敏度可达0.032%。此方法具有灵敏度高、结果准确、通量大等优点,可实现水稻中转基因成分的快速、高效检测。 相似文献

This paper discusses the capability of Guo et al.'s (2021) equations to determine the discharge of radial gates under submerged flow conditions. It was concluded that Guo et al.'s (2021) equations are associated with error reduction compared to the Incomplete Self-Similarity (ISS) theory and the calibration method. However, it does not have a significant advantage over Energy-Momentum (E-M) approach. Employing E-M principles, new equations were proposed to determine the discharge of radial gates, which has some advantages compared to Guo et al. (2021), such as (1) error reduction under partially and fully submerged flow conditions, (2) least dependence on the empirical constants, (3) uniformity of form over the entire submerged condition, and (4) no need to classify the submerged flow. Field calibration showed that the proposed equations in the present study for a single gate predict the discharge of parallel radial gates with a mean absolute error of less than 4.5% subject to the submerged operation of all open gates. 相似文献

The motivation of this work is to address real-time sequential inference of parameters with a full Bayesian formulation. First, the proper generalized decomposition (PGD) is used to reduce the computational evaluation of the posterior density in the online phase. Second, Transport Map sampling is used to build a deterministic coupling between a reference measure and the posterior measure. The determination of the transport maps involves the solution of a minimization problem. As the PGD model is quasi-analytical and under a variable separation form, the use of gradient and Hessian information speeds up the minimization algorithm. Eventually, uncertainty quantification on outputs of interest of the model can be easily performed due to the global feature of the PGD solution over all coordinate domains. 针对谱聚类融合模糊C-means(FCM)聚类的蛋白质相互作用(PPI)网络功能模块挖掘方法准确率不高、执行效率较低和易受假阳性影响的问题,提出一种基于模糊谱聚类的不确定PPI网络功能模块挖掘(FSC-FM)方法。首先,构建一个不确定PPI网络模型,使用边聚集系数给每一条蛋白质交互作用赋予一个存在概率测度,克服假阳性对实验结果的影响;第二,利用基于边聚集系数流行距离(FEC)策略改进谱聚类中的相似度计算,解决谱聚类算法对尺度参数敏感的问题,进而利用谱聚类算法对不确定PPI网络数据进行预处理,降低数据的维数,提高聚类的准确率;第三,设计基于密度的概率中心选取策略(DPCS)解决模糊C-means算法对初始聚类中心和聚类数目敏感的问题,并对预处理后的PPI数据进行FCM聚类,提高聚类的执行效率以及灵敏度;最后,采用改进的边期望稠密度(EED)对挖掘出的蛋白质功能模块进行过滤。在酵母菌DIP数据集上运行各个算法可知,FSC-FM与基于不确定图模型的检测蛋白质复合物(DCU)算法相比,F-measure值提高了27.92%,执行效率提高了27.92%;与在动态蛋白质相互作用网络中识别复合物的方法(CDUN)、演化算法(EA)、医学基因或蛋白质预测算法(MGPPA)相比也有更高的F-measure值和执行效率。实验结果表明,在不确定PPI网络中,FSC-FM适合用于功能模块的挖掘。 相似文献

Basins with various mineral resources coexisting and enriching often occupy an important strategic position. The exploration of various mineral resources is repetitive at present due to unshared data and imperfect management mechanism. This situation greatly increases the cost of energy exploitation in the country. Traditional data-sharing mode has several disadvantages, such as high cost, difficulty in confirming the right of data, and lack of incentive mechanism, which make achieving real data sharing difficult. In this paper, we propose a data-sharing mechanism based on blockchain and provide implementation suggestions and technical key points. Compared with traditional data-sharing methods, the proposed data-sharing mechanism can realize data sharing, ensure data quality, and protect intellectual property. Raney-type Ni precursor alloys containing 75 at.% Al and doped with 0, 0.75, 1.5 and 3.0 at.% Ti have been produced by a gas atomization process. The resulting powders have been classified by size fraction with subsequent investigation by powder XRD, SEM and EDX analysis. The undoped powders contain, as expected, the phases Ni2Al3, NiAl3 and an Al-eutectic. The Ti-doped powders contain an additional phase with the TiAl3 DO22 crystal structure. However, quantitative analysis of the XRD results indicate a far greater fraction of the TiAl3 phase is present than could be accounted for by a simple mass balance on Ti. This appears to be a (TixNi1−x)Al3 phase in which higher cooling rates favour small x (low Ti-site occupancy by Ti atoms). SEM and EDX analysis reveal that virtually all the available Ti is contained within the TiAl3 phase, with negligible Ti dissolved in either the Ni2Al3 or NiAl3 phases. 相似文献

Computer-Supported Collaborative Learning (CSCL) is concerned with how Information and Communication Technology (ICT) might facilitate learning in groups which can be co-located or distributed over a network of computers such as Internet. CSCL supports effective learning by means of communication of ideas and information among learners, collaborative access of essential documents, and feedback from instructors and peers on learning activities. As the cloud technologies are increasingly becoming popular and collaborative learning is evolving, new directions for development of collaborative learning tools deployed on cloud are proposed. Development of such learning tools requires access to substantial data stored in the cloud. Ensuring efficient access to such data is hindered by the high latencies of wide-area networks underlying the cloud infrastructures. To improve learners’ experience by accelerating data access, important files can be replicated so a group of learners can access data from nearby locations. Since a cloud environment is highly dynamic, resource availability, network latency, and learner requests may change. In this paper, we present the advantages of collaborative learning and focus on the importance of data replication in the design of such a dynamic cloud-based system that a collaborative learning portal uses. To this end, we introduce a highly distributed replication technique that determines optimal data locations to improve access performance by minimizing replication overhead (access and update). The problem is formulated using dynamic programming. This paper presents a control design for the one‐phase Stefan problem under actuator delay via a backstepping method. The Stefan problem represents a liquid‐solid phase change phenomenon which describes the time evolution of a material's temperature profile and the interface position. The actuator delay is modeled by a first‐order hyperbolic partial differential equation (PDE), resulting in a cascaded transport‐diffusion PDE system defined on a time‐varying spatial domain described by an ordinary differential equation (ODE). Two nonlinear backstepping transformations are utilized for the control design. The setpoint restriction is given to guarantee a physical constraint on the proposed controller for the melting process. This constraint ensures the exponential convergence of the moving interface to a setpoint and the exponential stability of the temperature equilibrium profile and the delayed controller in the norm. Furthermore, robustness analysis with respect to the delay mismatch between the plant and the controller is studied, which provides analogous results to the exact compensation by restricting the control gain. 相似文献
