This study evaluated the genetic similarity and prevalence of the stx1, stx2, eae, and ehxA genes in Escherichia coli isolated from pasteurized cow milk. Eighty‐seven E. coli isolates from pasteurized cow milk from 22 dairies located in northwestern Paraná state, Brazil, were analyzed. Genetic similarity was evaluated using enterobacterial repetitive intergenic consensus sequence polymerase chain reaction (ERIC‐PCR) and repetitive extragenic palindromic sequence PCR (REP‐PCR). E. coli isolates were also analyzed by PCR to investigate the presence of the stx1, stx2, eae, and ehxA genes. ERIC‐PCR and REP‐PCR clustered 87 bacterial isolates in 76 and 81 genomic profiles, respectively. Both techniques revealed high genetic diversity among the E. coli isolates, confirming the possibility of their use in epidemiological studies. The stx1, stx2, eae, and ehxA virulence genes were not detected in E. coli isolates, indicating a low prevalence of Shiga toxin‐producing E. coli in milk produced in the region studied. 相似文献

通过传统微生物培养分离手段从山西老陈醋醋醅中分离醋酸菌,采用形态学观察及分子生物学技术对其进行鉴定,并对分离菌株进行肠杆菌基因间保守重复序列(ERIC)分型及发酵特性研究。结果表明,共分离筛选出20株醋酸菌,经鉴定18株为巴氏醋杆菌(Acetobacter pasteurianus),2株为醋化醋杆菌(Acetobacter aceti)。经ERIC分型,18株巴氏醋杆菌属于5个分型(Ⅰ~Ⅴ型),2株醋化醋杆菌属于2个分型(Ⅰ和Ⅱ型)。经发酵特性研究,巴氏醋杆菌、醋化醋杆菌中Ⅱ型菌株的温度耐受性均优于Ⅰ型;巴氏醋杆菌Ⅰ型菌株产酸、产乙偶姻能力优于其他型的菌株。在此基础上,筛选出的1株性能优良的菌株,为巴氏醋杆菌SAV-CP 1884,其可耐受高温45 ℃、酒精度10%vol及pH 3,乙酸产量高达9.87 g/L、乙偶姻产量为0.86 mg/mL。 相似文献

Enterobacter sakazakii is an emerging foodborne pathogen associated with severe diseases in neonates. Infant formula milk (IFM) has been identified as one of the major contaminated sources and a transmission vehicle. To determine the phenotypic and genotypic characterization of this pathogen, 22 E. sakazakii strains isolated from IFM by an FDA-recommended method and PCR on the α-glucosidase gene were subtyped by random amplified polymorphic DNA (RAPD)-PCR, enterobacterial repetitive intergenic consensus (ERIC)-PCR, and antibiotic resistance patterns. At a similarity threshold of 80%, 16 ERIC-PCR fingerprint types were identified with a discriminatory power (D) of 0.933, and 18 RAPD-PCR types were identified with D of 0.973. Resistance to 9 antibiotics tested by disk diffusion assay revealed 6 antibiotic resistance patterns with D of 0.749. The comparison of characterization indicated that RAPD-PCR and ERIC-PCR have high discriminatory power showing genetic diversity of E. sakazakii isolates, and ERIC-PCR patterns showed a closer correlation than RAPD-PCR patterns to phenotypic characterization and the brands of IFM. Overall, the ERIC-PCR typing method could be used for tracing sources of E. sakazakii isolates in the food chain. 相似文献

建立空肠弯曲菌(Campylobacter jejuni)的ERIC-PCR分子生物学分型技术,比较ERIC-PCR分型和生化分型的分型效果。从菌株TY1273出发,运用L16(54)正交试验,对Mg2+、dNTPs、引物和TaqDNA聚合酶浓度等因素在较大范围水平内进行反应体系条件摸索,得到一个初步的优化体系;在此基础之上进行单因素的进一步小范围水平内的微调优化,得到最终的优化体系;最后以优化的ERIC-PCR方法对24株C.jejuni分离株分型;同时根据API Campy生化反应结果进行生化分型,比较ERIC-PCR分子分型方法和生化分型方法。结果显示ERIC-PCR方法将24株菌扩增均得到大小在100 bp-3000 bp之间的条带,并可将其分为22个基因型,分辨系数为0.92,具有较高的分辨力。生化分型将24株菌分为19个生化型,显示了菌株基因的多样性。表明ERIC-PCR技术比生化分型能更好的体现菌株的遗传多样性,且具有简便和分辨力搞等优点,可用于C.jejuni的多样性研究。 相似文献
