Sexually transmitted infections (STIs) caused by Neisseria gonorrhoeae, Chlamydia trachomatis and Mycoplasma genitalium are a common cause of pelvic inflammatory disease (PID) which can lead to tubal factor infertility (TFI). TFI is one of the most common causes of infertility, accounting for 30% of female fertility problems. STIs can also have an impact on pregnancy, leading to adverse pregnancy outcomes. Escalating antibiotic resistance in Neisseria gonorrhoeae and Mycoplasma genitalium represents a significant problem and can be therapeutically challenging. We present a comprehensive review of the current treatment options, as well as the molecular approach to this subject. We have given special attention to molecular epidemiology, molecular diagnostics, current and new treatments, and drug resistance.  相似文献

We scanned the genome of 77,815 Normande cattle with different Illumina SNP chips (Illumina Inc., San Diego, CA) to map recessive embryonic lethal mutations using homozygous haplotype deficiency. We detected 2 novel haplotypes on chromosomes 11 and 24 but did not confirm 6 previously reported haplotypes. The one on chromosome 11 showed a marked reduction in conception rates and moderate decrease in nonreturn rate in at-risk versus control mating, supporting late embryonic mortality. After fine mapping and analyzing whole-genome sequences, we prioritized a missense mutation in CAD (g.72399397T>C; p.Tyr452Cys)—a gene encoding a protein (carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, and dihydroorotase) essential for de novo pyrimidine biosynthesis—as a candidate causal variant. This transition mutation replaces a tyrosine residue, which is perfectly conserved among living organisms, with a cysteine residue in the carbamoyl-phosphate synthetase 2 domain of the protein. A single animal was confirmed to be homozygous for the mutation based on Sanger sequencing. However, large-scale genotyping of the candidate variant with the Illumina EuroG10k BeadChip revealed an absence of live homozygotes in a panel of 33,323 Normande animals and an absence of carriers in 348,593 animals from 19 other cattle breeds. These results support recessive embryonic lethality with nearly complete penetrance, as was previously reported in CAD mutants in several eukaryote species. The only homozygous cow had extremely poor udder conformation, suggesting a potential role of CAD in udder development, but no effect was detected when comparing daughter yield deviations of 250 heterozygous bulls with that of 2,912 homozygotes for the ancestral allele. Together, our results showed the importance of large-scale screening for homozygous haplotype deficiency with hundreds of thousands of animals, validating results with an independent data set, and considering unexpected live homozygotes, to avoid both false-positive and false-negative discoveries. These discoveries will be used primarily in mating decisions to avoid at-risk mating. In addition, we recommend including CAD in the breeding objectives of Normande cattle.  相似文献

Nearly 57,000 single-nucleotide polymorphisms (SNP) genotyped with the Illumina BovineSNP50 BeadChip (Illumina Inc., San Diego, CA) were investigated to determine usefulness of the associated SNP for genomic prediction. Genotypes were obtained for 12,591 bulls and cows, and SNP were selected based on 5,503 bulls with genotypes from a larger set of SNP. The following SNP were deleted: 6,572 that were monomorphic, 3,213 with scoring problems (primarily because of poor definition of clusters and excess number of clusters), and 3,649 with a minor allele frequency of <2%. Number of SNP for each minor allele frequency class (≥2%) was fairly uniform (777 to 1,004). For 5 contiguous SNP assigned to chromosome 7, no bulls were heterozygous, which indicated that those SNP are actually on the nonpseudoautosomal portion of the X chromosome. Another 178 SNP that were not assigned to a chromosome but that had many fewer heterozygotes than expected were also assigned to the X chromosome. Existence of Hardy-Weinberg equilibrium was investigated by comparing observed with expected heterozygosity. For 11 SNP, the observed percentage of heterozygous individuals differed from the expected by >15%; therefore, those SNP were deleted. For 2,628 SNP, the genotype at another SNP was highly correlated (i.e., genotypes were identical for >99.5% of bulls), and those were deleted. After edits, 40,874 SNP remained. A parent-progeny conflict was declared when the genotypes were alternate homozygotes. Mean number of conflicts was 2.3 when pedigree was correct and 2,411 when it was incorrect. The sire was genotyped for >93% of animals. Maternal grandsire genotype was similarly checked; however, because alternate homozygotes could be valid, a conflict threshold of 16% was used to indicate a need for further investigation. Genotyping consistency was investigated for 21 bulls genotyped twice with differences primarily from SNP that were not scored in one of the genotypes. Concordance for readable SNP was extremely high (99.96-100%). Thousands of SNP that were polymorphic in Holsteins were monomorphic in Jerseys or Brown Swiss, which indicated that breed-specific SNP sets are required or that all breeds need to be considered in the SNP selection process. Genotypes from the Illumina BovineSNP50 BeadChip are of high accuracy and provide the basis for genomic evaluations in the United States and Canada.  相似文献

The aim of this study was to quantify the impact of genotyping cows with reliable phenotypes for direct health traits on annual monetary genetic gain (AMGG) and discounted profit. The calculations were based on a deterministic approach using ZPLAN software (University of Hohenheim, Stuttgart, Germany). It was assumed that increases in reliability of the total merit index (TMI) of 5, 15, and 25 percentage points were achieved through genotyping 5,000, 25,000, and 50,000 cows, respectively. Costs for phenotyping, genotyping, and genomic estimated breeding values vary between €150 and €20 per cow. The gain in genotyping cows for traits with medium to high heritability is more than for direct health traits with low heritability. The AMGG is increased by 1.5% if the reliability of TMI is 5 percentage points higher (i.e., 5,000 cows genotyped) and 6.53% higher AMGG can be expected when the reliability of TMI is increased by 25 percentage points (i.e., 50,000 cows genotyped). The discounted profit depends not only on the costs of genotyping but also on the population size. This study indicates that genotyping cows with reliable phenotypes is feasible to speed up the availability of genomic estimated breeding values for direct health traits. But, because of the huge amount of valid phenotypes and genotypes needed to establish an efficient genomic evaluation, it is likely that financial constraints will be the main limiting factor for implementation into breeding program such as Fleckvieh Austria.  相似文献

Characterization and genotyping of the caprine kappa-casein variants   总被引:4,自引:0,他引:4  
Kappa-Casein (kappa-CN) is the milk protein that determines the size and specific function of milk micelles, and its cleavage by chymosin is responsible for milk coagulation. We have previously detected and characterized four variants of the goat kappa-CN in Spanish, French, and Italian breeds by screening the major part of the coding region in exon 4. Here we have sequenced and analyzed the full coding region of the kappa-CN gene which includes exons 3 and 4. No additional mutations were found, with exception of a single nucleotide substitution in exon 3, which had no amino acid change. However, the analysis of the association between the different mutations resulted in two new variants designated kappa-CN F and G. The novel variants are present in the Italian breeds Teramana, Girgentana, and Sarda (variant F). A protocol for rapid simultaneous genotyping of all known kappa-CN variants using the primer extension method was described, and a total of 210 animals from nine European breeds were genotyped. Alleles A and B are the most frequent variants occurring in the majority of breeds with highest prevalence of the B variant, except for the Canaria breed where the A allele is more frequent. Sequence data suggest that the F variant is the original type of caprine kappa-CN, other alleles being derived from this type following two different trunks by successive mutations.  相似文献

目的 了解福建省养殖环节牡蛎中诺如病毒(Norovirus)污染状况及基因分型,进一步定量分析诺如病毒污染水平,为食品中诺如病毒污染监测及安全风险评估提供数据支持。方法 2017年8月—2018年9月,于福建省某养殖基地采集牡蛎样品共244份,采用实时荧光定量聚合酶链式反应(PCR)法对样品中诺如病毒进行检测,确定其污染情况及基因分型。结果 211份有效样品中诺如病毒阳性率为30.33%(64/211),GI和GII组诺如病毒均有检出。GI组诺如病毒阳性率为16.59%(35/211),GII组阳性率为24.17%(51/211),GI和GII组同时为阳性的比例为10.43%(22/211)。诺如病毒含量在1.05×102~1.68×104基因拷贝/g(消化腺)之间。结论 福建省养殖环节牡蛎样品中存在诺如病毒污染,冬季阳性率较高,提示有关部门需在冬季采取更多针对性的风险管理干预措施,以降低食源性诺如病毒感染风险。  相似文献
