Objective To observe the effect of FTY720 and ICAM-1 mAb mono and combination therapy in cardiac silo-transplantation in rats. Methods Rats were randomly assigned to 9 groups, heart allo-transplantation were performed in abdominal site with micro-surgical technique. Recipients with allografts were treated with different doses of FTY720 and(or) ICAM-1 mAb. Graft survival, histopathology andlevel of serum IL-2, IFN-γ, IL-4, IL-10were investigated. Results Low doses of FTY720 (lmg/kg) combined with ICAM-1 mAb achieved synergistic effect in the prolongation of cardiac graft survival, combination index(CD =0.67. Conclusion Concomitant therapy of FTY720 and ICAM-1 mAb achieved a synergistic effect in the prolongation of heart allograft survival in rats.  相似文献

Insulin-loaded microspheres were prepared by alternating deposition film layers that were composed of insulin and poly(vinyl sulfate) potassium on the surface of poly(lactic acid) (PLA) microspheres. The preparation of the insulin-loaded microspheres was optimized by an orthogonal test design, and the relationship between drug loading (DL) and film layers was studied. The particle size, DL and encapsulation efficiency of the obtained insulin-loaded microspheres with 10 films were 5.25?±?0.15?µm, 111.33?±?1.15?mg/g and 33.7?±?0.19%, respectively. Following this, the physical characteristics of the insulin-loaded microspheres were investigated. The results from scanning electron microscopy and a laser particle size analyzer (LPSA) indicated the spherical morphology, rough surface and increasing particle sizes of the insulin-loaded microspheres, which were compared to those of PLA microspheres. An in vitro release study showed that the insulin-loaded microspheres were stable in HCl solution (pH 1.0) and released insulin slowly in phosphate-buffered solution (pH 6.8). Finally, the drug efficacy of the prepared insulin-loaded microspheres via oral administration was evaluated in rats with diabetes induced by streptozotocin, and an obvious dose-dependent hypoglycemic effect was observed. This preliminary data could illustrate the prospect of using microspheres for the oral delivery of insulin.  相似文献

目的:探讨长期低铅染毒对大鼠脾脏免疫细胞超微结构的影响。方法:建立低铅染毒的大鼠模型,8周后测定血铅并应用透射电镜技术观察大鼠脾脏免疫细胞的超微结构。结果:①电镜观察,脾脏免疫细胞出现不同程度的线粒体肿胀、空泡化或固缩;内质网肿胀、脱颗粒;②以上变化均随着铅染毒剂量的增加而加重。讨论:长期低铅染毒即可导致脾脏免疫细胞的损伤,为进一步研究铅对机体免疫系统的毒性作用提供了形态学依据。  相似文献

研究不同功率的半导体激光照射对大鼠损伤神经电生理功能恢复的影响。结果:15mW的半导体激光照射可促进神经纤维的再生过程,第6、8周时激光组的运动神经传导速度明显高于对照组,而5mW和10mW半导体激光照射组与对照组比较无明显差异。结论:15mW半导体激光照射大鼠2的坐骨神经,有促进损伤神经功能恢复的作用。  相似文献

Human growth hormone (hGH) and prolactin (hPRL) have a low sequence homology, but both bind and activate hPRL receptors. hGH also binds hGH receptors. hGH has 22 and 20 kDa forms; residues 32-46 have been deleted by alternative RNA splicing to create the smaller form. hGH requires F44 for activity through the hPRL receptor, but not for activity through the hGH receptor. The deletion of F44 from hGH has the same effect as removal of residues 32-46 (approximately 200-fold loss in activity), indicating the importance of F44 in hGH when activating the hPRL receptor. In contrast, when the homologous F50 is deleted from hPRL little or no activity is lost, indicating that this highly conserved phenylalanine is not required for the action of hPRL. Deletion of residues 41-52 (a non-conserved sequence homologous to residues 32-46 of hGH) reduced the activity of hPRL by >14 000-fold. This region is essential for the biological activity of hPRL. As these two proteins have evolved from a common ancestor, they have retained the requirement for this region but need different structural elements to activate hPRL receptors. Such diversity represents an opportunity to fine-tune hormone activity.  相似文献

通过观察大气污染物引起大鼠肺损伤的病理组织学变化,分析肺组织内血红素氧合酶1(HO-1)的表达量的变化,为大气污染所致肺损伤的生物学标记物的研究提供科学依据。实验方法如下:将30只Wistar大鼠,随机分为3个实验组(3d、7d、30d组)和3个对照组(3d、7d、30d组),对实验组大鼠染尘染毒后提取所有大鼠肺组织,观察其病理组织学变化;RT-PCR技术检测各组肺组织中HO-1的mRNA表达;Western-blot技术检测各组肺组织中HO-1蛋白的表达。结果显示:实验组(7d及30d)的肺组织病理形态学评分显著高于对照组(7d及30d),实验组(7d及30d)大鼠肺组织HO-1mRNA表达与对照组相比,具有显著性差异(P均〈0.05)。随着吸入大气混合污染物的时间延长,肺组织内的HO-1蛋白表达水平逐渐增高,实验组(7d及30d)与对照组(7d及30d)相比,差异有显著性意义(P〈0.05)。在吸入大气混合污染物后早期(7d),HO-1转录水平升高。持续吸入混合大气污染物(30d),大鼠肺组织出现明显的病理组织学变化,HO-1转录水平、蛋白表达水平均比对照组明显升高。得出结论:可将HO-1作为大气混合污染物所致的肺损伤的早期生物学标记物;采取某些手段增加肺组织HO-1的表达,可能会减少混合大气污染物对肺组织造成的损伤。  相似文献

In this work, we address the problem of compact cell topology selection for miniaturization of rat‐race couplers. The principal objective of the design process is to achieve the smallest possible footprint of the coupler, while maintaining the required levels of electrical parameters imposed on its components. Our approach permits identification of the minimum achievable coupler area, provided that the circuit is composed of a given compact cell and folded lines. This allows for the quantitative assessment of a set of considered cells with respect to the miniaturization capabilities they exhibit under certain design specifications. The proposed method is validated using 6 different cells with unified parameterization to identify the smallest rectangular‐like rat‐race coupler described by 2 design specifications. The obtained results attest that circuit topology and electrical parameters of the reference design are critical factors determining the final miniaturization rate. The proof‐of‐concept prototype devices occupy merely 8% of the conventional coupler area, while preserving fractional bandwidths (20% and 13.5%) of their conventional counterparts. The experimental results confirm the claims inferred from the numerical data.  相似文献

A compact reconfigurable rat‐race coupler with tunable frequency and tunable power dividing ratio is proposed for the first time. Varactors and two single control voltages are used to obtain both the tunable frequency and the tunable power dividing ratio in this article. The structure of the rat‐race coupler involves 50 Ω parallel‐strip lines only and a phase inverter is used for size reduction. Theoretical equations for the relationship among S‐parameters and the capacitance of varactors are derived. The graphic method is used to choose capacitance for the desired operation frequency and the desired power dividing ratio. For demonstration, a prototype is designed and fabricated. The measured results show that the rat‐race coupler's frequency and the power dividing ratio can be effectively tuned in 0.69 GHz ~ 0.81 GHz and 3 dB ~ 14 dB, respectively with isolation better than 20 dB, phase difference less than 7°and return loss better than 20 dB. The theoretical simulation, electromagnetic simulation, and measured results show good agreement in this design.  相似文献

This article proposes an analytical design methodology for dual‐band hybrid couplers and baluns structures for any arbitrary frequency ratio using a stub‐loaded transmission line. An analysis of changing the impedance behavior of the stub, is carried out for the two bands of operation, which along with a dispersive analysis, emphasizes certain conditions where the existing methodology is not applicable. In addition, an extra degree of freedom has been included to increase the solutions for a given frequency ratio, thus providing greater flexibility and feasibility of the proposed structure. The design methodology is experimentally validated with the design and fabrication of dual‐band branch‐line and rat‐race couplers for various commercial frequency bands. © 2011 Wiley Periodicals, Inc.
